1Heshouwu decoction, a Chinese herb for tonifying kidney, ameliorates hypothalamic-pituitary-testicular axis secretion in aging rats显示文摘An increasing amount of evidence demonstrates the anti-aging effect of Heshouwu in pill form. In this study, a subacute aging rat model was established by continuous intraperitoneal injection of D-galactose and treated with Heshouwu decoction (a Chinese herb for tonifying the kidney, comprising Heshouwu pill, Herba Epimedii, Radix Salviae Miltiorrhiae, and Poria). Heshouwu pill treated rats were the positive control group. Radioimmunoassay, immunohistochemical staining, and western blot assay showed hypothalamic gonadotropin-releasing hormone, hypothalamic substance P, and serum gonadotropin levels to be significantly increased in the model rats; the concentrations of hypothalamic β-endorphin, and serum levels of insulin-like growth factor 1 and testosterone were significantly decreased. 17β- and 3β-hydroxysteroid dehydrogenase expression in testicular tissue was also decreased. Intragastric administration of Heshouwu decoction at high (9.6 g/mL/100 g), medium (4.8 g/mL/100 g), and low (2.4 g/mL/100 g) doses, Heshouwu decoction pretreatment at a medium dose (4.8 g/mL/100 g), and Heshouwu pill (2.06 g/mL/100 g) significantly reversed these changes. Heshouwu decoction pretreatment and high-dose Heshouwu decoction had the greatest anti-aging effects. These experimental findings indicate that Heshouwu decoction can improve hypothalamic-pituitary-testicular axis secretion in a subacute aging rat model, and prevent and delay gonadal axis aging, with an effect superior to that of Heshouwu pill.Siyun Niu Suru Kou Xiaochun Zhou Liang Ding 2012Neural Regeneration Research2012,7,21:3
2Studies on the relationship between the senescence of Leydig cells and the regulation of insulin/IGF-1 signaling pathway in rats delayed by Heshouwuyin显示文摘This work aims to investigate the relationship between the senescence of testicular Leydig cells and the regulation of insulin/IGF-1 signaling pathway in rats delayed by Heshouwuyin.The oxidative damage technique was used to establish Leydig cell aging model.Flow cytometry was used to detect the changes of cell cycle in each group.Detection of mRNA and protein expression levels of key genes of insulin/IGF-1 signaling pathway in Leydig cells by Real-time PCR,immunofluorescence and Western blotting.Cells were treated with insulin receptor/IGF receptor specific inhibitor BMS 754807,flow cytometry was used to detect the apoptotic level of Leydig cells.The results showed that compared with the normal group,the proportion ofβgalactosidase positive cells in the aging group was more than 60%(P<0.05),and the proportion of G1 phase increased significantly,while that of S phase decreased significantly(P<0.05);Changes in the expression of key genes of the insulin/IGF-1 signaling pathway:The expression levels of INSR,IRS1,IRS2,IGF-1 mRNA and protein in the aging group were significantly lower than those in the normal group(P<0.05),and the mRNA expression of IGFBP3 was significantly higher than in the normal group(P<0.05),Heshouwuyin can reverse the above phenomenon.Song Yicheng Jiang Liping Niu Shuang Duan Yulei Zhu Juan Niu Siyun Qi Feng 2021解剖学杂志2021,44,S01:0
