| 1 | Pandanus amaryllifolius leaf extract increases insulin sensitivity in high-fat diet-induced obese mice显示文摘Objective: To examine the effect of Pandanus amaryllifolius(P. amaryllifolius) leaf extract on the insulin resistance state in obese ICR mice.Methods: Obesity was induced in mice fed with high-fat diet(45% fat) for 12 weeks. After the first six weeks on the diet, the obese mice were administered with the water extract of P. amaryllifolius leaf at 125 and 250 mg/kg/day, respectively for another six weeks. At the5 th week of treatment, oral glucose tolerance test was conducted. After six weeks of treatment, the levels of blood glucose, serum insulin, leptin, adiponectin, and lipid profiles were determined. The liver, muscle and epididymal fat tissues were removed for measuring the biochemical parameters and protein expression, as well as histological examination.Results: Six weeks of treatment with P. amaryllifolius led to a significant reduction in the blood glucose level as well as improvement in the insulin resistance. P. amaryllifolius also increased the liver glycogen storage and serum adiponectin and decreased the serum leptin levels. A reduction in the serum and hepatic triglyceride, and non-esterified fatty acid levels was also observed. The histological examination showed that the obese mice treated with P. amaryllifolius reduced the lipid droplet in liver tissue and adipocyte size in epididymal fat tissue. The treatment also increased the protein expression of glucose transporter 4 in the muscle and fat tissues.Conclusions: The treatment with P. amaryllifolius could decrease several parameters of impaired glucose and lipid metabolism. | 2 | Serum proteins in chronic hepatitis B patients treated with peginterferon alfa-2b显示文摘AIM: To study the differential protein profile in serum of hepatitis B patients.METHODS: Serum samples were obtained from patients with chronic hepatitis B who were receiving peginterferon alfa-2b.The serum samples were subjected to albumin depletion and analyzed by two-dimensional gel electrophoresis(2-DE).Differentially expressed protein spots were identified by electrospray ionizationquadrupole time-of-flight mass spectrometry.Alpha2-HS-glycoprotein,complement component C3c and CD5 antigen were further analyzed by an enzymelinked immunosorbent assay and immunonephelometry.RESULTS: Nineteen patients with HBeAg-positive chronic hepatitis B(CHB) were studied.These patients were followed for at least 1 year after treatment and were classified according to their treatment response: responders(n = 9) and non-responders(n = 10).2-DE and MS/MS analysis were performed to compare the serum proteins before initiating peginterferon alfa2b.From the quantitative analysis of the 2-D gel,7 proteins were detected between the two groups at different levels before treatment.Among these potential candidates,serum levels of alpha-2-HS-glycoprotein,complement component C3c and CD5 antigen-like precursor were further analyzed.In the validation phase,23 subjects,9 sustained responders and 14 nonresponders,were recruited.Interestingly,the levels of alpha-2-HS-glycoprotein and complement component C3c were elevated in the serum of the non-responders compared to the responders.CONCLUSION: Serum alpha-2-HS-glycoprotein and complement component C3c may be potential serum biomarkers in predicting the treatment response of peginterferon alfa-2b in patients with CHB prior to treatment. | Sunida Kuakarn Poorichaya SomParn Pisit Tangkijvanich Varocha Mahachai Visith Thongboonkerd Nattiya Hirankarn | 2013 | World Journal of Gastroenterology2013,19,31: | 4 |

| 11 | Assessment of pesticide contaminated sediment using biological response of tropical chironomid,Chironomus javanus Kiffer as biomarker显示文摘Objective: To investigate the use of a biomarker for assessment of the effects on the tropical chironomid, Chironomus javanus(C. javanus), Kiffer of sediment contaminated with an insecticide(chlorpyrifos).Methods: A wide range of biological responses to the tropical chironomid exposed were measured, including survival, growth rate and Acetylcholinesterase(AChE) activity.Results: The measured median lethal concentration(96 h LC_(50)) of chlorpyrifos to C. javanus was 0.056(95% CI 0.024-0.124)μg/kg. For sub-chronic levels of chlorpyrifos between 0.001 and 0.25μg/kg administered for 10 days, the effects on the growth of C. javanus were reduced(larva size, head structure width and dry weight) at the significance level(P<0.01) and the effects were concentration dependent. Following exposure to chlorpyrifos at the level of 0.001μg/kg for 48 and 96 h, the AChE activity in C. javanus was inhibited compared with control samples(P<0.05).Conclusions: This study demonstrated that C. javanus was sensitive when exposed to chlorpyrifos.
