| 2 | Celastrol enhances transcription factor EB (TFEB)-mediated autophagy and mitigates Tau pathology:Implications for Alzheimer's disease therapy显示文摘Alzheimer's disease(AD),characterized by the accumulation of protein aggregates including phosphorylated Tau aggregates,is the most common neurodegenerative disorder with limited therapeutic agents.Autophagy plays a critical role in the degradation of phosphorylated Tau aggregates,and transcription factor EB(TFEB)is a master regulator of autophagy and lysosomal biogenesis.Thus,small-molecule autophagy enhancers targeting TFEB hold promise for AD therapy.Here,we found that celastrol,an active ingredient isolated from the root extracts of Tripterygium wilfordii(Lei Gong Teng in Chinese)enhanced TFEB-mediated autophagy and lysosomal biogenesis in vitro and in mouse brains.Importantly,celastrol reduced phosphorylated Tau aggregates and attenuated memory dysfunction and cognitive deficits in P301S Tau and 3xTg mice,two commonly used AD animal models.Mechanistical studies suggest that TFEB-mediated autophagy-lysosomal pathway is responsible for phosphorylated Tau degradation in response to celastrol.Overall,our findings indicate that Celastrol is a novel TFEB activator that promotes the degradation of phosphorylated Tau aggregates and improves memory in AD animal models.Therefore,Celastrol shows potential as a novel agent for the treatment and/or prevention of AD and other tauopathies. | Chuanbin Yang Chengfu Su Ashok Iyaswamy Senthil Kumar Krishnamoorthi Zhou Zhu Sichang Yang Benjamin Chunkit Tong Jia Liu Sravan G.Sreenivasmurthy Xinjie Guan Yuxuan Kan Aston Jiaxi Wu Alexis Shiying Huang Jieqiong Tan Kingho Cheung Juxian Song Min Li | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 2 |

| 7 | Cross-sectional analysis of obesity and high blood pressure among undergraduate students of a university medical college in South India显示文摘Objective:To estimate the prevalence of obesity and high blood pressure among undergradu-ate students of a university medical college.Methods:A cross-sectional study was conducted at a medical college among 434 medical stu-dents.A questionnaire was used to collect basic demographic details,followed by anthropometric measurements.Body mass index(BMI)was classified according to the World Health Organization classification.Blood pressure was measured with a standard mercury sphygmomanometer and clas-sified according to the seventh report of the Joint National Committee(JNC VII)on prevention,detection,evaluation,and treatment of high blood pressure.Data were entered into and analyzed with SPSS version 15.0.Results:Nearly 65.0% of students had normal BMI,9.9% were underweight,17.9% were overweight,and 7.6% were obese.Obesity was more prevalent among males than among females on the basis of anthropometric variables such as BMI,waist-hip ratio,and waist-stature ratio,and this difference was found to be statistically significant.Blood pressure was in the normal range among 55.0% of the students,36.6% had blood pressure in the prehypertensive range,7.6% had blood pressure in the stage 1 category of the JNC VII criteria,and 0.5% had blood pressure in the stage 2 category of the JNC VII criteria.Among the students who had blood pressure greater than 140/90 mm Hg,63.0% were males and 37.0% were females,and this difference was statistically significant.Conclusion:The prevalence of obesity was 7.6% and that of high blood pressure was 8.1% among the medical students,which were higher than those reported in the literature for the same age group and warrant further evaluation. | Sravan Kumar Chenji Chythra R.Rao Saranya Sivanesan Veena Kamath Asha Kamath | 2018 | Family Medicine and Community Health2018,6,2: | 1 |

| 12 | Modified distance regularized level set evolution for brain ventricles segmentation显示文摘Neurodegenerative disorders are commonly characterized by atrophy of the brain which is caused by neuronal loss.Ventricles are one of the prominent structures in the brain;their shape changes,due to their content,the cerebrospinal fluid.Analyzing the morphological changes of ventricles,aids in the diagnosis of atrophy,for which the region of interest needs to be separated from the background.This study presents a modified distance regularized level set evolution segmentation method,incorporating regional intensity information.The proposed method is implemented for segmenting ventricles from brain images for normal and atrophy subjects of magnetic resonance imaging and computed tomography images.Results of the proposed method were compared with ground truth images and produced sensitivity in the range of 65%–90%,specificity in the range of 98%–99%,and accuracy in the range of 95%–98%.Peak signal to noise ratio and structural similarity index were also used as performance measures for determining segmentation accuracy:95%and 0.95,respectively.The parameters of level set formulation vary for different datasets.An optimization procedure was followed to fine tune parameters.The proposed method was found to be efficient and robust against noisy images.The proposed method is adaptive and multimodal. | Thirumagal Jayaraman Sravan Reddy M Manjunatha Mahadevappa Anup Sadhu Pranab Kumar Dutta | 2020 | Visual Computing for Industry,Biomedicine,and Art2020,3,1: | 0 |
