1Cloning and Analysis of TasA Gene in Bacillus subtilis HAS显示文摘In order to clarify the mechanism for the inhibitory action of the Bacillus subtilis strain HAS on Sporisorium scitamineum(Syd.),which can cause sugarcane smut,the full-length TasA gene which encodes a protein with broad-spectrum antimicrobial activity,was PCR-amplified from HAS,and cloned into pMD18-T vector.Sequence analysis indicated that the full-length Tas A cloned from HAS consisted of 786 nucleotides,and shared 99% homology in nucleotide sequence with the TasA gene sequence published in Genbank(AJ871386.1).It was predicted that the molecular weight of Tas A protein was about 28 kD.Base transitions or transversions occurred at positions 104,164,169,250,399,623 and 627,at the 2^(nd),2^(nd),3^(rd),1^(st),3^(rd),2^(nd) and 3^(rd) bases of TasA codons.The mutations in the seven bases may cause the missense mutations of the polypeptide chain.Compared with the amino acid sequences of the TasA protein encoded by Bacillus subtilis subsp.subtilis str.168,mutations in two amino acids at positions 150 and 209of the protein encoded by the cloned TasA gene were found,and as a result,an alanine was replaced with a threonine,and a glutamic acid was replaced with an asparagine.Guoruo XIONG Gengfeng ZHAO Wenwei CAI Suran WU Benpeng YANG Shuzhen ZHANG 2017Agricultural Biotechnology2017,6,2:0
2Integrated Green Prevention and Control Techniques for Kiwifruit Canker Disease in 'Guichang' Kiwifruit in Xiuwen County, Guizhou Province显示文摘Kiwifruit canker disease seriously affects the yield and quality of'Guichang'kiwifruit in Xiuwen County,Guizhou Province.In order to scientifically,safely,greenly and efficiently prevent and control the disease,theory was combined with prevention and control techniques to optimize existing prevention and control techniques,so as to improve the production yield and quality of kiwifruit.Specifically,biocontrol strains targeting local kiwifruit canker disease were screened,and reduced and mixed use of agrochemicals with improved efficiency was studied;and the effects and application techniques of disease resistance inducers and bioorganic fertilizers in inducing systemic disease resistance in kiwifruit trees were explored,and finally,an integrated green prevention and control scheme for kiwifruit canker disease that is suitable for kiwifruit production areas in Guizhou Province and has strong operability was proposed.This study provides technical support for green,efficient,standardized production technical services and sustainable and healthy development of kiwifruit industry.Miao LI Xian WEI Rong WU Shouying TANG Suran WAN Liqian JIANG Song BAI 2023Agricultural Biotechnology2023,12,5:0
3Control Effects of 12 Kinds of Insecticides on Sugarcane Pests at Different Periods显示文摘[Objective]The paper was to study control effects of 12 kinds of insecticides on sugarcane pests at different periods.[Method]By field experiment,the control effects of different insecticides against sugarcane pests at planting stage,seedling stage and tillering stage of sugarcane were studied.[Result]After seedcane was treated by Chlorantraniliprole,Durivo and Seed soaking,the dead heart seedlings were significantly reduced in fields,and the control effects were87.13%±12.87%,85.22%±14.78%,82.71%±17.29%,respectively;Durivo(seedling emergence rate is 63.00%±10.30%)and Seed soaking(seedling emergence rate is 57.75%±4.50%)could also significantly enhance seedling emergence rate.After'removal-irrigation'treatment during seedling stage,the control effects of Flubendiamide(76.26%±4.31%)and Chlorantraniliprole(61.66%±6.65%)against sugarcane stem borer were significantly higher than Virtako(60.17%±7.06%),Deltamethrin(38.75%±9.88%),lambda-cyhalothrin(54.06%±6.83%),18%Dimehypo AS(37.98%±10.31%)and Dipterex(38.64%±9.34%).During tillering stage,with the increased application of 3.6%Dimehypo GR in cultivation and earthing up,the control effect on stem borer was significantly increased,and the control effect reached(72.03%±17.06%)as the dosage was 120 kg/hm2;when the dosage of phoxim exceeded 30 kg/hm2,the control effect on underground pests reached 44.00%±14.80%,and the control effect did not have significant difference as the dosage was ranged from 30kg/hm2to 60 kg/hm2.[Conclusion]The paper provided scientific basis for selection of different control methods,different chemicals and dosages against pests during different periods.Wu Suran Xiong Guoru Zhang Shuzhen Zhang Yuliang Wang Jungang Zeng Jun Yang Benpeng 2013Plant Diseases and Pests2013,4,5:0
4Resistance Evaluation of 23 Sugarcane Varieties to Smut显示文摘[Objective] The paper aimed to screen sugarcane varieties and species layout suitable for Hainan sugarcane area.[Method] Using mixed spores of Ustilago scitaminea Sydow,smut resistance of 21 major sugarcane varieties in national sugarcane regional trial as well as ROC22 and ROC16 were identified through artificial inoculation test and field natural incidence.[Result] Yunzhe 99-596,Yuegan 42,Guitang 29 and Funong 39 reached highly resistant and resistant levels to smut in artificial inoculation test.[Conclusion] Most of the varieties showed better resistance to smut in newly planted sugarcane and ratoon sugarcane except Yunzhe 03-103,Liucheng 03-1137 and ROC22.Xiong Guoru Cai Wenwei Wang Wenzhi Wu Suran Yang Benpeng Zhang Shuzhen 2017Plant Diseases and Pests2017,8,3:0
5Screening and Identification of Antagonistic Strain HAS against Sporisorium scitaminea Sydow显示文摘The present study was to screen and identify a biocontrol strain with antagonistic effect against Sporisorium scitaminea Sydow. 16 S rDNA gene fragments were amplified by universal primers P0 and P6. The results showed that 16 S rDNA gene fragment with the size of 1 533 bp was obtained,and its sequence had the homology of 99% with the sequence of Bacillus subtilis. The morphological characteristics of the strain were as follows: the mycelium of strain HAS was short-rod shape,and could move; Gram staining was positive; 3% KOH solubility test was negative; flagellum was grown around the cells; spores were grown in the middle,elliptic,and cysts were slightly swelled. The strain was preliminarily identified as B. subtilis. Dual culture with pathogenic fungi showed that the strain had good inhibitory effect against various pathogenic fungi causing diseases on sugarcane and other crops.
