| 1 | Increasing the frequency of CIK cells adoptive immunotherapy may decrease risk of death in gastric cancer patients显示文摘AIM: To analyze the correlation between cytokineinduced killer (CIK) cells adoptive immunotherapy and cancer-related death in gastric cancer patients. METHODS: One hundred and fifty-six gastric cancer patients after operation at the Third Affiliated Hospital of Soochow University were enrolled in this study. Their clinical data including demographic characteristics, operation time, tumor size, pathological type and staging, tumor metastasis, outcome of chemotherapy or CIK cells adoptive immunotherapy, survival time or time of death were collected with a standard structured questionnaire. Kaplan-Meier method was used to estimate the median survival time, and the 2- and 5- year survival rates. Hazard risk (HR) and 95% confidence interval (95% CI) of CIK cells adoptive immunotherapy for gastric cancer were calculated using the two-stage time-dependent covariates Cox model. RESULTS: The survival time of gastric cancer patients was longer after CIK cells adoptive immunotherapy than after chemotherapy (χ 2 = 10.907, P = 0.001). The median survival time of gastric cancer patients was also longer after CIK cells adoptive immunotherapy than after chemotherapy (49 mo vs 27 mo, P < 0.05). The 2- and 5-year survival rates of gastric cancer patients were significantly higher after CIK cells adoptive immunotherapy than after chemotherapy (73.5% vs 52.6%, 40.4% vs 23.9%, P < 0.05). A significant difference was observed in the survival curve for patients who received CIK cells adoptive immunotherapy (0, 1-10, 11-25, and over 25 frequencies) (χ 2 = 14.534, P = 0.002). The frequencies of CIK cells adoptive immunotherapy were significantly related with the decreasing risk of death in gastric cancer patients after adjustment for sex and age of the patients, tumor stage and relapse (HR = 0.54, 95% CI: 0.36-0.80) when the first stage Cox model was used to define the subjects who remained alive beyond 36 mo as survivors. However, no correlation was observed between the frequencies of death in CIK cells adoptive immunotherapy and the risk of gastric cancer patients (HR = 1.09, 95% CI: 0.63-0.89) when the second stage Cox model was used to define the subjects who survived for more than 36 mo as survivors. | 4 | Quantum secret sharing between multiparty and multiparty with Bell states and Bell measurements显示文摘We present a quantum secret sharing scheme between multiparty (m members in Group 1) and multiparty (n members in Group 2), and analyze its security. This scheme takes EPR pairs in Bell states as quantum resources. In order to obtain the shared key, all members only need to perform Bell measurements, rather than perform any local unitary operation. | 5 | Formation of complexes of Rubisco-Rubisco activase from La^3+, Ce^3+ treatment spinach显示文摘The complex of Rubisco and Rubisco activase from LaCl3-, CeCl3- treated spinach in vivo is induced. SDS-PAGE result shows that the purified proteins from LaCl3-, CeCl3- treated spinach have not only large and small subunits (55 kD, 14.4 kD) of Rubisco, but also two large subunits of 45 kD and 41 kD near the large subunits of Rubisco. Native-PAGE shows that the purified proteins from LaCl3-, CeCl3- treated spinach have not only a band of Rubisco (560 kD), but also a band of about 1100 kD, about twice distant from Rubisco, which might be a complex of Rubisco and Rubisco activase. The purified enzyme activities from LaCl3-, CeCl3-treated spinach are 1.8 and 2.8 times that of the control,the intensities of absorption and fluorescence are sig- nificantly higher than that of the purified Rubisco from the control, and the total sulfhydryl groups and available sulfhydryl groups are 36—39 –SH per mol enzyme, 14—25 –SH per mol enzyme more than those of the purified Rubisco from the control, respectively. The CD spectra show that the secondary structure of the purified enzyme from LaCl3-, CeCl3 -treated spinach is very dif- ferent from the control. | 6 | Association between Serum Homocysteine and Oxidative Stress in Elderly Patients with Obstructive Sleep Apnea/hypopnea Syndrome显示文摘Objective Elderly patients with obstructive sleep apnea/hypopnea syndrome(OSAHS)has a higher risk of cardiovascular and cerebrovascular disease.However,changes of homocysteine(Hcy)as markers of cardiovascular and cerebrovascular disease associated with OSAHS and their mechanism have not been elucidated so far.This study aims to investigate the changes of both serum Hcy and oxidative stress and their possible links with OSAHS in elderly patients.Methods Based on polysomnogram(PSG)and age,83 patients with OSAHS were recruited and divided into elderly-OSAHS(n=32)and non-elderly OSAHS groups(n=51).Fifty two subjects without OSAHS were divided into elderly control(n=29)and non-elderly control groups(n=23).A total of 135 subjects were included in the present study.All subjects were recorded for PSG variables and the contents of homocysteine(Hcy),malonaldehyde(MDA),and glutathione(GSH)which were detected after sleep.Serum homocysteine was measured by cyclophorase.MDA and GSH were measured by spectrophotometer.Results(1)The serum levels of Hcy showed significant difference among the four groups(P<0.05).The concentrations of Hcy in elderly OSAHS patients were higher than in other groups,while those in the elderly control group were higher than in the non-elderly control the concentrations in the non-elderly OSAHS group were higher than in the non-elderly control.(2)The concentrations of MDA and GSH changed at an equal pace with Hcy in the four groups.(3)Multielement linearity regression analysis indicated a statistically significant relationship between Hcy concentration and age,MDA,GSH,and apnea hypopnea index(AHI).Conclusions(1)The concentrations of Hcy and oxidative stress have increased with advancing age.(2)The concentrations of Hcy and oxidative stress have further increased in the elderly patients with OSAHS.(3)Oxidative stress might cause high-level serum Hcy in the elderly patients with OSAHS. | LING WANG,JIE LI 2,YAN XIE,AND XUE-GUANG ZHANG Laboratory of Clinical Immunology,the First Affiliated Hospital of Soochow University,Suzhou 215006,Jiangsu,China | 2010 | Biomedical and Environmental Sciences2010，23，1： | 15 |

| 12 | Expression of hIGF-I in the silk glands of transgenic silkworms and in transformed silkworm cells显示文摘To express human insulin-like growth factor-I (hIGF-I) in transformed Bombyx mori cultured cells and silk glands, the transgenic vector pigA3GFP-hIGF-ie-neo was constructed with a neomycin resistance gene driven by the baculovirus ie-1 promoter, and with the hIGF-I gene under the control of the silkworm sericin promoter Ser-1. The stably transformed BmN cells expressing hIGF-I were selected by using the antibiotic G418 at a final concentration of 700-800 μg/mL after the BmN cells were transfected with the piggyBac vector and the helper plasmid. The specific band of hIGF-I was detected in the transformed cells by Western blot. The expression level of hIGF-I, determined by ELISA, was about 7800 pg in 5×105 cells. Analysis of the chromosomal insertion sites by inverse PCR showed that exogenous DNA could be inserted into the cell genome randomly or at TTAA target sequence specifically for piggyBac element transposition. The transgenic vector pigA3GFP-hIGF-ie-neo was transferred into the eggs using sperm-mediated gene transfer. Finally, two transgenic silkworms were obtained after screening for the neo and gfp genes and verified by PCR and dot hybridization. It is shown that while the solid fraction is 0.9, MnS inclusions began to precipitate in the final stage of solidification. The solidification process of 49MnVS3 non-quenched and tempered steel during heating has been observed in situ using a confocal scanning laser microscope (CSLM), which agrees well with the thermodynamics calculation.
