1Proliferation of intestinal crypt cells by gastrin-induced ornithine decarboxylase显示文摘AIM:to determine whether the gastrin stimulated intestinalcrypt cell(IEC-6)proliferation by induction of ornithinedecarboxylase(ODC).METHODS:IEC-6 cells were grown in DMEM containing50mL·L^(-1)dialyzed fetal bovine serum for 24h and then weretreated with gastrin.The proliferative capability of the cellswas monitored subsequently on d 1,2,3,and 4 aftertreatment with MTT assay at aborbance 570nm.The cellularODC mRNA expression,ODC activity,and putrescinecontent were examined by RT-PCR method,radiometrictechnique and high-performance liquid chromatography(HPLC)analysis respectively after 12h of treatment.RESULTS:On d1 after exposure of IEC-6 cells topentagastrin,the proliferation increased initially and reacheda peak on d3 at 250μg·L^(-1)concentration.Pentagastrin 500μg·L^(-1)increased cell proliferation on day 1 and day 2,andthen decreased,Compared with control group,pentagastrin250μg·L^(-1)increased ODC mRNA level by 1.09-fold(P<0.05),ODC activity by 1.71-fold(P<0.0]),and putrescinecontent 5.30-fold(P<0.01)respectively.Similarly,pentagastrin of 500μg·L^(-1)also increased ODC mRNA levelby 1.16-fold(P<0.05),ODC activity 1.63-fold(P<0.05),and putrescine content 4.4t-fold(P<0.01)respectively.But there was not significant difference between them.CONCLUSION:Gastrin is an agent which promotes IEC-6 cellproliferation involved in regulating ODC activity mechanism.Zi-Li Zhang Wei-Wen Chen Piei Institute,Guangzhou University of Traditional Chinese Medicine(TCM),Guangzhou 510405,China 2002World Journal of Gastroenterology2002,8,1:11
2Harmine induces apoptosis in HepG2 cells via mitochondrial signaling pathway显示文摘BACKGROUND:Harmine has antitumor and antinociceptive effects,and inhibits human DNA topoisomerase.However no detailed data are available on the mechanisms of action of harmine in hepatocellular carcinoma.This study aimed to investigate the effects of harmine on proliferation and apoptosis and the underlying mechanisms in the human hepatocellular carcinoma cell line HepG2.METHODS:The proliferation of HepG2 cells was determined by the cell counting kit-8 (CCK-8) assay and the clone formation test.The morphology of HepG2 cells was examined using fluorescence microscopy after Hoechst 33258 staining Annexin V/propidium iodide (PI) was used to analyze apoptosis and PI to analyze the cell cycle.Western blotting was used to assess expression of the apoptosis-regulated genes Bcl-2,Bax,Bcl-xl,Mcl-1,caspase-3,and caspase-9 Mitochondrial transmembrane potential (Ψ m) was determined using JC-1.RESULTS:Harmine inhibited the proliferation of HepG2 cells in a dose-dependent manner.Hoechst 33258 staining revealed nuclear fragmentation and chromosomal condensation,cell shrinkage,and attachment loss in HepG2 cells treated with harmine.The percentage of the sub/G1 fraction was increased in a concentration-dependent manner,indicating apoptotic cell death.PI staining showed that harmine changed the cell cycle distribution,by decreasing the proportion of cells inG0/G1 and increasing the proportion in S and G2/M.Harmine induced apoptosis in a concentration-dependent manner,with rates of 20.0%,32.7% and 64.9%,respectively.JC-1 revealed a decrease in Ψ m.Apoptosis of HepG2 cells was associated with caspase-3 and caspase-9 activation,down-regulation of Bcl-2,Mcl-1,and Bcl-xl,and no change in Bax.CONCLUSIONS:Harmine had an anti-proliferative effect in HepG2 cells by inducing apoptosis.Mitochondrial signal pathways were involved in the apoptosis.The cancer-specific selectivity shown in this study suggested that harmine is a promising novel drug for human hepatocellular carcinoma.Ming-Rong Cao,Qiang Li,Zhi-Long Liu,Hui-Hui Liu,Wei Wang,Xiao-Li Liao,Yun-Long Pan and Jian-Wei Jiang Department of General Surgery,First Affiliated Hospital,Jinan University,Guangzhou 510632,China Department of Anesthesiology,First Affiliated Hospital,Guangzhou University of TCM,Guangzhou 510632,China Department of Biochemistry,Medical College of Jinan University,Guangzhou 510632,China 2011Hepatobiliary & Pancreatic Diseases International2011,10,6:11
3Influence of BOL on hyaluronic acid,laminin and hyperplasia in hepatofibrotic rats显示文摘AIM: To study the anti-hepatofibrosis mechanism of Bie Jie Jian oral liquid (BOL).METHODS: The model was induced by subcutaneous injection of CCl4. BOL was administered and the change of serum hyaluronic acid (HA) and laminin (LN) was observed and the degeneration of liver cells and the degree of fibre hyperplasia analyzed. Changes of ultra micro-structure in liver cells were observed in some samples.RESULTS: HA was reduced in both the groups with Iow and high dosage of BOL, which showed a remarkable difference as compared with that of the model group ( Iow dosage group: 376.15 μg/L ± 35.48 μg/L vs 806.07 μg/L ± 98.49 μg/L, P < 0.05; high dosage group: 340.14 μg/L ± 30.18 μg/L vs 806.07μg/L± 98.49 μg/L, P<0.05). The LN content of Iow and high dosage group of BOL was lower than that of model group (Iow dosage group: 71.99 μg/L± 8.15 μg/L vs 133.94μg/L± 14.45 μg/L, P< 0.01; high dosage group: 71.68 ig/L± 11.62 μg/L vs 133.94 μg/L ± 14.45 μg/L, P<0.01) and colchicine group (Iow dosage group: 71.99 μg/L ± 8.15 μg/Lvs 118.28 μg/L ± 16.13 μg/L, P< 0.05; high dosage group:71.68 μg/L ± 11.62 μg/L vs 118.28 μg/L ± 16.13 μg/L,P<0.05). Examined by Ridit, BOL could reduce the degeneration and necrosis of liver cells (X2 = 11.99P<0.05), the degree of fibre hyperplasia (X2 = 13.24P<0.05) and the pathological change of ultra micro-structure as well.CONCLUSION: The BOL has certain therapeutic effect on the experiment hepatofibrosis. 4Jiang Tang Fang Long Jiao Nang in Treating Non-Insulin Dependent Diabetes Mellitus with Hearing Loss显示文摘Objective To study the efficacy and safety of the Jiang Tang Fang Long Jiao Nang (Hypoglycemic Anti-Deafness Capsule) in treating non-insulin dependent diabetes mellitus with hearing loss. Methods Two hundred ninety six patients with non-insulin dependent diabetes mellitus and hearing loss were randomly assigned to a treatment group(n=164, 208 ears) and a control group(n=132, 184 ears). Patients in the treatment group were treated with the Jiang Tang Fang Long Jiao Nang and supplement herbal preparations as indicated by traditional Chinese medicine dialectical assessment, while control patients received glibenclamide and conventional treatments for deafness. Hearing, fasting blood glucose(FBG), post-prandial blood glucose(PBG), 24 hour urine sugar, platelet function indices, blood superoxide dismutase(SOD) and lipid peroxides(LPO) levels, and symptom improvement were compared between the two groups. Results The rate of hearing improvement was 56.7% for the treatment group and 26.6% for the control. FBG, PBG and 24 hour urine sugar improved in both groups, but the last two were superior in the treatment group compared to the control. Symptoms improvement was also superior in the treatment group compared to the control. In patients receiving Jiang Tang Fang Long Jiao Nang treatment, platelet function indices, SOD and LPO were all improved, while only LOP improvement was noticed in control patients. No acute or long-term toxicity was demonstrated for the Jiang Tang Fang Long Jiao Nang in animal tests. The Jiang Tang Fang Long Jiao Nang lowered blood glucose and serum triglycerides in a rat model of alloxan-induced diabetes. 7THE EXTENSIVENESS AND SPECIFICITY OF EFFECT OF ELECTROACUPUNCTURE AT DIFFERENT ACUPOINTS ON NOCICEPTIVE RESPONSE OF CONVERGENT NEURONS IN TRIGEMINAL NUCLEUS CAUDALIS显示文摘Experiments were carried out on rats anaesthetized with uraethane. The sponta-neous discharges and nociceptive responses of convergent neurons in the right trigerninal nucleus cau-dalis(TNC) to noxious stimuli at receptive field (cheek) were recorded extracellularly with glass mi-cro-electrode. Electroacupuncture (EA ) was applied at bilateral ' Xiaguan' (ST 7 on face ) or 'Zusanli' (ST 36 on shank) acupoint with Iow (2V) and high (18V) intensity. The noclceptive re-sponse of convergent neurons in TNC could be inhihited by low intensity EA applied at 'Xiaguan' butnot 'Zusanlil', showing the specificity of acupoints. High intensity EA at either 'Xiaguan' or 'Zusan-li' also reduced the nociceptive responses, showing the analgesic extensiveness of acupoints. We sug-gest that 'the gate of control' mechanism plays a main role in low intensity EA and 'diffuse noxiousinhibitory controls' (DNIC) rnechanism does so in high intensity EA.The results suggest that we should pay attention to the location of acupoints,Xu Weidong, Liu Xiang. 16THE CO-RELATIONSHIP BETWEEN NEIGUAN POINT AND THE HEART AND REGULATION OF AMYGDALOID NUCLEI显示文摘In this present study, the ruodel of the rabbit with acute myocardial ischemia(AMI) was used to explore mechanisms of interrelation hetween Neiguan acupoint (PC 6) of the Peri-cardium Meridian and the heart hy employing physiological and morphologicaI methods. It wasdemonstrated that electroacupuncture (EA) at Neiguan (PC 6 ) couId raise the eIectrical excitahility ofischemic myocardium, lessen the dispersity of recovery excitability, correct the disorder of electricalactivity; protect myocardiac glycogen and phosphorylase and alleviate their depletion, increase contentof ribonuclei acid (RNA ), etc., indicating that EA could help oxidative rnetaholism carry out normal-Iy and irnProve nutritional state of the ischemic myocardiurn. The key point of EA action is probablyregulates the coronary microclrculation of heart and reclistributes myocardial blood flow.
