1Electro-mechanically controlled assembly of reconfigurable 3D mesostructures and electronic devices based on dielectric elastomer platforms显示文摘The manufacture of 3D mesostructures is receiving rapidly increasing attention,because of the fundamental significance and practical applications across wide-ranging areas.The recently developed approach of buckling-guided assembly allows deterministic formation of complex 3D mesostructures in a broad set of functional materials;with feature sizes spanning nanoscale to centimeter-scale.Previous studies mostly exploited mechanically controlled assembly platforms using elastomer substrates;which limits the capabilities to achieve on-demand local assembly;and to reshape assembled mesostructures into distinct 3D configurations.This work introduces a set of design concepts and assembly strategies to utilize dielectric elastomer actuators as powerful platforms for the electro-mechanically controlled 3D assembly.Capabilities of sequential-local loading with desired strain distributions allow access to precisely tailored 3D mesostructures that can be reshaped into distinct geometries,as demonstrated by experimental and theoretical studies of?30 examples.A reconfigurable inductive-capacitive radio-frequency circuit consisting of morphable 3D capacitors serves as an application example.Wenbo Pang Xu Cheng Haojie Zhao Xiaogang Guo Ziyao Ji Guorui Li Yiming Liang Zhaoguo Xue Honglie Song Fan Zhang Zheng Xu Lei Sang Wen Huang Tiefeng Li Yihui Zhang 2020National Science Review2020,7,2:8
2Preparation of AZO Nanoparticles, Ceramic Targets and Thin Films by a Co-precipitaition Method显示文摘We comprehensively study the co-precipitation preparation of aluminum doped zinc oxide(AZO) nanoparticles, ceramic target and thin fi lm deposition. The nanoparticles calcined below 700 ℃ possessed pure wurtzite structure of ZnO. When the calcination temperature exceeded 700 ℃, ZnAl_2O_4 phase appeared. The resistivity and relative density of the AZO target pressed from nanoparticles were 3×10^(-3) Ω?cm and 99.1%, respectively. The minimum resistivity of AZO thin films prepared by DC sputtering of the ceramic target reached 4.1×10^(–4) Ω?cm with the mobility of 33 cm^2/v?s and the carrier concentration of 4.5 ×10^(20) cm^(-3). 3Culture of patient-derived multicellular clusters in suspended hydrogel capsules for pre-clinical personalized drug screening显示文摘A personalized medication regimen provides precise treatment for an individual and can be guided by pre-clinical drug screening.The economical and high-efficiency simulation of the liver tumor microenvironment(TME)in a drug-screening model has high value yet challenging to accomplish.Herein,we propose a simulation of the liver TME with suspended alginate-gelatin hydrogel capsules encapsulating patient-derived liver tumor multicellular clusters,and the culture of patient-derived tumor organoids(PDTOs)for personalized pre-clinical drug screening.The hydrogel capsule offers a 3D matrix environment with mechanical and biological properties similar to those of the liver in vivo.As a result,18 of the 28 patient-derived multicellular clusters were successfully cultured as PDTOs.These PDTOs,along with hepatocyte growth factor(HGF)of non-cellular components,preserve stromal cells,including cancer-associated fibroblasts(CAFs)and vascular endothelial cells(VECs).They also maintain stable expression of molecular markers and tumor heterogeneity similar to those of the original liver tumors.Drugs,including cabazitaxel,oxaliplatin,and sorafenib,were tested in PDTOs.The sensitivity of PDTOs to these drugs differs between individuals.The sensitivity of one PDTO to oxaliplatin was validated using magnetic resonance imaging(MRI)and biochemical tests after oxaliplatin clinical treatment of the corresponding patient.Therefore,this approach is promising for economical,accurate,and high-throughput drug screening for personalized treatment.Haijiang Dong Zequn Li Suchen Bian Guangyuan Song Wenfeng Song Mingqi Zhang Haiyang Xie Shusen Zheng Xuxu Yang Tiefeng Li Penghong Song 2022Bioactive Materials2022,7,12:0
4Structure and transport mechanism of the human calcium pump SPCA1显示文摘Secretory-pathway Ca^(2+)-ATPases(SPCAs)play critical roles in maintaining Ca^(2+)homeostasis,but the exact mechanism of SPCAsmediated Ca^(2+)transport remains unclear.Here,we determined six cryo-electron microscopy(cryo-EM)structures of human SPCA1(hSPCA1)in a series of intermediate states,revealing a near-complete conformational cycle.With the aid of molecular dynamics simulations,these structures offer a clear structural basis for Ca^(2+)entry and release in hSPCA1.We found that hSPCA1 undergoes unique conformational changes during ATP binding and phosphorylation compared to other well-studied P-type II ATPases.In addition,we observed a conformational distortion of the Ca^(2+)-binding site induced by the separation of transmembrane helices 4L and 6,unveiling a distinct Ca^(2+)release mechanism.Particularly,we determined a structure of the long-sought CaE2P state of P-type IIA ATPases,providing valuable insights into the Ca^(2+)transport cycle.Together,these findings enhance our understanding of Ca^(2+)transport by hSPCA1 and broaden our knowledge of P-type ATPases.Mengqi Wu Cang Wu Tiefeng Song Kewu Pan Yong Wang Zhongmin Liu 2023Cell Research2023,33,7:0
