2Inhibition of 12-lipoxygenase reduces proliferation and induces apoptosis of hepatocellular carcinoma cells in vitro and in vivo显示文摘BACKGROUND:12-lipoxygenase(12-LOX) has been reported to be an important gene in cancer cell proliferation and survival,and tumor metastasis.However,its role in hepatocellular carcinoma(HCC) cells remains unknown.METHODS:Expression of 12-LOX was assessed in a diethylnitrosamine-induced rat HCC model,and in SMMC-7721,HepG2 and L-02 cells using immunohistochemical staining and reverse transcriptase-polymerase chain reaction(RT-PCR).GST-π and Ki-67 were determined in vivo by immunohistochemical staining.Apoptosis was evaluated by TUNEL assay.Cell viability and apoptosis were determined by MTT assay and flow cytometry,respectively.Apoptosis-related proteins in SMMC-7721 and HepG2 cells were detected by Western blotting.RESULTS:Immunohistochemical staining and RT-PCR showed that 12-LOX was over-expressed in rat HCC and two HCC cell lines,while the expression was inhibited by baicalein,a specific inhibitor of 12-LOX.Baicalein inhibited cell proliferation and induced apoptosis in rat HCC and both cell lines in a dose-and time-dependent manner.Our in vivo study demonstrated that baicalein also reduced neoplastic nodules.Mechanistically,baicalein reduced Bcl-2 protein expression coupled with a slight increase of the expression of Bax and activation of caspase-3.Furthermore,baicalein inhibited the activation of ERK-1/2(phosphorylated).Interestingly,the effects of baicalein were reversed by 12(S)-HETE,a metabolite of 12-LOX.CONCLUSIONS:Inhibition of 12-LOX leads to reduced numbers of HCC cells,partially caused by increased apoptosis.12-LOX may be a potential molecular target for HCC prevention and treatment.Xi-Ming Xu,Guang-Jin Yuan,Jun-Jian Deng,Hong-Ting Guo,Miao Xiang,Fang Yang,Wei Ge and Shi-You Chen Cancer Center, Renmin Hospital of Wuhan University, Wuhan 430060, China Cancer Center, the 82nd Hospital of the Chinese PLA, Huai’an 223001, China Department of Physiology, Medical College of Wuhan University, Wuhan 430071, China Department of Physiology & Pharmacology, University of Georgia, Athens, GA 30602, USA 2012Hepatobiliary & Pancreatic Diseases International2012,11,2:20

4Early Pleistocene hominid teeth recovered in Mohui cave in Bubing Basin, Guangxi, South China显示文摘Two hominid teeth recovered in Mohui cave are morphologically distinguished from Australopithecus in Af-rica, but close to Homo erectus in China. These teeth are therefore provisionally assigned to Homo erectus. The associ-ated mammalian fauna include Gigantopithecus blacki, Nes-toritherium sp., Sus xiaozhu, Sus peii and Ailuropoda microta, which are typical early Pleistocene taxa in South China. The general characteristics of the Mohui faunal assemblage are similar to the Longgupo site, which is dated to 2 Ma, imply-ing a contemporaneity for the two sites. To date, compared with the discoveries in Africa, far fewer early Pleistocene hominid fossils have been recovered in Asia, and there are intensive controversies concerning their stratigraphic prove-nience and typological and temporal positions. 6Efficacy and safety of Yunxiangjing derived from Chinese herbal medicine administered as an enema in the management of acute radiation-induced proctitis in patients with pelvic malignancy显示文摘OBJECTIVE: To assess the efficacy and safety of Yunxiangjing(YXJ), derived from Chinese herbal medicine, in the management of acute radiation-induced proctitis(ARIP) in patients with pelvic malignancy.METHODS: Forty-eight patients with grade 2 ARIP were administered YXJ as an enema(1∶30 dilution)for 2 weeks and followed up for 2 years. All were assessed for response and ARIP grade. Quality of life(QOL) was assessed with the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire-Core 30.RESULTS: Of the 48 patients, six(12.5%) achieved complete remission of ARIP and 28(58.3%) showed a decrease from grade 2 to grade 1 ARIP. No patient experienced a grade ≥3 toxicity. At the end of radiotherapy, patients showed significant improvements in QOL(P < 0.05). Two years after treatment, 46 patients showed no late toxicity, with only two experiencing grade 1 late toxicity.CONCLUSION: YXJ can be used as an enema to manage acute radiation-induced proctitis in cancer patients undergoing radiotherapy. 7Distribution and drug resistance of pathogenic bacteria in emergency patients显示文摘BACKGROUND Antibiotic resistance has become a global threat for human health,calling for rational use of antibiotics.AIM To analyze the distribution and drug resistance of the bacteria,providing the prerequisite for use of antibiotics in emergency patients.METHODS A total of 2048 emergency patients from 2013 to 2017 were enrolled.Their clinical examination specimens were collected,followed by isolation of bacteria.The bacterial identification and drug susceptibility testing were carried out.RESULTS A total of 3387 pathogens were isolated.The top six pathogens were Acinetobacter baumannii(660 strains),Staphylococcus aureus(436 strains),Klebsiella pneumoniae(347 strains),Pseudomonas aeruginosa(338 strains),Escherichia coli(237 strains),and Candida albicans(207 strains).The isolation rates of these pathogens decreased year by year except Klebsiella pneumoniae,which increased from 7.1%to 12.1%.Acinetobacter baumannii is a widely-resistant strain,with multiple resistances to imipenem,ciprofloxacin,minocycline and tigecycline.The Staphylococcus aureus had high resistance rates to levofloxacin,penicillin G,and tetracycline.But the susceptibility of it to vancomycin and tigecycline were 100%.Klebsiella pneumoniae had high resistance rates to imipenem,cefoperazone/sulbactam,amikacin,and ciprofloxacin,with the lowest resistance rate to tigecycline.The resistance rates of Pseudomonas aeruginosa to cefoperazone/sulbactam and imipenem were higher,with the resistance rate to amikacin below 10%.Besides,Escherichia coli had high resistance rates to ciprofloxacin and cefoperazone/sulbactam and low resistance rates to imipenem,amikacin,and tigecycline.CONCLUSION The pathogenic bacteria isolated from the emergency patients were mainly Acinetobacter baumannii,Staphylococcus aureus,Klebsiella pneumoniae,Pseudomonas aeruginosa,Escherichia coli,and Candida albicans.The detection rates of drugresistant bacteria were high,with different bacteria having multiple drug resistances to commonly used antimicrobial agents,guiding the rational use of drugs and reducing the production of multidrug-resistant bacteria.Wei Huai Qing-Bian Ma Jia-Jia Zheng Yang Zhao Qiang-Rong Zhai 2019World Journal of Clinical Cases2019,7,20:6

9Mineral phase and structure changes during roasting of fine-grained carbonaceous gold ores and their effects on gold leaching efficiency显示文摘While roasting has been widely applied to reduce the negative effect of carbonaceous matters on gold extraction from fine-grained carbonaceous gold ores, the phase and structure changes of minerals during roasting and their in fluences on the leachi ng rate of gold have not been fully understood. This limits the extraction of carbonaceous gold deposits. The current work examines the oxidation process of a fine-grained carbonaceous gold ore during roasting using a range of techniques including X-ray diffraction (XRD), seanning electron microscopy (SEM), Energy Dispersive Spectrometer (EDS) analysis and pore structure analysis together with gold leaching tests. The results show that during the process of oxidative roasting, the carbonaceous matters (organic carbon and graphitic carbon) and pyrite were completely decomposed at 600 ℃ with the carbonaceous components burned and pyrite oxidized into hematite. At 650 ℃, while dolomite was decomposed into calcia, magnesia, calcium sulfate etc., the calcine structure became loose and porous, leading to a high gold leaching rate from the roasted product. Above 750 ℃, the porous calcite structure started to collapse along with the agglomeration, leading to the secondary encapsulation of gold particles, which contributed to the sharp drop in the gold leaching rate of the roasted product. 11Systems pharmacology dissection of action mechanisms for herbs in osteoporosis treatment显示文摘Objective:Osteoporosis has become the biggest cause of non-fatal health issue.Currently,the limitations of traditional anti-osteoporosis drugs such as long-term ill-effects and drug resistance,have raised concerns toward complementary and alternative therapies,particularly herbal medicines and their natural active compounds.Thus,this study aimed to provide an integrative analysis of active chemicals,drug targets and interacting pathways of the herbs for osteoporosis treatment.Methods:Here,we introduced a systematic pharmacology model,combining the absorption,distribution,metabolism,and excretion(ADME)screening model,drug targeting and network pharmacology,to probe into the therapeutic mechanisms of herbs in osteoporosis.Results:We obtained 86 natural compounds with favorable pharmacokinetic profiles and their 58 targets from seven osteoporosis-related herbs.Network analysis revealed that they probably synergistically work through multiple mechanisms,such as suppressing inflammatory response,maintaining bone metabolism or improving organism immunity,to benefit patients with osteoporosis.Furthermore,experimental results showed that all the five compounds(calycosin,asperosaponin VI,hederagenin,betulinic acid and luteolin)enhanced osteoblast proliferation and differentiation in vitro,which corroborated the validity of this system pharmacology approach.Notably,gentisin and aureusidin among the identified compounds were first predicted to be associated with osteoporosis.Conclusion:Herbs and their natural compounds,being characterized as the classical combination therapies,might be engaged in multiple mechanisms to coordinately improve the osteoporosis symptoms.This work may contribute to offer novel strategies and clues for the therapy and drug discovery of osteoporosis and other complex diseases.Ying Huai Wen-juan Zhang Wei Wang Kai Dang Shan-feng Jiang Dan-ming Li Meng Li Qiang Hao Zhi-ping Miao Yu Li Ai-rong Qian 2021Chinese Herbal Medicines2021,13,3:3
12ATP-binding cassette transporter enhances tolerance to DDT in Tetrahymena显示文摘The reuse of dichlorodiphenyltrichloroethane(DDT) as an indoor residual spray was permitted by the World Health Organization in 2007, and approximately 14 countries still use DDT to control disease vectors. The extensive exposure of insects to DDT has resulted in the emergence of DDT resistance, especially in mosquitoes, and the mechanism for this resistance in mosquitoes has been widely reported. Spraying can also introduce DDT directly into surface water, and DDT can subsequently accumulate in microorganisms, but the mechanism for the resistance to DDT degradation in microorganisms is unclear. Using whole-genome microarray analysis, we detected an abcb15 gene that was up-regulated in a specific manner by DDT treatment in T. thermophile. The deduced ABCB15 peptide sequence had two transmembrane domains(TMDs) and two nucleotide-binding domains(NBDs) to form the structure TMD-NBD-TMD-NBD, and each NBD contained three conserved motifs: Walker-A, C-loop, and Walker-B, which indicated the T. thermophila abcb15 was a typical ABC transporter gene. The expression of ABCB15 fused with a C-terminal green fluorescent protein was found to be on the periphery of the cell, suggesting that ABCB15 was a membrane pump protein. In addition, cells with abcb15 partially knocked down(abcb15-KD) grew slower than wild-type cells in the presence of 256 mg L-1 DDT, indicating the tolerance of abcb15-KD strain to DDT exposure was decreased. 14Nonvolatile memristor based on heterostructure of 2D room-temperature ferroelectric α-In2Se3 and WSe2显示文摘Two-dimensional(2D) ferroelectricity is considered to have significant potential for information storage in the future. Semiconducting ferroelectrics that are stable at room temperature afford many possibilities for the assembly of various high-performance heterostructures and fabricating multifuntional devices.Herein, we report the synthesis of a stable van der Waals(vd W) single-crystal semiconductor α-In2 Se3.Piezoresponse force microscopy(PFM) measurements demonstrated the out-of-plane ferroelectricity in ~15 layers α-In2 Se3 at room temperature. Both ferroelectric domains with opposite polarization and the tested amplitude and phase curve proved that this semiconductor exhibits hysteresis behavior during polarization.In the α-In2Se3/WSe2 vertical heterostructure device, the switchable diode effect and nonvolatile memory phenomenon showed a high on/off ratio and a small switching voltage. The distinct resistance switches were further analyzed by band alignment of the heterostructure under different polarizations by first principle calculations. 16Crack status analysis for concrete dams based on measured entropy显示文摘The integrity and safety of concrete dams are seriously affected by the existing cracks in dam bodies, and some serious cracks may cause dam failure or disaster. The propagation of cracks in concrete dams is accompanied by changes in energy distribution, which can be represented by changes in the structure's system entropy. Therefore, the entropy theory can be used in analyzing the behavior of dam cracks. Due to the randomness and locality of crack propagation, it is difficult to predict the location of cracks by traditional monitoring methods. To solve this problem, the influence of spatial positions of monitoring points on inspection zones is represented by a weight index, and the weight index is determined by the distance measure method proposed in this paper. Through the weighted linear fusion method, the entropy of multiple monitoring points is obtained for analyzing the behavior of dam cracks in the selected zones. Meanwhile, the catastrophe theory is used as the variation criterion of an entropy sequence in order to predict the instability time of dam cracks. Case studies are put forward on a high arch dam, and the fusion entropy is calculated according to the monitoring data from strain gauges. 17Structural Damage Recognition Based on Perturbations of Curvature Mode Shape and Frequency显示文摘A new method for structural damage identification is presented based on perturbations of curvature mode shape and frequency.Firstly,the structure's mass and stiffness matrices are expressed as functions of its elements'physical parameters,which reflect their damage states. According to differences in the curvature mode shape of the structure in undamaged and damaged states,possible damage equations and determined damage equations are established and used to solve for the elements'damage parameters.Then,to ensure the accuracy of the recognition result,a reasonable solution is substituted into a damage-checking equation based on the change of frequency of the damaged structure.Numerical examples are used to show that,to identify the damage,only one order mode needs to be tested.When the degree of damage is low, first-order perturbation equations can be used to recognize the damage with sufficient accuracy.
