2Integrated network analyses identify MYB4R1 neofunctionalization in the UV-B adaptation of Tartary buckwheat显示文摘A hallmark of adaptive evolution is innovation in gene function,which is associated with the development of distinct roles for genes during plant evolution;however,assessing functional innovation over long periods of time is not trivial.Tartary buckwheat(Fagopyrum tataricum)originated in the Himalayan region and has been exposed to intense UV-B radiation for a long time,making it an ideal species for studying novel UV-B response mechanisms in plants.Here,we developed a workflow to obtain a co-functional network of UV-B responses using data from more than 10,000 samples in more than 80 projects with multi-species and multiomics data.Dissecting the entire network revealed that flavonoid biosynthesis was most significantly related to the UV-B response.Importantly,we found that the regulatory factor MYB4R1,which resides at the core of the network,has undergone neofunctionalization.In vitro and in vivo experiments demonstrated that MYB4R1 regulates flavonoid and anthocyanin accumulation in response to UV-B in buckwheat by binding to L-box motifs in the FtCHS,FtFLS,and FtUFGT promoters.We used deep learning to develop a visual discrimination model of buckwheat flavonoid content based on natural populations exposed to global UV-B radiation.Our study highlights the critical role of gene neofunctionalization in UV-B adaptation.Moyang Liu Wenjun Sun Zhaotang Ma Chaocheng Guo Jiahao Chen Qi Wu Xiyin Wang Hui Chen 2022Plant Communications2022,3,6:1
3Study on Isolation, Identification of A Petroleum Hydrocarbon Degrading Bacterium Bacillus fusiformis sp. and Influence of Environmental Factors on Degradation Efficiency显示文摘A hydrocarbon degrading bacterium KL2-13 was isolated from ten sites of oil contaminated soil in the Karamay oilfield. It was identified as the Bacillus fusiformis sp. bacterium based on its morphological and physiological characteristics and the 16S rDNA sequence analysis. The factors influencing the hydrocarbon degradation by the bacterium KL2-13 were determined. The test results have showed that the hydrocarbon degrading bacterium KL2-13 requires an optimum pH range of 6-8, and the optimum inoculation quantity is 3%. The low-concentration metal ions Fe2+, Mg2+ and Ca2+ can improve the degradation ability of the bacteria KL2-13.Atoo low concentration of Tween-80 does not show obvious promotion to the de- grading bacterium KL2-13, and an excessively high concentration can decrease the degradation ability of the bacterium, the best dosage of which is 2%. 5Is Participating in MOOC Forums Important for Students? A Data-driven Study from the Perspective of the Supernetwork显示文摘Purpose: Compared with traditional course materials used in the classroom, the massive open online course(MOOC) forum that delivers unlimited learning content to students has various advantages. Yet MOOC has also received criticism recently, notably the problem of extremely low participation rates in its discussion forums. This study aims to explore the correlation between forum activity and student course grade in MOOC, and identify more accurately the forum activity levels of participants and the quality of threads in MOOC.Design/Methodology/Approach: We crawled students' tests, final exams, exercises, discussions performance data and total scores from a course in Chinese College MOOC from May 2014 to August 2014. And we use the data to analyze the correlation between Forum Participation and Course Performance based on nonparametric tests as well as multiple linear regressions with the software of R. The study provides definitions and algorithms of super degrees based on the supernetwork model to help find high-quality threads and active participants.Findings: A positive correlation between forum activity and course grade is found in this study. Students who participate in the forum have better performance than those who do not. Using the definitions and algorithms of super degrees in the supernetwork, forum activity levels of participants as well as the quality of threads they employ are identified. Research limitation: Only limited representative forum participants and threads are used to analyze the activity level and significance of the MOOC forum. Also, the study only investigates one Chinese course on information retrieval. More data and more data sources could be helpful in better understanding the MOOC forum phenomenon.Practical implications: As super degrees can reveal more latent information and recognize high-quality threads as well as active participants, these parameters can be used to assess needs to improve forum settings and alleviate the problem of low forum participation. The proposed super degrees can be applied in social network domains for further research.Originality/Value: Definitions and algorithms of super degrees are provided and used for forum analysis.
