| 1 | Two RING-Finger Ubiquitin E3 Ligases Regulate the Degradation of SPX4, An Internal Phosphate Sensor, for Phosphate Homeostasis and Signaling in Rice显示文摘SPX-domain-containing proteins (SPXs) play an important role in inorganic phosphate (Pi) sensing,signaling,and transport in eukaryotes.In plants,SPXs are known to integrate cellular Pi status and negatively regulate the activity of Pi central regulators,the PHOSPATE STARVATION RESPONSE proteins (PHRs).The stability of SPXs,such as SPX4,is reduced under Pi-deficient conditions.However,the mechanisms by which SPXs are degraded remain unclear.In this study,using a yeast-twhybrid screen we iden.tified two RING-finger ubiquitin E3 ligases regulating SPX4 degradation,designated SDEL1 and SDEL2,which were post-transcriptionally induced by Pi starvation.We found that both SDELs were located in the nucleus and cytoplasm,had ubiquitin E3 ligase activity,and directly ubiquitinated the K^213 and K^299 lysine residues in SPX4 to regulate its stability.Furthermore,we found that PHR2,a Pi central regulator in rice,could compete with SDELs by interacting with SPX4 under Pi-sufficient conditions,which protected SPX4 from ubiquitination and degradation.Consistent with the biochemical function of SDEL1 and SDEL2,overexpression of SDEL1 or SDEL2 resulted in Pi overaccumulation and induced Pi-starvation signaling even under Pi-sufficient conditions.Conversely,their loss-of-function mutants displayed decreased Pi accumulation and reduced Pi-starvation signaling.Collectively,our study revealed that SDEL1 and SDEL2 facilitate the degradation of SPX4 to modulate PHR2 activity and regulate Pi homeostasis and Pi signaling in response to external Pi availability in rice. | Wenyuan Ruan Meina Guo Xueqing Wang Zhenhui Guo Zhuang Xu Lei Xu Hongyu Zhao Haiji Sun Chengqi Yan Keke Yi | 2019 | Molecular Plant2019,12,8: | 14 |

| 3 | Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments,therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival,proliferation,apoptosis,autophagy of cardiomyocytes,endothelial cells,fibroblasts,monocytes,and stem cells.These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis,for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog,etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence,drug therapy,gene therapy,protein therapy,cell therapy,and exosome therapy have been emerging and are known as novel therapies.In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways,which contribute to inhibiting cardiomyocytes death,attenuating inflammation,enhancing angiogenesis,etc.so as to repair and re-functionalize damaged hearts. | Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 11 |
| 4 | Reworking effects in the Holocene Zhuye Lake sediments:A case study by pollen concentrates AMS ^(14)C dating显示文摘Transported by wind and water,the relatively old sediments can deposit in the terminal lake of an inland drainage basin.The reworking effect can affect the lake sediments 14 C dating and explanations for proxies.The Zhuye Lake is the terminal lake of the Shiyang River Basin.Previous studies indicated that sediments in different locations of the lake basin showed different climatic change patterns.And then,some radiocarbon dates were inverted for some Late Pleistocene sections.Whether this phenomenon is related with the reworking effect? | 5 | Effects of Target and Distractor Saturations on the Cognitive Performance of an Integrated Display Interface显示文摘Color coding is often used to enhance decision quality in complex man-machine interfaces of integrated display systems. However, people are easily distracted by irrelevant colors and by the numerous data points and complex structures in the interface. Although an increasing number of studies are seriously focusing on the problem of achieving efficient color coding, few are able to determine the effects of target and distractor saturations on cognitive performance. To study the performances of target colors among distractors, a systematic experiment is conducted to assess the influence of high and low saturated targets on cognitive performance, and the affecting extent of different saturated distractors of homogeneous colors on targets. According to the analysis of the reaction time through the non-parametric statistical method, a calculation method of the cognitive performance of each color is proposed. Based on the calculation of the color differences and the accumulation of the reaction times, it is shown that with the different saturated distractors of homogeneous colors, the high saturated yellow targets perform better than the low saturated ones, and the green and blue targets have moderate performances. When searching for a singleton target placed on a black background, the color difference between the target and the distractor should be more than 20ΔE*ab units in the yellow saturation coding, whereas the color difference should be more than 40ΔE*ab units in the blue and green saturation coding. In addition, as regards saturation coding, the influence of the color difference between the target and the background on cognitive performance is greater than that of the color difference between the target and the distractor. Seemingly, the hue attribute determines whether the saturation difference between the target and the distractor affects the cognitive performance. Based on the experimental results, the simulation design of the instrument dials in a flight situation awareness interface is completed and tested. | 6 | Improvement in grain refinement efficiency of Mg-Zr master alloy for magnesium alloy by friction stir processing显示文摘Previous studies have proved that the zirconium(Zr)alloying and grain refining performance of a Mg-Zr master alloy on Mg alloy is closely related to the distribution of Zr particle size,and a Mg-Zr master alloy with more Zr particles in size range of 1-5μm exhibits a better refining efficiency.In this paper,friction stir processing(FSP)was used to modify the Zr particles size distribution of a commercially available Mg-30 wt.%Zr master alloy,and the subsequent grain refinement ability was studied by trials on a typical Mg-3Nd-0.2Zn-0.6Zr(wt.%,NZ30K)alloy.It is found that plenty of large Zr particles in the as-received Mg-30%Zr master alloy are broken by FSP.Grain refinement tests reveal that the refining efficiency of Mg-30%Zr alloy is significantly improved by FSP,which is attributed to the better distribution of Zr particles.The refinement effect by adding 0.6%FSP-ed Mg-30%Zr is approximately equivalent to that by adding 1.0%as-received Mg-30%Zr.Due to the easy and convenient operation of FSP,this study provides a new method to develop a more efficient Mg-Zr refiner. | Chengqi Wang Ming Sun Feiyan Zheng Liming Peng Wenjiang Ding | 2014 | Journal of Magnesium and Alloys2014,2,3: | 7 |
| 7 | Early Holocene environment at a key location of the northwest boundary of the Asian summer monsoon: a synthesis on chronologies of Zhuye Lake, Northwest China显示文摘The intensified monsoon increases summer rainfall and creates wet conditions in the Asian summer monsoon region during the early Holocene. Along with millennial-scale changes of the monsoon intensity, it is still unclear whether the boundary of the monsoon region changes according to monsoon variability. Investigations into the early Holocene environment in monsoon marginal zones are crucial for understanding the monsoon boundary changes. Zhuye Lake is located at the northwest edge of the Asian summer monsoon, the northern Qilian Mountains, which are less affected by modern summer monsoon water vapor. Previous studies have reached different conclusions regarding the early Holocene climatic and environmental changes based on different dating methods(14C and OSL(optically stimulated luminescence)) and materials(shells, carbonate, pollen concentrates and bulk organic carbon). In this study, we synthesized 102 14C dates and 35 OSL dates from ten Holocene sedimentary sections and ten paleo-shorelines in the lake basin. A comparison between ages from different dating methods and materials generally shows that carbon reservoir effects are relatively slight in Zhuye Lake while the disordered chronologies are mainly related to the erosion processes and reworking effects. In addition, proxy data, including lithology, pollen, total organic carbon and carbonate, were collected from different sites of Zhuye Lake. According to the new synthesis, the early Holocene environment was relatively humid, associated with high runoff and lake water levels. The result indicates that the monsoon boundary moves to the north during the period of the intensified monsoon. A typical arid-area lake was formed during the mid-Holocene when carbonate accumulation and high organic matter contents were the main features of this period. The lake retreated strongly during the late Holocene, showing a drought trend. | 13 | Characterization and Proteomic Analysis of Novel Rice Lesion Mimic Mutant with Enhanced Disease Resistance显示文摘Lesion mmic mutants(LMMs)are plants that spontaneously form lesions without pathogeninfection or external stimulus and exhibit resistance to pathogens.Here,a rice LMM was created by ethylmethane sulfonate mutagenesis,named as hpil(hydrogen peroxide induced lesion).Diaminobenzidineand trypan blue staining showed that large amounts of H_(2)O_(2) were produced and cell death was occurredat and around the parts of lesion mimic in the rice leaves.The phenotype of hpil is controlled by a singlerecessive gene,localized at a 2 Mb interval on chromosome 2.The data suggested that hpil is a novelLMM with enhanced bacterial and fungal disease resistance,and multiple pathogenesis-related proteins(PRs)were up-regulated.The proteomes of leaves at three positions(different degrees of lesion mimicseverity)were characterized in hpil compared with its wild type plant.Differentially expressed proteinswere detected by two dimensional difference gel electrophoresis and 274 proteins were identified byMALDITOF/TOFTM.These proteins were related to metabolic process,cellular process and response tostimulus,with mostly down-regulated in hpil leaves.Many of these proteins were related to the Calvincycle,photosynthetic electron transport chain,glycolysis/gluconeogenesis and phosphonates pathways.Some resistance-related proteins including 14-3-3 proteins,OsPR10 and antioxidases such asperoxidase,superoxide dismutase and ascorbate peroxidase were up-regulated in leaves with lesionmimic.These results provide the foundation for cloning of the target gene and shed light on themechanism involved in autaimmunity of rice. | Yang Yong Lin Qiujun Chen Xinyu Liang Weifang Fu Yuwen Xu Zhengjin Wu Yuanhua Wang Xuming Zhou Jie Yu Chulang Yan Chengqi Mei Qiong Chen Jianping | 2021 | Rice science2021,28,5: | 2 |

| 15 | Sequence Signatures of Nucleosome Positioning in Caenorhabditis elegans显示文摘Our recent investigation in the protist Trichomonas vaginalis suggested a DNA sequence periodicity with a unit length of 120.9 nt, which represents a sequence signature for nucleosome positioning. We now extended our observation in higher eukaryotes and identified a similar periodicity of 175 nt in length in Caenorhabditis elegans. In the process of defining the sequence compositional characteristics, we found that the 10.5-nt periodicity, the sequence signature of DNA double helix, may not be sufficient for cross-nucleosome positioning but provides essential guiding rails to facilitate positioning. We further dissected nucleosome-protected sequences and identified a strong positive purine (AG) gradient from the 5′-end to the 3′-end, and also learnt that the nucleosome-enriched regions are GC-rich as compared to the nucleosome-free sequences as purine content is positively correlated with GC content. Sequence characterization allowed us to develop a hidden Markov model (HMM) algorithm for decoding nucleosome positioning computationally, and based on a set of training data from the fifth chromosome of C. elegans, our algorithm predicted 60%-70% of the well-positioned nucleosomes, which is 15%-20% higher than random positioning.
