1Effect of burden and origin sites of premature ventricular contractions on left ventricular function by 7-day Holter monitor显示文摘Recent studies have shown that premature ventricular contractions(PVCs) could enlarge the heart,but its risk factors are incompletely understood as a single 24-hour recording cannot reflect the true PVC burden due to day-to-day variability.Our purpose was to investigate the effect of burden and origin sites on left ventricular(LV) function in patients with PVCs by 7-day Holter electrocardiography(ECG).From May 2012 to August2013,112 consecutive patients with PVCs were recruited from the authors' affiliated hospital.All patients received2-dimensional transthoracic echocardiography,12-lead routing ECG and 7-days Holter ECG.Serum N-terminal probrain natriuretic peptide(NT-proBNP) levels were measured.A total of 102 participants with PVCs were included in the final analysis.Origin of PVCs from the tricuspid annulus had the highest burden and NT-proBNP level.LV papillary muscle had a higher LV ejection fraction(EF) level and a lower LV end-systolic dimension(ESD) than other PVC foci(P<0.05).The high burden group had a higher LV end-diastolic dimension(EDD) and LVESD but lower LVEF than the other two groups(P<0.05).Female,older age,physical work,and history of PVCs had a significantly positive correlation with symptoms.Male,older age,physical work,and high burden were positive predictors of enlarged LVEDD,LVESD and higher serum NT-proBNP level,but lower LVEF.Seven-day dynamic ECG Holter monitor showed the true PVC burden on patients with PVCs.PVCs with a lower burden or origin from the LV papillary muscle and the fascicle were relatively benign,while PVCs with a higher burden or origin from the tricuspid annulus may lead to cardiac dysfunction.Wenhua Xu Mingfang Li Minglong Chen Bing Yang Daowu Wang Xiangqing Kong Hongwu Chen Weizhu Ju Kai Gu Kejiang Cao Hailei Liu Qi Jiang Jiaojiao Shi Yan Cui Hong Wang 2015The Journal of Biomedical Research2015,29,6:4
2Structural biology of SARS-CoV-2:open the door for novel therapies显示文摘Severe Acute Respiratory Syndrome Coronavirus-2(SARS-CoV-2)is the causative agent of the pandemic disease COVID-19,which is so far without efficacious treatment.The discovery of therapy reagents for treating COVID-19 are urgently needed,and the structures of the potential drug-target proteins in the viral life cycle are particularly important.SARS-CoV-2,a member of the Orthocoronavirinae subfamily containing the largest RNA genome,encodes 29 proteins including nonstructural,structural and accessory proteins which are involved in viral adsorption,entry and uncoating,nucleic acid replication and transcription,assembly and release,etc.These proteins individually act as a partner of the replication machinery or involved in forming the complexes with host cellular factors to participate in the essential physiological activities.This review summarizes the representative structures and typically potential therapy agents that target SARS-CoV-2 or some critical proteins for viral pathogenesis,providing insights into the mechanisms underlying viral infection,prevention of infection,and treatment.Indeed,these studies open the door for COVID therapies,leading to ways to prevent and treat COVID-19,especially,treatment of the disease caused by the viral variants are imperative.Weizhu Yan Yanhui Zheng Xiaotao Zeng Bin He Wei Cheng 2022Signal Transduction and Targeted Therapy2022,7,2:2
3Large Stable Photoinduced Anisotropy and Optical Information Storage in Poly [4-bis (N, N-oxyethylene) imino-4- nitroasobenzene Succinyl] Film by Two-Wave Mixing显示文摘Optical information storage caused by photoinduced anisotropy in poly[4'-bis(N,N-oxyethylene)imino-4-nitroazobenzene succinyl]film is studied by two-wave mixing.Using Nd:YAG second-harmonic pulses(100ps),grating of orientation of azobenzene molecules is formed in the film by photoinduced anisotropy.Up to third-order diffraction was observed.After turning off one of the two pump lights,the signals remained.When the weaker one of two pump lights was turned off,the intensities of diffraction beams of another pump light almost did not decrease.The information recorded in the film can be 100%maintained for a long time(monitored over 100 days without obvious decrease).WEI Xiong YAN Xingzhong LIN Weizhu MO Dang LIANG Zhaoxi 1995Chinese Physics Letters1995,12,12:0
4Epicardial adipose tissue around the superior vena cava: A single center study of factors related to atrial fibrillation显示文摘The superior vena cava(SVC)is the main component of non-pulmonary vein(PV)ectopy in patients with atrial fibrillation(AF).Researchers have found that epicardial adipose tissue(EAT)volume is related to the AF substrate,which can be defined by the low voltage area(LVA).This study aimed to investigate the relationship between SVC-EAT and SVC-AF.Twenty-six patients with SVC-AF triggers were identified as the SVC-AF group.Other three groups were defined and included as the LVA-AF group(LVA>5%),non-LVA-AF group(LVA<5%),and physical examination(PE)group.EAT around left atrium(LA-EAT)and SVC-EAT volumes were obtained using a cardiac risk assessment module.According to the SVC/LA-EAT ratio,there are significant differences between the SVC-AF group and the three control groups(the SVC-AF group 0.092±0.041 vs.the LVA-AF group 0.054±0.026,the non-LVA-AF group 0.052±0.022,and the PE group 0.052±0.019,all P<0.001).Receiver operating characteristic curve analysis suggests the optimal cut-off point of SVC/LA-EAT ratio is 6.8%for detecting SVC-AF patients,with 81.1%sensitivity,73.1%specificity,and an area under the curve of 0.83(95%confidence interval,0.75-0.91).Those with SVC-AF have a higher SVC/LA-EAT ratio and empirical SVC isolation could be considered if the SVC/LA-EAT ratio was over 6.8%.Xingxing Sun Jun Wang Qing Yan Weizhu Ju Fengxiang Zhang Gang Yang Kai Gu Hailei Liu Zidun wang Xiaohong Jiang Mingfang Li Di Xu Yi Xu Hongwu Chen Minglong Chen 2022The Journal of Biomedical Research2022,36,6:0
5Crystal structure and catalytic mechanism of the MbnBC holoenzyme required for methanobactin biosynthesis显示文摘Methanobactins(Mbns)are a family of copper binding peptides involved in copper uptake by methanotrophs,and are potential therapeutic agents for treating diseases characterized by disordered copper accumulation.Mbns are produced via modification of MbnA precursor peptides at cysteine residues catalyzed by the core biosynthetic machinery containing MbnB,an iron-dependent enzyme,and MbnC.However,mechanistic details underlying the catalysis of the MbnBC holoenzyme remain unclear.Here,we present crystal structures of MbnABC complexes from two distinct species,revealing that the leader peptide of the substrate MbnA binds MbnC for recruitment of the MbnBC holoenzyme,while the core peptide of MbnA resides in the catalytic cavity created by the MbnB-MbnC interaction which harbors a unique tri-iron cluster.Ligation of the substrate sulfhydryl group to the tri-iron center achieves a dioxygen-dependent reaction for oxazolone thioamide installation.Structural analysis of the MbnABC complexes together with functional investigation of MbnB variants identifed a conserved catalytic aspartate residue as a general base required for MbnBC-mediated MbnA modifcation.Together,our study reveals the similar architecture and function of MbnBC complexes from different species,demonstrating an evolutionarily conserved catalytic mechanism of the MbnBC holoenzymes.Chao Dou Zhaolin Long Shoujie Li Dan Zhou Ying Jin Li Zhang Xuan Zhang Yanhui Zheng Lin Li Xiaofeng Zhu Zheng Liu Siyu He Weizhu Yan Lulu Yang Jie Xiong Xianghui Fu Shiqian Qi Haiyan Ren She Chen Lunzhi Dai Binju Wang Wei Cheng 2022Cell Research2022,32,3:0
