1Arginine, N-carbamylglutamate,and glutamine exert protective effects against oxidative stress in rat intestine显示文摘The objective of the current study is to evaluate the effects of dietary supplementation with arginine(ARG), N-carbamylglutamate(NCG), and glutamine(GLN) on rat intestinal morphology and antioxidant status under oxidative stress. Rats were fed for 30 d with one of the following iso-nitrogenous diets:basal diet(BD), BD plus 1% ARG, BD plus 0.1% NCG, and BD plus 1% GLN. On day 28, half of the rats fed BD were intraperitoneally injected with 12 mg/kg body weight of diquat(DT; i.e., the DT group) and the other half was intraperitoneally injected with sterile solution(i,e., the control group). The other diet groups were intraperitoneally injected with 12 mg/kg body weight of DT(i,e., DT + 1% GLN [DT + GLN],DT + 1% ARG [DT + ARG], and DT + 0.1% NCG [DT + NCG]). Rat jejunum samples obtained at 48 h after DT injection were analyzed. Results showed that DT significantly decreased catalase(CAT) activity and glutathione(GSH) content by 58.25% and 56.57%, respectively, and elevated malondialdehyde(MDA)content and crypt depth(CD) by 19.39% and 22.13%, respectively, in the jejunum(P < 0.05, relative to the control group). Compared with the DT group, the DT + GLN group exhibited significantly improved villus height(VH), villus width(VW), villus surface area(VSA), CD and total antioxidant capacity(T-AOC) activity(P < 0.05); the DT + ARG group exhibited significantly increased the ratio of VH to CD(H:D) and TAOC activity(P < 0.05); the DT + GLN, DT + ARG and DT + NCG groups exhibited significantly enhanced CAT activity and GSH content as well as decreased MDA content(P < 0.05). Moreover, VH, VW, VSA, CD and GSH content in the DT + GLN group were higher whereas MDA content was lower compared with the corresponding values observed in both the DT + ARG and the DT + NCG groups(P < 0.05). The H:D ratio in the DT + ARG group significantly increased compared with that in the DT + NCG and DT + GLN groups(P < 0.05). 2Roles of dietary supplementation with arginine or N-carbamylglutamate in modulating the inflammation, antioxidant property, and m RNA expression of antioxidant-relative signaling molecules in the spleen of rats under oxidative stress显示文摘This study evaluated the effects of arginine(Arg) or N-carbamylglutamate(NCG) on inflammation, antioxidant property, and antioxidant-related gene expression in rat spleen under oxidative stress. A total of 52 rats were randomly distributed into 4 treatment groups with 13 replicates per group. Rats were fed a basal diet(BD) or BD supplemented with Arg or NCG for 30 days. On day 28, half of the BD-fed rats were intraperitoneally injected with sterile saline(control group), and the other half with 12 mg/kg body weight of diquat(DT; DT group). The other 2 diet groups were intraperitoneally injected with 12 mg/kg body weight of DT with either Arg(1%)(DT t Arg) or NCG(0.1%)(DT t NCG). Rat spleen samples were collected for analysis at 48 h after DT injection. Results showed that DT damaged the antioxidant defense in rats compared with the control group(P < 0.05). Compared with the DT group, the DT t Arg and DT t NCG groups manifested improved anti-hydroxyl radical, catalase, and total superoxide dismutase(T-SOD) activities, increased glutathione content(P < 0.05), and decreased malondialdehyde content(P < 0.05).Moreover, compared with the DT group, the DT t Arg and DT t NCG groups enhanced mRNA expression of superoxide dismutase(SOD), glutathione peroxidase 1(GPx1), glutathione reductase(GR), nuclear factor erythroid 2-related factor 2(Nrf2), Kelch-like ECH-associated protein 1(Keap-1), and mammalian target of rapamycin(m TOR)(P < 0.05). Both NCG and Arg significantly increased anti-inflammatory cytokine mRNA level but suppressed the pro-inflammatory cytokine mRNA expression under oxidative stress(P < 0.05). In summary, NCG and Arg effectively alleviated oxidative stress, improved the antioxidant capacity and regulated the antioxidant-related signaling molecular expression in rat spleen. 3New insights into the role of spermine in enhancing the antioxidant capacity of rat spleen and liver under oxidative stress显示文摘Oxidative stress can damage cellular antioxidant defense and reduce livestock production efficiency.Spermine is a ubiquitous cellular component that plays important roles in stabilizing nucleic acids,modulating cell growth and differentiation, and regulating ion channel activities. Spermine has the potential to alleviate the effects of oxidative stress. However, to date no information is available about the effect of spermine administration on antioxidant property of the liver and spleen in any mammalian in vivo system. This study aims to investigate the protective effect of spermine on rat liver and spleen under oxidative stress. Rats received intragastric administration of either 0.4 μmol/g body weight of spermine or saline once a day for 3 days. The rats in each treatment were then injected with either diquat or sterile saline at 12 mg/kg body weight. Liver and spleen samples were collected 48 h after the last spermine ingestion.Results showed that regardless of diquat treatment, spermine administration significantly reduced the malondialdehyde(MDA) content by 23.78% in the liver and by 5.75% in the spleen, respectively(P < 0.05).Spermine administration also enhanced the catalase(CAT) activity, anti-hydroxyl radical(AHR) capacity and glutathione(GSH) content by 38.68%, 15.53% and 1.32% in the spleen, respectively(P< 0.05). There were interactions between spermine administration and diquat injection about anti-superoxide anion(ASA),AHR capacity, CAT activity, GSH content, and total antioxidant capacity(T-AOC) in the liver and about ASA capacity and T-AOC in the spleen of weaned rats(P < 0.05). Compared with the control group, spermine administration significantly increased the AHR capacity, CAT activity, GSH content, and T-AOC by 40.23%,31.15%, 30.25%, 35.37% in the liver, respectively(P < 0.05) and increased the T-AOC by 8% in the spleen of weaned rats(P < 0.05). 4Effects of saccharicterpenin on antioxidant status and urinary metabolic profile of rats显示文摘Saccharicterpenin is a new green additive agent that is derived from the extract of Theaceae plants and has the ability to improve immunity and meat quality, increase the digestive enzyme activity, and enhance the intestinal development and growth of animals. However, the antioxidant status and systematic changes in metabolic biochemistry associated with saccharicterpenin supplementation in animals are still unknown. This study examined the effects of saccharicterpenin on the antioxidant status and urinary metabolic profile of rats. Sixteen rats were randomly distributed to 2 groups. One group was treated with 400 mg/kg body weight of saccharicterpenin, and the other group was treated with equal amount of saline. Results revealed that saccharicterpenin significantly increased the capacities of antihydroxyl radical(13.18%) and anti-superoxide anion(14.36%), the total antioxidant capacity(48.27%),and the activities of total superoxide dismutase(3.68%), catalase(21.52%), glutathione peroxidase(5.83%)and glutathione S-transferase(29.59%)(P < 0.05). By contrast, the contents of malondialdehyde and glutathione were not significantly affected by saccharicterpenin(P > 0.05). Saccharicterpenin supplementation significantly increased the urinary levels of bile acids, ethanol. α-ketoglutarate, and a-hydroxy butyrate but decreased the level of N-acetylglutamate(P < 0.05). 5Spermine protects intestinal barrier integrity through ras-related C3 botulinum toxin substrate 1/phospholipase C-γ1 signaling pathway in piglets显示文摘Weaning stress can cause tight junctions damage and intestinal permeability enhancement,which leads to intestinal imbalance and growth retardation,thereby causing damage to piglet growth and development.Spermine can reduce stress.However,the mechanism of spermine modulating the intestinal integrity in pigs remains largely unknown.This study aims to examine whether spermine protects the intestinal barrier integrity of piglets through ras-related C3 botulinum toxin substrate 1(Rac1)/phospholipase C-g1(PLC-γ1)signaling pathway.In vivo,80 piglets were categorised into 4 control groups and 4 spermine groups(10 piglets per group).The piglets were fed with normal saline or spermine at 0.4 mmol/kg BW for 7 h and 3,6 and 9 d.In vitro,we investigated whether spermine protects the intestinal barrier after a tumor necrosis factor a(TNF-a)challenge through Rac1/PLC-γ1 signaling pathway.The in vivo study found that spermine supplementation increased tight junction protein mRNA levels and Rac1/PLC-γ1 signaling pathway gene expression in the jejunum of piglets.The serum D-lactate content was significantly decreased after spermine supplementation(P<0.05).The in vitro study found that 0.1 mmol/L spermine increased the levels of tight junction protein expression,Rac1/PLC-γ1 signaling pathway and transepithelial electrical resistance,and decreased paracellular permeability(P<0.05).Further experiments demonstrated that spermine supplementation enhanced the levels of tight junction protein expression,Rac1/PLC-γ1 signaling pathway and transepithelial electrical resistance,and decreased paracellular permeability compared with the NSC-23766 and U73122 treatment with spermine after TNF-a challenge(P<0.05).Collectively,spermine protects intestinal barrier integrity through Rac1/PLC-γ1 signaling pathway in piglets.Guangmang Liu Xiaomei Xu Caimei Wu Gang Jia Hua Zhao Xiaoling Chen Gang Tian Jingyi Cai Jing Wang 2022Animal Nutrition2022,,1:2
6Transfer of β-hydroxy-β-methylbutyrate from sows to their offspring and its impact on muscle fiber type transformation and performance in pigs显示文摘Background: Previous studies suggested that supplementation of lactating sows with β-hydroxy-β-methylbutyrate(HMB) could improve the performance of weaning pigs, but there were little information in the muscle fiber type transformation of the offspring and the subsequent performance in pigs from weaning through finishing in response to maternal HMB consumption. The purpose of this study was to determine the effect of supplementing lactating sows with HMB on skeletal muscle fiber type transformation and growth of the offspring during d 28 and180 after birth. A total of 20 sows according to their body weight were divided into the control(CON, n = 10) or HMB groups(HMB, n = 10). Sows in the HMB group were supplemented with β-hydroxy-β-methylbutyrate calcium(HMB-Ca) 2 g/kg feed during d 1 to 27 of lactation. After weaning, 48 mixed sex piglets were blocked by sow treatment and fed standard diets for post-weaning, growing, finishing periods. Growth performance was recorded during d 28 to 180 after birth. Pigs were slaughtered on d 28(n = 6/treatment) and 180(n = 6/treatment) postnatal and the longissimus dorsi(LD) was collected, respectively.Results: The HMB-fed sows during lactation showed increased HMB concentration(P < 0.05) in milk and LD of weaning piglets(P < 0.05). In addition, offsprings in HMB group had a higher finishing BW and lean percentage than did pigs in CON group(P < 0.05), meanwhile, compared with pigs from sows fed the CON diet, pigs from sows fed HMB diet showed higher type Ⅱ muscle fiber cross-sectional area(CSA), elevated myosin heavy chain(MyHC) Ⅱb and Sox6 mRNA, and fast-MyHC protein levels in LD(P < 0.05).Conclusions: HMB supplemented to sow diets throughout lactation increases the levels of HMB in maternal milk and skeletal muscle of pigs during d 28 after birth and promotes subsequent performance of pigs between d 28 and 180 of age by enhancing glycolytic muscle fiber transformation.Haifeng Wan Jiatao Zhu Caimei Wu Pan Zhou Yong Shen Yan Lin Shengyu Xu Lianqiang Che Bin Feng Jian Li Zhengfeng Fang De Wu 2017Journal of Animal Science and Biotechnology2017,8,2:2

9An emerging role of arecoline on growth performance,intestinal digestion and absorption capacities and intestinal structural integrity of adult grass carp(Ctenopharyngodon idella)显示文摘Arecoline is an alkaloid with important pharmacological effects in the plant areca nut,which has been demonstrated to be an agonist of muscarinic receptors(M receptor).This study explored the influences of dietary arecoline on growth performance,intestinal digestion and absorption abilities,antioxidant capacity,and the apical junction complex(AJC)of adult grass carp(Ctenopharyngodon idella).Adult grass carp(608to 1512 g)were fed at 6 graded levels of dietary arecoline(0,0.5,1.0,1.5,2.0,and 2.5 mg/kg diet)for 9weeks.The results suggested that appropriate dietary supplementation of arecoline(1.0 mg/kg)increased growth parameters and intestinal growth in adult grass carp(P<0.05),enhanced digestion and absorption capacities(P<0.05),up-regulated muscarinic receptor 3(M3)mRNA level(P<0.05),increased the content of neuropeptide fish substance P(P<0.05),improved antioxidant capacity by activating the Keap1a/Nrf2signaling pathway(P<0.05),reduced intestinal mucosal permeability(P<0.05),and increased m RNA levels of tight junction(TJ)and adherent junction AJ-related proteins in fish by inhibiting the RhoA/ROCK signaling pathway(RhoA/ROCK/MLCK/NMII)(P<0.05).In addition,the appropriate arecoline supplementation for adult grass carp was determined to be 1.20,1.21,1.07,and 1.19 mg/kg based on percentage weight gain,lipase activity,serum diamine oxidase,and protein carbonyl,respectively.Overall,to the best of our knowledge,we investigated for the first time the effects and possible mechanisms of dietary arecoline on intestinal digestive and absorptive capacities and structural integrity in fish and evaluated the appropriate level of supplementation.Na Yao Lin Feng Weidan Jiang Pei Wu Hongmei Ren Hequn Shi Ling Tang Shuwei Li Caimei Wu Hua Li Yang Liu Xiaoqiu Zhou 2023Animal Nutrition2023,,4:0
