| 1 | Insights into triterpene synthesis and unsaturated fatty-acid accumulation provided by chromosomallevel genome analysis of Akebia trifoliata subsp . australis显示文摘Akebia trifoliata subsp.australis is a well-known medicinal and potential woody oil plant in China.The limited genetic information available for A.trifoliata subsp.australis has hindered its exploitation.Here,a high-quality chromosomelevel genome sequence of A.trifoliata subsp.australis is reported.The de novo genome assembly of 682.14 Mb was generated with a scaffold N50 of 43.11 Mb.The genome includes 25,598 protein-coding genes,and 71.18%(485.55 Mb)of the assembled sequences were identi fi ed as repetitive sequences.An ongoing massive burst of long terminal repeat(LTR)insertions,which occurred~1.0 million years ago,has contributed a large proportion of LTRs in the genome of A.trifoliata subsp.australis.Phylogenetic analysis shows that A.trifoliata subsp.australis is closely related to Aquilegia coerulea and forms a clade with Papaver somniferum and Nelumbo nucifera,which supports the well-established hypothesis of a close relationship between basal eudicot species.The expansion of UDP-glucoronosyl and UDP-glucosyl transferase gene families and fi-amyrin synthase-like genes and the exclusive contraction of terpene synthase gene families may be responsible for the abundant oleanane-type triterpenoids in A.trifoliata subsp.australis.Furthermore,the acyl-ACP desaturase gene family,including 12 stearoyl-acyl-carrier protein desaturase(SAD)genes,has expanded exclusively.A combined transcriptome and fatty-acid analysis of seeds at fi ve developmental stages revealed that homologs of SADs,acyl-lipid desaturase omega fatty acid desaturases(FADs),and oleosins were highly expressed,consistent with the rapid increase in the content of fatty acids,especially unsaturated fatty acids.The genomic sequences of A.trifoliata subsp.australis will be a valuable resource for comparative genomic analyses and molecular breeding. | Hui Huang Juan Liang Qi Tan Linfeng Ou Xiaolin Li Caihong Zhong Huilin Huang Ian Max Moller Xianjin Wu Songquan Song | 2021 | Horticulture Research2021,8,1: | 5 |
| 2 | Fine equiaxed β grains and superior tensile property in Ti-6Al-4V alloy deposited by coaxial electron beam wire feeding additive manufacturing显示文摘Coarse columnar β grains result in anisotropic mechanical properties in Ti alloys deposited by additive manufacturing. This study reports that Ti-6Al-4V alloy fabricated by coaxial electron beam wire feeding additive manufacturing presents a weak anisotropy, high strength and ductility. The superior tensile property arises from a microstructure with fine equiaxed β grains(EGβ), discontinuous grain boundary α phase and short intragranular α lamellae. | 3 | Priming of NLRP3 inflammasome activation by Msn kinase MINK1 in macrophages显示文摘The nucleotide-binding domain,leucine-rich-repeat containing family,pyrin domain-containing 3(NLRP3)inflammasome is essential in inflammation and inflammatory disorders.Phosphorylation at various sites on NLRP3 differentially regulates inflammasome activation.The Ser725 phosphorylation site on NLRP3 is depicted in multiple inflammasome activation scenarios,but the importance and regulation of this site has not been clarified.The present study revealed that the phosphorylation of Ser725 was an essential step for the priming of the NLRP3 inflammasome in macrophages.We also showed that Ser725 was directly phosphorylated by misshapen(Msn)/NIK-related kinase 1(MINK1),depending on the direct interaction between MINK1 and the NLRP3 LRR domain.MINK1 deficiency reduced NLRP3 activation and suppressed inflammatory responses in mouse models of acute sepsis and peritonitis.Reactive oxygen species(ROS)upregulated the kinase activity of MINK1 and subsequently promoted inflammasome priming via NLRP3 Ser725 phosphorylation.Eliminating ROS suppressed NLRP3 activation and reduced sepsis and peritonitis symptoms in a MINK1-dependent manner.Altogether,our study reveals a direct regulation of the NLRP3 inflammasome by Msn family kinase MINK1 and suggests that modulation of MINK1 activity is a potential intervention strategy for inflammasomerelated diseases. | Kaixiang Zhu Xuexiao Jin Zhexu Chi Sheng Chen Songquan Wu Richard D.Sloan Xuai Lin Dante Neculai Di Wang Hu Hu Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,10: | 2 |

| 12 | {332}<113> Twinning transfer behavior and its effect on the twin shape in a beta-type Ti-23.1Nb-2.0Zr-1.0O alloy显示文摘The interaction between twins and grain boundaries(GB) has an important influence on the deformation and fracture behavior of materials. In the present work, {332}<113> twinning transfer(TT) behavior and its effect on the twin shape in a deformed β-type Ti-23.1 Nb-2.0 Zr-1.0 O Ti alloy were investigated experimentally and with molecular dynamics simulation. The crystallographic alignment factor of the two twinning systems in the neighboring grains and the misorientation angle of the grain pairs were found to influence the occurrence of TT. This further determines the twin shape: twins present a ruler shape when TT occurs but are in a lenticular shape otherwise. Such different twin shapes are attributed to the local stress states related to TT occurring or not. | 15 | Deformation twinning in octahe dron-base d face-centere d cubic metallic structures:Localized shear-force dipoles drive atomic displacements显示文摘Twinning is found to impart favorable mechanical,physical and chemical properties to nanostructured materials.Deformation twinning prevails in face-centered cubic(FCC)nanocrystalline materials upon loading.In FCC structures,the<112>{111}deformation twinning is traditionally believed to nucleate and grow through layer-by-layer emission of 1/6<112>Shockley partial dislocations on consecutive{111}planes.We report that deformation twinning is able to occur in crystalline(Fe,Nb)_(23)Zr_(6)nanoparticles(NPs)that have a large Mn 23 Th 6-type FCC structure with a Zr-octahedron as a motif.Based on direct atomic-scale observations,we discover a new zero-net-strain path for the<112>{111}deformation twin-ning in FCC structures.To form a[¯1¯12]/(111)twin,for example,short(¯1¯11)planes within two adjacent(111)plane layers in the repeated three-layer sequence of(111)planes are shear deformed continuously by a shear-force dipole along the[11¯2]direction like a domino effect,whereas the other(111)plane in the repeated sequence remains intact.In addition,a loading criterion for deformation twinning of a FCC NP under uniaxial compression is proposed based on our observations.Our work here not only extends the fundamental understanding on deformation twinning in FCC structures,but also opens up studies of deformation behaviors in a class of Mn 23 Th 6-type FCC materials. | Hengfei Gu Chengze Liu Fusen Yuan Fuzhou Han Yingdong Zhang Muhammad Ali Wenbin Guo Jie Ren Lifeng Zhang Songquan Wu Geping Li | 2022 | Journal of Materials Science & Technology2022,,31: | 0 |
