1A Convenient and Biosafe Replicon with Accessory Genes of SARS-CoV-2 and Its Potential Application in Antiviral Drug Discovery显示文摘SARS-CoV-2 causes the pandemic of COVID-19 and no effective drugs for this disease are available thus far.Due to the high infectivity and pathogenicity of this virus,all studies on the live virus are strictly confined in the biosafety level 3(BSL3)laboratory but this would hinder the basic research and antiviral drug development of SARS-CoV-2 because the BSL3 facility is not commonly available and the work in the containment is costly and laborious.In this study,we constructed a reverse genetics system of SARS-CoV-2 by assembling the viral cDNA in a bacterial artificial chromosome(BAC)vector with deletion of the spike(S)gene.Transfection of the cDNA into cells results in the production of an RNA replicon that keeps the capability of genome or subgenome replication but is deficient in virion assembly and infection due to the absence of S protein.Therefore,such a replicon system is not infectious and can be used in ordinary biological laboratories.We confirmed the efficient replication of the replicon by demonstrating the expression of the subgenomic RNAs which have similar profiles to the wild-type virus.By mutational analysis of nsp12 and nsp14,we showed that the RNA polymerase,exonuclease,and cap N7 methyltransferase play essential roles in genome replication and sgRNA production.We also created a SARS-CoV-2 replicon carrying a luciferase reporter gene and this system was validated by the inhibition assays with known anti-SARS-CoV-2 inhibitors.Thus,such a one-plasmid system is biosafe and convenient to use,which will benefit both fundamental research and development of antiviral drugs.Yun-Yun Jin Hanwen Lin Liu Cao Wei-Chen Wu Yanxi Ji Liubing Du Yiling Jiang Yanchun Xie Kuijie Tong Fan Xing Fuxiang Zheng Mang Shi Ji-An Pan Xiaoxue Peng Deyin Guo 2021Virologica Sinica2021,36,5:2

3An improved self-calibration approach based on adaptive genetic algorithm for position-based visual servo显示文摘An improved self-calibrating algorithm for visual servo based on adaptive genetic algorithm is proposed in this paper. Our approach introduces an extension of Mendonca-Cipolla and G. Chesi's self-calibration for the position-based visual servo technique which exploits the singular value property of the essential matrix. Specifically,a suitable dynamic online cost function is generated according to the property of the three singular values. The visual servo process is carried out simultaneous to the dynamic self-calibration,and then the cost function is minimized using the adaptive genetic algorithm instead of the gradient descent method in G. Chesi's approach. Moreover,this method overcomes the limitation that the initial parameters must be selected close to the true value,which is not constant in many cases. It is not necessary to know exactly the camera intrinsic parameters when using our approach,instead,coarse coding bounds of the five parameters are enough for the algorithm,which can be done once and for all off-line. Besides,this algorithm does not require knowledge of the 3D model of the object. 7New insights into multiple provenances evolution of the Jurassic from heavy minerals characteristics in southern Junggar Basin, NW China显示文摘The method of random forest was used to classify the heavy mineral assemblages of 2 418 Jurassic samples in the southern Junggar Basin, and determine the distribution of the heavy mineral assemblages from the same provenance systems. Based on the analysis of heavy minerals assemblages, ZTR index, sedimentary characteristics, U-Pb zircon ages, whole-rock geochemical and paleocurrent direction analysis, the study reveals that five important provenances were providing sediments to the southern Junggar Basin in the Jurassic period: The North Tianshan(NTS), Central Tianshan(CTS), Bogda Mountains, Zhayier Mountains and Kalamaili Mountains. During the Early Jurassic, NTS-CTS, Kalamaili Mountains and Zhayier Mountains are primary provenances, Bogda Mountains started to uplift and supply clastic materials in the Middle Jurassic. There are three sedimentary area in the Jurassic of southern Junggar Basin: the western part, the central part and the eastern part. In the western part, the clastic materials of the Early Jurassic was mainly from NTS blocks and Zhayier Mountains, and the sediments were dominantly derived from the Zhayier Mountains during the Middle–Late Jurassic. In the central part, the main provenance of the Early Jurassic switched from NTS to CTS. In the Xishanyao Formation, the main source went back to NTS again. The NTS was the primary provenance during the sedimentary periods of Toutunhe Formation and Qigu Formation. In the eastern part, the contribution of CTS and Kalamaili Mountains were considered as major provenances in the Early Jurassic-Xishanyao Formation, small proportion of sediments were from NTS. The Bogda mountains uplifted and started to provide sediments to the Junggar Basin in the sedimentary period of Xishanyao Formation, and became the major source during the Toutunhe Formation period, with small amount of sediments from CTS. 9TinII intron, an enhancer to affect the function of the cytoplasmic male sterility related gene T in Brassica juncea显示文摘The T gene,which was cloned from the mitochondria of tumorous stem mustard(Brassica juncea var.tumida),is a cytoplasmic male sterility(CMS)-related gene that can produce two transcripts,T1170 and T1243.The latter is transcribed with the uncleaved intron TinII.In our previous study,transgenic Arabidopsis thaliana plants over-expressing the T1243 transcript(OE-T1243)showed a severe male-sterile phenotype,whereas OE-T1170 plants did not.However,the functional mechanism of the T gene in B.Juncea remained unknown.In this study,microscopic analyses of paraffin sections of anthers confirmed that OE-T1243 plants did not produce normal pollen,whereas OE-T1170 plants did.We analyzed the transcription of 15 anther development-related genes and found that transcript levels of nozzle/sporocyteless and barely any meristem 1 and 2 were markedly lower in OE-T1243 plants than those in wild type,while the transcript levels of these genes in OE-T1170 plants were unchanged.To investigate the potential roles of TinII,we inserted the TinII sequence upstream of a minimal region(60)of the cauliflower mosaic virus 35S promoter fused to the 5′end of theβ-glucuronidase(GUS)reporter gene.Analysis of the transgenic plants suggested that TinII acted as an enhancer to significantly increase GUS expression.The potential action mechanism is that the TinII intron acts as an enhancer to affect the function of the CMS-related gene T.JIN ZhuPing WU LingLing CAO JiaShu CHEN ZhuJun PEI YanXi 2013Science China(Life Sciences)2013,56,12:0
10The competitive and synergistic effect between adsorption enthalpy and capacity in D_(2)/H_(2)separation of M_(2)(m-dobdc)frameworks显示文摘Hydrogen isotope separation is a challenging task due to their similar properties.Herein,based on the chemical affinity quantum sieve(CAQS)effect,the D_(2)/H_(2)separation performance of M_(2)(m-dobdc)(M=Co,Ni,Mg,Mn;m-dobdc^(4-)=4,6-dioxido-1,3-benzenedicarboxylate),a series of honeycomb-shaped MOFs with high stability and abundant open metal sites,are studied by gases sorption and breakthrough experiments,in which two critical factors,gas uptake and adsorption enthalpy,are taken into consideration.Among these MOFs,Co_(2)(m-dobdc)exhibits the longest D_(2)retention time of 180 min/g(H_(2)/D_(2)/Ne:1/1/98)at 77 K because of its second-highest adsorption enthalpy(10.7 kJ/mol for H_(2)and 11.8 kJ/mol for D_(2))and the best sorption capacity(5.22 mmol/g for H_(2)and 5.49 mmol/g for D_(2))under low pressure of 1 kPa and 77 K,which make it a promising material for industrial hydrogen isotope separation.Moreover,the results indicate that H_(2)and D_(2)capacities under low pressure(about 1 kPa)dominate the final D_(2)/H_(2)separation property of MOFs.Fan Wu Liqiong Li Yanxi Tan El-Sayed M.El-Sayed Daqiang Yuan 2021Chinese Chemical Letters2021,32,11:0
