1Expression of Beclin1 in non small cell lung cancer and its clinical significance显示文摘Objective:To investigate the expression of autophagy related gene;Beclin1 in human non-small cell lung cancer(NSCLC)tissues.Methods:Protein expression of Beclin1 was determined by immunofluorescence staining and Western Blot,mRNA expression was analyzed by RT-PCR.Results:Immunofluorescence staining revealed that the level of Beclin1 expression in lung cancer was significantly lower than that in adjacent noncancerous tissues and normal tissues(expression rate 8.3%,P=0.000).The Beclin1 mRNA expression in lung cancer,adjacent noncancerous tissues and normal tissues was 1.372(±0.475)1.721(±0.521)and 1.553(±0.554)when F = 15.0,P < 0.01.Beclin1 protein expression in lung cancer,adjacent noncancerous tissues and normal tissues was 3.453(±0.852)5.423(±1.351)and 6.878(±0.997)F = 11.2,P < 0.01.Conclusion:The protein and mRNA expression of Beclin1 in lung cancer was much lower than those in and around cancer tissues and normal tissues,those differences having statistical significance.However in adjacent noncancerous tissues and normal tissues,the expression showed no difference.Quan Liu Jianjun Wang Yongcheng Pan Lifeng Meng Xi Zhan 2008Journal of Nanjing Medical University2008,22,2:6
2Cupric ion release and cytotoxicity for Yuangong Cu-IUDs and the release behavior of indomethacin for medicated 220 Cu-IUD显示文摘These years Yuangong copper-bearing intrauterine devices (Cu-IUDs) have been used because of less side effects in use. The corrosion of copper is essential to the success of contraception, and the release behavior of indomethacin from medicated Cu-IUD is related to its therapeutic effect. In this study, analytical methods were established to investigate the release behavior of cupric ion of three kinds of Yuangong Cu-IUDs and indomethacin of medicated Yuangong 220 Cu-IUD. Cu-IUDs were incubated in simulated uterine solution (SUS). The concentrations of cupric ion and indomethacin were analyzed by flame atomic absorption spectrometer (FAAS) for 60 days and UV/vis ?3310 spectrophotometer for 60 days, respectively. The morphology of copper after corrosion was characterized by SEM. In addition, we detected cytotoxicity by MTT of L929 mouse fibroblasts cells caused by extracts of the three Yuangong Cu-IUDs. The release behavior of cupric ion for three kinds of Yuangong Cu-IUDs was biphasic, which consisted of the initial burst release and then slow and constant release. In vitro release experiment confirmed a biphasic release of indomethacin from Yuangong 220. The copper wire of Yuangong Cu-IUDs showed uneven corrosion. The RGR value of Yuangong 365 Cu-IUD was smaller than that of medicated Yuangong 220 Cu-IUD and RGR value of medicated Yuangong 220 Cu-IUD was smaller than that of Yuangong 300 Cu-IUD. The cupric ion release and indomethacin release showed biphasic. Indomethacin increased the cupric ion release rate and might diminish the adverse effects caused by burst release of cupric ion. The toxicity grade of these three Yuangong Cu-IUDs was 4. 3Toxicity and clinical reasonable application of Taoren(Semen Persicae) based on ancient and modern literature research显示文摘OBJECTIVE:To probe the toxicity and relative application theory of the commonly-used traditional Chinese herbal drug Taoren(Semen Persicae),and set up a correct attitude and principle and method to use Taoren(Semen Persicae) for treating the syndrome of stagnation of blood stasis and others in TCM clinic.METHODS:In this study,we analyzed and probed the ancient and modern literature research about Taoren(Semen Persicae),and summarized the realization of its toxicity and application contraindications in ancient herbals and the research assertion of its processing,drug-nature,pharmacologic actions,adverse reaction,and clinical reasonable application in modern literature.RESULTS:We found that some TCM doctors were worried about the effect of Taoren(Semen Persicae) 's disintegrating blood stasis to impair body's healthy Qi and its toxicity,and were not good at using this herb.And some patients were afraid of its toxic and side-effect not to take it.In the ancient and modern literatures some proper hates of Taoren(Semen Persicae) existed,and the toxicity component was also clear-cut,and the applications of Taoren(Semen Persicae) were in many fields especially the gynecological and traumatological diseases.The key root of toxicity generation and unreasonable application of Taoren(Semen Persicae) lies in taking without syndrome differentiation or using with overdosage.CONCLUSION:Under the precondition of correct processing,treatment based on syndrome differentiation,and taking the dosage stipulated by laws to apply Taoren(Semen Persicae) should be quite safe.The ancient and modern literature records and researches about Taoren(Semen Persicae) provide the determinate reference for understanding Taoren(Semen Persicae)'s efficacy and drug-nature(toxicity) more objectively and also for further correctly clinic recognition and research on Taoren(Semen Persicae).Shengyan Xi Linchao Qian Haiying Tong Lifeng Yue Hui Zhao Dawei Wang Dawei Lu Pengcheng Li Xinrong Wang 2013Journal of Traditional Chinese Medicine2013,33,2:5
