2Biological evaluations of decellularized extracellular matrix collagen microparticles prepared based on plant enzymes and aqueous two-phase method显示文摘For patients with extensive full-thickness burns who do not have sufficient autologous splitthickness skin for skin grafts,the application of biological skin substitutes may be considered.The aim of this study was to find an optimal new type method for the production of a biovital skin substitute based on acellular dermal matrix(ADM)and preclinical evaluations.In this work,25 methods of ADM production were assessed.The proposed methods are based on the use of the following enzymes:papain,Carica papaya lipase(CPL),and purification using a polymer/salt aqueous two-phase system.The obtained ADM samples were characterized via scanning electron microscopy(SEM),porosity measurement and water vapor transmission test.Results showed that the collagen bundles of ADM microparticles were intact and orderly.Through differential scanning calorimetry(DSC),thermo gravimetric analysis(TGA)and biocompatibility tests,the results indicated that the proportion of papain and CPL was the same and 5 h processing time are the optimum conditions for ADM preparation and the material showed good biocompatibility.Our results suggested that the potential of developing this kind of decellularization process to manufacture ADM scaffolds for clinical application.YaWen Liu Ching-Cheng Huang YuanYuan Wang Jun Xu GuoDing Wang XinPeng Bai 2021Regenerative Biomaterials2021,8,2:1
3Sensitive direct-conversion X-ray detectors formed by ZnO nanowire field emitters andβ-Ga_(2)O_(3)photoconductor targets with an electron bombardment induced photoconductivity mechanism显示文摘Sensitive X-ray detection is needed in diverse areas motivated by a common desire to reduce radiation dose.Cold cathode X-ray detectors operating with a photoelectron multiplication mechanism called electron bombardment induced photoconductivity(EBIPC)have emerged as promising candidates for low-dose X-ray detection.Herein,the cold cathode detectors formed by ZnO nanowire field emitters andβ-Ga_(2)O_(3)photoconductor targets were proposed for sensitive direct-conversion X-ray detection.The charge carrier transport mechanism of EBIPC effect in X-ray detectors was investigated to achieve a high internal gain(2.9×10^(2))and high detection sensitivity(3.0×10^(3)μCGy^(−1)_(air)cm^(−2))for a 6 keV X-ray at the electric field of 22.5 Vμm−1.Furthermore,the proposed X-ray detectors showed the features of fast response time(40 ms),long-term stability(0.6%for 1 h),and low detection limit(0.28 mGy_(air)s^(−1)),suggesting that the direct-conversion cold cathode X-ray detectors are ideal candidates for low-energy X-ray detecting and imaging applications.ZHIPENG ZHANG MANNI CHEN XINPENG BAI KAI WANG HUANJUN CHEN SHAOZHI DENG JUN CHEN 2021Photonics Research2021,9,12:1
4Extraction and characterization of bovine collagen Type V and its effects on cell behaviors显示文摘Collagen Type V(Col.V)plays an essential role in cell behaviors and has attracted increasing attention in recent years.Highpurity Col.V is needed for evaluating its biological properties.In this research,the enzymatic hydrolysis process was combined with ultrafiltration to purify Col.V from the bovine cornea.The purity of Col.V was determined to be above 90%by both sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)and high-performance liquid chromatography methods.The effect of Col.V on cell behaviors was evaluated.The circular dichroism spectroscopy results demonstrated that the extracted Col.V exhibited a complete triple helix structure.SDS-PAGE suggested that the molecular weight of Col.V was 440 kDa.The self-assembly experiment revealed that the proportion of Col.V in the collagen mixture can affect the Col.I fiber diameter.The cell culture results implied that Col.V can inhibit fibroblasts(L929)proliferation.The L929 showed maximum mobility when the addition of Col.V was 30%.Thus,Col.V has the effect of inhibiting L929 proliferation and promoting migration.The high-purity Col.V provides useful information for further understanding its biological implications.Jun Xu Xi Luo Yang Zhang Jianping Gao Ching-Cheng Huang Xinpeng Bai Guifeng Zhang 2022Regenerative Biomaterials2022,9,1:0
5New insights into the dispersion history and adaptive evolution of taxon Aegilops tauschii in China显示文摘Aegilops tauschii,the wild progenitor of wheat D-genome and a valuable germplasm for wheat improvement,has a wide natural distribution from eastern Turkey to China.However,the phylogenetic relationship and dispersion history of Ae.tauschii in China has not been scientifically clarified.In this study,we genotyped 208 accessions(with 104 in China)using dd RAD sequencing and 55K SNP array,and classified the population into six sublineages.Three possible spreading routes or events were identified,resulting in specific distribution patterns,with four sublineages found in Xinjiang,one in Qinghai,two in Shaanxi and one in Henan.We also established the correlation of SNP-based,karyotypebased and spike-morphology-based techniques to demonstrate the internal classification of Ae.tauschii,and developed consensus dataset with 1245 putative accessions by merging data previously published.Our analysis suggested that eight inter-lineage accessions could be assigned to the putative Lineage 3and these accessions would help to conserve the genetic diversity of the species.By developing the consensus phylogenetic relationships of Ae.tauschii,our work validated the hypothesis on the dispersal history of Ae.tauschii in China,and contributed to the efficient and comprehensive germplasm-mining of the species.Hao Li Fang Nie Lele Zhu Menghua Mu Ruixiao Fan Jingyao Li Aaqib Shaheen Yifan Liu Can Li Wenjuan Liu Huihui Liang Xinpeng Zhao Shenglong Bai Guanghui Guo Zheng Li Yiheng Hu Yuannian Jiao Jonathan Adams Assaf Distelfeld Guiling Sun Suoping Li Yun Zhou Chun-Peng Song 2022Journal of Genetics and Genomics2022,49,3:0
6STRATEGIES FOR A LOW-CARBON FOOD SYSTEM IN CHINA显示文摘In China,there has been insufficient study of whole food system greenhouse gas(GHG)accounting,which limits the development of mitigation strategies and may preclude the achievement of carbon peak and carbon neutrality goals.The paper presents the development of a carbon extension of NUFER(NUtrient flows in Food chain,Environment and Resources use model),a food system GHG emission accounting model that covers land use and land-use change,agricultural production,and post-production subsectors.The spatiotemporal characteristics of GHG emissions were investigated for the Chinese food system(CFS)from 1992 to 2017,with a focus on GHG emissions from the entire system.The potential to achieve a low-carbon food system in China was explored.The net GHG emissions from the CFS increased from 785Tg CO_(2)equivalent(CO_(2)-eq)in 1992 to 1080 Tg CO_(2)-eq in 2017.Agricultural activities accounted for more than half of the total emissions during the study period,while agricultural energy was the largest contributor to the GHG increase.In 2017,highest emitting regions were located in central and southern China(Guangdong and Hunan),the North China Plain(Shandong,Henan and Jiangsu)and Northeast China(Heilongjiang and Inner Mongolia)and contributed to over half of the total GHG emissions.Meanwhile,Xinjiang,Qinghai and Tibet are shown as carbon sink areas.It was found that foodsystem GHG emissions could be reduced to 355 Tg CO_(2)-eq,where enhancing endpoint mitigation technologies,transforming social-economic and diet conditions,and increasing agricultural productivities can contribute to 60%,25%and 15%,respectively.Synergistic mitigation effects were found to exist in agricultural activities.Xinpeng JIN Xiangwen FAN Yuanchao HU Zhaohai BAI Lin MA 2023Frontiers of Agricultural Science and Engineering2023,10,2:0
7Introgression of QTL from Aegilops tauschii enhances yield-related traits in common wheat显示文摘To break the narrow diversity bottleneck of the wheat D genome, a set of Aegilops tauschii-wheat introgression(A-WI) lines was developed by crossing Ae. tauschii accession T015 with common wheat elite cultivar Zhoumai 18(Zhou18). A high-density genetic map was constructed based on Single Nucleotide Polymorphism(SNP) markers and 15 yield-related traits were evaluated in 11 environments for detecting quantitative trait loci(QTL). A total of 27 environmentally stable QTL were identified in at least five environments, 20 of which were derived from Ae. tauschii T015, explaining up to 24.27% of the phenotypic variations. The major QTL for kernel length(KL), QKl-2D.5, was delimited to a physical interval of approximately 2.6 Mb harboring 52 candidate genes. Three Kompetitive Allele Specific PCR(KASP)markers were successfully developed based on nonsynonymous nucleotide mutations of candidate gene AetT093_2Dv1G100900.1 and showed that A-WI lines with the T015 haplotype had significantly longer KL than the Zhou18 haplotype across all 11 environments. Four primary valuable A-WIs with good trait performance and carrying yield-related QTL were selected for breeding improvement.
