1Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized,whether and how liver-intrinsic factors drive metastatic spread is incompletely understood.Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver,which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models,respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling,which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors.In summary,this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance.Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention.Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng 2021Cellular & Molecular Immunology2021,18,4:5
2Flexible microbubble-based Fabry-Pérot cavity for sensitive ultrasound detection and wide-view photoacoustic imaging显示文摘Interaction of acoustic waves and microbubbles occurs in numerous biomedical applications including ultrasound imaging,drug delivery,lithotripsy treatment,and cell manipulation,wherein the acoustically driven microbub-blcs routincly act as active microscale oscillators or actuators.In contrast,microbubbles were utilized here as passive reccivers to detect broadband ultrasound waves in aqueous environments.The microbubble was photo-thermally generated on a microstructured optical fiber(MOF)tip,forming a flexible Fabry-Perot cavity whose gas-water interface was sensitive to ultrasound waves.The MOF severed as both a low-loss waveguide and a compact light condenser,allowing high-efficiency generation and stabilization of ultrasmall microbubbles.Integrated with all-fiber interferometry,a 10 um diameter microbubble exhibited a low noise-cquivalcnt pressur elevei of^3.4 mPa/Hz^1/2 and a broad bandwidth of^0.8 MHz,capable of detecting weak ultrasounds emittedfrom red blood cells irradiated by pulsed laser light.With advantages of high sensitivity,compact size,and low cost,the microbubble-based ultrasound sensor has great potential in biomedical imaging and sensing applications.JUN MA YANG HE XUE BAI LI-PENG SUN KA CHEN KYUNGHWAN OH BAI-OU GUAN 2020Photonics Research2020,8,10:4
3Trait complementarity between fine roots of Stipa purpurea and their associated arbuscular mycorrhizal fungi along a precipitation gradient in Tibetan alpine steppe显示文摘Development of fine roots and formation of symbiosis with arbuscular mycorrhizal(AM) fungi represent two strategies for plants to acquire nutrient and water from soil. Here, we elucidated how fine root development and symbolized mycorrhizal fungi with Stipa purpurea responded to the precipitation change in Tibetan alpine steppe ecosystem across a precipitation gradient from 50 mm to 400 mm. As precipitation increased, the proportion of thinner fine roots(diameter < 0.4 mm) in total roots increased significantly; while the mycorrhizal colonization percentage, either associated with thinner or thicker roots, decreased. This phenomenon indicated that fine root development and symbolized mycorrhizal fungi are likely alternative, and plant preferred to develop fine root rather than build a symbiotic relationship with mycorrhizal fungi in more benign niches with higher precipitation. Also, root diameter was negatively correlated with specific root length(SRL), but positively correlated with AM fungal colonization percentage, indicating thicker-root species rely more on mycorrhizal fungi in alpine steppe.
