1Analysis of Phthalate Esters in Air, Soil and Plants in Plastic Film Greenhouse显示文摘The phthalate esters such as DMP, DEP, DBP and DEHP in air, soil and plant samples in plastic film greenhouse were clean up with fine silica gel column and determined with HPLC. It was found that the concentrations of PEs in air and soil samples in plastic film greenhouse are much higher than those of contrast samples. 2Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
3Mesoporous Ag nanocubes synthesized via selectively oxidative etching at room temperature for surface- enhanced Raman spectroscopy显示文摘Silver nanocubes enriched with {100} facets have been extensively used for surface-enhanced Raman scattering. Herein, we report a new water-phase synthesis method for weU-defined Ag nanocubes with tunable sizes via a two-step procedure at room temperature. First, irregularly shaped Ag nanoparticles (INPs) were prepared by reducing silver ammonia solution using ethylal. Second, the agglomerated INPs were selectively etched with HNO3 and C1- to yield {100} facet-rich mesoporous Ag nanocubes. The mechanism of Ag-nanocube formation and growth was investigated in detail by elucidating the involved chemical reactions and physical changes at each step during the synthesis. The addition of C1- anions was responsible for facilitating Ag nanoparticle growth by removing surface-adsorbed Ag+ species, thereby eliminating inter-particle repulsive forces. This agglomeration was found crucial for the subsequent selective oxidation of Ag nanoparticles because the protective agent used, polyvinylpyrrolidone (PVP), was the most effective one for adsorption on the surfaces of Ag nanoparticles of size greater than approximately 50 nm. Importantly mesopores were found inside the Ag nanocubes; this can be attributed to the unavoidable imperfect packing during the agglomeration of INPs. 4Molecular cloning and expression analysis of FTZ-F1 in the GIFT tilapia,Oreochromis niloticus显示文摘The FTZ-F1 genes encode orphan receptors of the nuclear receptor superfamily and in mammals have been found to play important roles in the proper development of the adrenal-gonadal axis and sex-determination.We isolated the homologue of FTZ-F1 in genetically improved farmed tilapia(gfFTZ-F1).The full-length cDNA was isolated from the ovary,which included an open reading frame encoding a predicted protein of 486 amino acids.Sequence,tissue distribution and phylogenic analysis of the FTZ-F1 showed that the gfFTZ-F1 belonged to SF-1/Ad4BP group and that gfFTZ-F1 transcripts were only expressed in the gonads and kidney but not in other tissues.Likewise our data suggests that the gfFTZ-F1 gene may share similar functions with other fish and mammalian counterparts,though further study is needed to make any definitive conclusions.Jinling CAO Jianjie CHEN Zhongliang JIANG Yongju LUO Xi GAN 2012Zoological Research2012,33,4:2
5Pretreatment of Shaoyao Gancao Decoction(芍药甘草汤)Alters Pharmacokinetics of Intravenous Paclitaxel in Rats显示文摘Objective:To investigate the effect of Shaoyao Gancao Decoction(芍药甘草汤,SGD) on the pharmacokinetics of intravenously administered paclitaxel in rats.Methods:Paclitaxel was intravenously administered to rats(3 mg/kg) with or without the concomitant administration of SGD(752 mg/kg,a single day or 14 consecutive days pretreatment).The paclitaxel in the serum was quantified using a simple and rapid ultra performance liquid chromatography(UPLC) method for the pharmacokinetic study.The pharmacokinetic parameters were calculated via a non-compartment model using the computer program DAS 2.0.Results:The pharmacokinetic parameters of paclitaxel were significantly altered in response to 14 consecutive days of pretreatment with SGD.The area under the curve(AUC_(0-t),from 4 820 ± 197 to 4 205 ± 186 ng·mL^(-1)·h^(-1))and AUC_(0-∞)(from 5 237 ±280 to 4 514 ± 210 ng·mL^(-1)·h^(-1)) significantly decreased in response to the 14-day pretreatment with SGD.The values of V_(dss)(L/kg) were 10.74 ±1.08 and 9.35 ±0.49,those of CL(L/kg) were0.67 ±0.03 and 0.57 ±0.03 and the t_(1/2)(h) values were 11.17 ±0.84 and 11.32 ±0.93,respectively,for the14-day SGD pretreatment and intravenous paclitaxel alone.The AUC_(0-t) and AUC_(0-∞) values decreased by13%and 14%(P<0.01),respectively.The area under the curve decreased significantly(P<0.01),and the total clearance increased by 1.2-fold(P<0.01),after 14 consecutive days of pretreatment with SGD.A single-day pretreatment with SGD did not significantly affect the pharmacokinetic parameters of paclitaxel.Conclusions:SGD administration for 14 consecutive days increased the metabolism of paclitaxel,while a 1-day pretreatment had little effect.The results would contribute important information to the study on interaction between Chinese medicines and chemotherapy and also help to utilize SGD better in the adjunctive therapy of cancer patients.WANG Yang HUANG Xi ZHONG Mei-zuo LU Ruo-huang XIA Zi-an FAN Rong LIU Bo HUANG Wei GAN Ping-ping 2017Chinese Journal of Integrative Medicine2017,23,1:2

8Concentration Characteristics and Sources of Chemical Elements in Atmospheric Fine Particles (PM_(2.5)) in Autumn in Xi'an City显示文摘[Objective] The aim was to study the concentration characteristics and sources of chemical elements in atmospheric fine particles (PM2.5) in autumn in Xi'an City. [Method] By means of mini-volume sampler, PM2.5 samples in atmosphere in Xi'an were collected in October 2009, and the concentration characteristics and sources of elements in PM2.5 were analyzed. [Result] The average mass concentration of PM2.5 in atmosphere in autumn in Xi'an City was 168.44 μg/m3 which was higher than that of Beijing and Pearl River Delta area, and the minimum and maximum value were 53.29 and 358.16 μg/m3, respectively. The mass concentration of S, Zn, K, Cl, Ca and Fe in PM2.5 was above 1.0 μg/m3, being at high pollution level. In addition, K had obvious correlation with organic carbon (OC) and element carbon (EC), with the correlation coefficients of 0.76 and 0.75 (P<0.000 1), respectively, and it showed that OC and EC had the same source as K, namely biomass burning had certain contribution to OC and EC. Enrichment factors analysis revealed that K, Ca, Fe, Ti, Mn and Cr mainly came from earth crust, rock weathering and other natural sources, while anthropogenic pollution sources had great effects on S, Zn, Cl, Pb, Br, Mo, Cd and As which were affected by soil dust and other natural sources slightly, and Cd had the highest enrichment factor and mainly came from metal smelting. In a word, coal combustion, biomass burning, vehicle emissions, metallurgical, chemical industry and dust were the main sources of PM2.5 in autumn in Xi'an.
