1Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11
2Computer Aided Simulation on Precision Cold Forging of Universal Joint Cross显示文摘The 3D FEM numerical simulation on multi-action precision cold forging technology of universal joint cross and differential spider is done in this article using DEFORM Software, a commercial computer aided engineering software specializing in forming and heat treatment simulation technology, and suitable for cold, warm and hot forging process. The material flow properties, the dynamic variation of stress and strain in the process of deformation and the load-stroke curve have also been achieved. A good consistency is exhibited between simulation results and practical data. Based on the DEFORM simulation results, the optimized procedure has been found for forging a universal joint cross. What should be emphasized here is that a better understanding of practical forging characters and the environmental factors can greatly improve the simulation accuracy thus make the simulation results more reliable.H. 3Nighttime sap flow of Acacia mangium and its implications for nighttime transpiration and stem water storage显示文摘Aims Nighttime sap flow of trees may indicate transpiration and/or recharge of stem water storage at night.This paper deals with the water use of Acacia mangium at night in the hilly lands of subtropical South China.Our primary goal was to reveal and understand the nature of nighttime sap flow and its functional significance.Methods Granier’s thermal dissipation method was used to determine the nighttime sap flux of A.mangium.Gas exchange system was used to estimate nighttime leaf transpiration and stomatal conductance of studied trees.Important Findings Nighttimesap flowwas substantial and showed seasonal variation similar to the patterns of daytime sap flowin A.mangium.Mean nighttime sap flow was higher in the less precipitation year of 2004(1122.4 mm)than in the more precipitation year of 2005(1342.5 mm)since more daytime transpiration and low soil water availability in the relatively dry 2004 can be the cause of more nighttime sap flow.Although vapor pressure deficit and air temperature were significantly correlated with nighttime sap flow,they could only explain a small fraction of the variance in nighttime sap flow.The total accumulated water loss(E_(L))by transpiration of canopy leaves was only;2.6–8.5%of the total nighttime sap flow(E_(t))during the nights of July 17–18 and 18–19,2006.Therefore,it is likely that the nighttime sap flow was mainly used for refillingwater in the trunk.The stem diameter at breast height,basal area and sapwood area explained much more variance of nighttime water recharge than environmental factors and other tree form features,such as tree height,stem length below the branch,and canopy size.The contribution of nighttime water recharge to the total transpiration ranged from 14.7 to 30.3%depending on different DBH class and was considerably higher in the dry season compared to the wet season.Hua Wang Ping Zhao Dirk Hölscher Quan Wang Ping Lu Xi A.Cai Xiao P.Zeng 2012Journal of Plant Ecology2012,5,3:5
