| 1 | One Stone Four Birds:A Novel Liposomal Delivery System Multi-functionalized with Ginsenoside Rh2 for Tumor Targeting Therapy显示文摘Liposomes hold great potential in anti-cancer drug delivery and the targeting treatment of tumors.However,the clinical therapeutic efficacy of liposomes is still limited by the complexity of tumor microenvironment(TME)and the insufficient accumulation in tumor sites.Meanwhile,the application of cholesterol and polyethylene glycol(PEG),which are usually used to prolong the blood circulation and stabilize the structure of liposomes respectively,has been questioned due to various disadvantages.Herein,we developed a ginsenoside Rh2-based multifunctional liposome system(Rh2-lipo)to effectively address these challenges once for all.Different with the conventional’wooden’liposomes,Rh2-lipo is a much more brilliant carrier with multiple functions.In Rh2-lipo,both cholesterol and PEG were substituted by Rh2,which works as membrane stabilizer,long-circulating stealther,active targeting ligand,and chemotherapy adjuvant at the same time.Firstly,Rh2 could keep the stability of liposomes and avoid the shortcomings caused by cholesterol.Secondly,Rh2-lipo showed a specifically prolonged circulation behavior in the blood.Thirdly,the accumulation of the liposomes in the tumor was significantly enhanced by the interaction of glucose transporter of tumor cells with Rh2.Fourth,Rh2-lipo could remodel the structure and reverse the immunosuppressive environment in TME.When tested in a 4T1 breast carcinoma xenograft model,the paclitaxel-loaded Rh2-lipo realized high efficient tumor growth suppression.Therefore,Rh2-lipo not only innovatively challenges the position of cholesterol as a liposome component,but also provides another innovative potential system with multiple functions for anti-cancer drug delivery. | Chao Hong Jianming Liang Jiaxuan Xia Ying Zhu Yizhen Guo Anni Wang Chunyi Lu Hongwei Ren Chen Chen Shiyi Li Dan Wang Huaxing Zhan Jianxin Wang | 2020 | Nano-Micro Letters2020,12,10: | 8 |
| 2 | Numerical Study of the Effect of Imaginary Circle Diameter and Initial Flow Field on the Aerodynamic Field in a Tangentially Fired Furnace显示文摘In this paper, the effect of the imaginary circle diameter opi and the initial flow field on the aerodynamic field in a tangentially fired furnace was studied by numerical simulation and experiments in the cold model. Results show that merely reducing the imaginary circle diameter rki can not significantly reduce the rotatiollal diameter op in the range considered. The flow still rotates coullter-clockwise stably and does not change rotation direction when the direction of all jet axes are defiected suddenly to the opposite rotation direction by up to 5.4' in a countereclockwise llow field. It is the first time that the numerical sAnulation results were obtained which agreed quite well with this experimental phenomena qualitatively. The experimental data, i.e., the rotational diameter & and the maximum velocity on the syrnmetric central line of fUrnace Vm, are only a bit larger than the simulation results. It is shown that the initial flow field has an important influence on the aerodynamic field in the furnace. Other measures have to be taken as wel1 in order to reduce & to resist slagging and high temperature corrosion of furnace tubes. | 3 | Assessment of Urban Climate Environment and Configuration of Ventilation Corridor:A Refined Study in Xi'an显示文摘Integrating urban spatial landscape(USL) parameters into refined climate environment assessment is important. By taking the central urban area(CUA) of Xi’an, China as an example, this study develops an evaluation method based on Urban Climatic Map(UCMap) technology. We define surface urban heat island intensity(SUHI) and surface ventilation potential coefficient(VPC), which can effectively reflect local urban climate. Based on SUHI and VPC,we analyze the influences of seven typical USL metrics including building height(BH), building density(BD), floor area ratio(FAR), sky view factor(SVF), frontal area index(FAI), surface roughness length(RL), and vegetation cover(VC). Then, we construct a comprehensive evaluation model and create an urban climate zoning map on a 100-m resolution. The climate optimization on the map is performed for configuration of possible ventilation corridors and identification of associated control indicators. The results show that the main factors affecting SUHI in the CUA of Xi’an are VC and BD, which explain 87.9% of the variation in SUHI, while VPC explains 50% of the variation in SUHI. The main factors affecting VPC are BH, FAR, FAI, and RL, all of which contribute to more than 95% of the variation in VPC. The evaluation model constructed by SUHI, VPC, and VC can divide the CUA into climate resource spaces, climate preservation spaces, climate sensitive spaces, and climate restoration spaces. On this basis, a ventilation corridor network of 3 level-1 corridors(each over 500 m wide), 6 level-2 corridors(each over 500 m wide) and 13 level-3 corridors(each over 50 m wide) is established. | 4 | MAPK/ERK regulation of P53 in human epidermoid carcinoma cell line A431显示文摘Objective:To observe the impact of activation and inhibition of mitogen activated protein kinases(MAPK)/extracellular signalregulated protein kinase(ERK)signaling pathway on the proliferation and apoptosis of cutaneous squamous cell carcinoma(SCC).cells and investigate the interaction mechanism between MAPK/ERK signaling pathway and tumor suppressor gene P53 in SCC.Methods:Human A431 cells were cultured and divided into MAPK/ERK inhibition groups with low-,medium-and highconcentration of inhibitors(PD98059+DMSO),MAPK/ERK activation groups with low-,medium-and high-concentration of stimuli(IGF+PBS)and blank control group(DMSO).The cell proliferation in vitro was detected by MTT assay,with the cell apoptosis detected by flow cytometry(FCM)and the protein expression of P-ERK and P53 detected by western blot in each group.Results:The A431 cell proliferation was inhibited by different concentrations of PD98059 with a clear concentration-effect and time-effect relationship(p<.05);and the cell proliferation was promoted by the different concentrations of IGF with a clear concentration-effect and time-effect relationship(p<.05).The FCM results showed a significant increase in the apoptosis rate of A431 cells which were treated with PD98059,with a clear concentration-effect relationship(p<.05);while the apoptosis rate was decreased significantly after A431 cells were treated with IGF,also with a concentration-effect relationship(p<.05).The western blot results showed that the expression of P-ERK protein was decreased but the expression of P53 was increased after A431 cells were treated with PD98059.With the concentration of PD98059 going up,the decrease in P-ERK and the increase in P53 were more significant(p<.05);while the expression of P-ERK protein was increased but the expression of P53 was decreased after A431 cells were treated with IGF.With the concentration of IGF going up,the increase in P-ERK and the decrease in P53 were more significant(p<.05).According to Pearson correlation analysis,the expression of P53 was negatively correlated to that of P-ERK(p<.05).Conclusions:After MAPK/ERK signaling pathway was activated by IGF in A431 cells,the expression of pro-apoptotic factor P53 was decreased with the ability of cell proliferation enhanced and the ability of apoptosis reduced.However,after the inhibition of MAPK/ERK signaling pathway,the expression of pro-apoptotic factor P53 was increased with the ability of cell proliferation reduced and the ability of apoptosis increased. | Yuqin Hao Chunyi Kang Xin Zhang Shuxia Kang Xia Liu | 2018 | Discussion of Clinical Cases2018,5,4: | 0 |
