1Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 2A small molecular electron acceptor based on asymmetric hexacyclic core of thieno[1,2-b]indaceno[5,6-b']thienothiophene for efficient fullerene-free polymer solar cells显示文摘A novel A-D-A(acceptor-donor-acceptor)type non-fullerene small molecule,A201,consisting of an asymmetric thieno[1,2-b]indaceno[5,6-b^0]thienothiophene(TITT)unit as middle D part and 2-(3-oxo-2,3-dihydroinden-1-ylidene)malononitrile(IC)groups as end-capped A parts was designed and synthesized.The asymmetric TITT building block showed a higher dipole moment of 0.85 Debye(1 Debye=3.33564?10^(à30)cm)compared with the symmetric analogues of indacenodithiophene(IDT)and indacenodithieno[3,2-b]thiophene(IDTT)of 0.098 and 0.13 Debye,respectively.The solution-processed bulk heterojunction solar cells using a benzotriazole(BTA)-based polymer of J71 as donor and A201 as acceptor,showed a power conversion efficiency(PCE)of 9.36%with an open-circuit voltage(V_(oc))of0.88 V,a short-circuit current(J_(sc))of 13.15 m A cm^(à2),and a fill factor(FF)of 0.67,under the illumination of AM 1.5G at 100 m W cm^(à2).The high PCE of this material combination could be attributed to its broad absorption spectrum and the high hole mobility(l_h)and electron mobility(l_e)of 9.56?10^(à4)and 5.17?10^(à4)cm^2V^(à1)s^(à1),respectively.These results indicate that the asymmetric electron-donating segments are promising to construct A-D-A type small molecular acceptors,which could largely enhance the diversity of building blocks to design photovoltaic materials.Wenchao Zhai Ailing Tang Bo Xiao Xiaochen Wang Fan Chen Erjun Zhou 2018Science Bulletin2018,63,13:1
3Constitutive expression of a chrysanthemum phospholipase Dα gene in Chrysanthemum morifolium enhances drought tolerance显示文摘Drought causes water shortage and consequent retardation of plants growth and development.Therefore,improving the drought tolerance of plants is necessary for expanding cultivation and resource promotion.Increasing evidence indicates that phospholipase is involved in the response of plants to drought stress.The objective of this study was to create new drought-tolerant chrysanthemum germplasm,which lays a foundation for the study of the molecular mechanism of phospholipase mediated stress response in chrysanthemum.CmPLDαhas the closest relationship with sunflower HaPLDα,and belongs to the PLDαfamily.CmPLDαover-expressing plants showed a slight shrinking under 20%PEG6000 treatment.The survival rate increased significantly by 1.7−1.8 times that of the wild type.Relative water content(RWC)of CmPLDαover-expressing plants were nearly 10%higher than that of the wild type.Relative electrical conductivity and MDA content were significantly lower than those of the wild type.ABA content of the over-expression lines Z1,Z2 were 1.3 and 1.22 times that of wild type,but ABA content of antisense lines F1,F2 was approximately 0.83 and 0.81 of those of wild type.Most plants of antisense transgenic lines F1,F2 were wrinkled,with a wilting index of 5 and 6,and the survival rate was also lower than that of the wild type after recovery growth.RWC of antisense lines were lower than over-expression lines,relative electrical conductivity and MDA content were significantly higher than those of the wild type.In summary,CmPLDαcould enhance tolerance of chrysanthemum to drought conditions.Lisheng Zhai Xiaochen Zhu Shujin Yang Chunsun Gu Peng Liu Aiping Song Jiafu Jiang Zhiyong Guan Weimin Fang Fadi Chen Sumei Chen 2021Ornamental Plant Research2021,1,1:1
