1Lactate metabolism in human health and disease显示文摘The current understanding of lactate extends from its origins as a byproduct of glycolysis to its role in tumor metabolism,as identified by studies on the Warburg effect.The lactate shuttle hypothesis suggests that lactate plays an important role as a bridging signaling molecule that coordinates signaling among different cells,organs and tissues.Lactylation is a posttranslational modification initially reported by Professor Yingming Zhao’s research group in 2019.Subsequent studies confirmed that lactylation is a vital component of lactate function and is involved in tumor proliferation,neural excitation,inflammation and other biological processes.An indispensable substance for various physiological cellular functions,lactate plays a regulatory role in different aspects of energy metabolism and signal transduction.Therefore,a comprehensive review and summary of lactate is presented to clarify the role of lactate in disease and to provide a reference and direction for future research.This review offers a systematic overview of lactate homeostasis and its roles in physiological and pathological processes,as well as a comprehensive overview of the effects of lactylation in various diseases,particularly inflammation and cancer.Xiaolu Li Yanyan Yang Bei Zhang Xiaotong Lin Xiuxiu Fu Yi An Yulin Zou Jian-Xun Wang Zhibin Wang Tao Yu 2022Signal Transduction and Targeted Therapy2022,7,10:9
2CRISPR/Cas9 therapeutics:progress and prospects显示文摘Clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)gene-editing technology is the ideal tool of the future for treating diseases by permanently correcting deleterious base mutations or disrupting disease-causing genes with great precision and efficiency.A variety of efficient Cas9 variants and derivatives have been developed to cope with the complex genomic changes that occur during diseases.However,strategies to effectively deliver the CRISPR system to diseased cells in vivo are currently lacking,and nonviral vectors with target recognition functions may be the focus of future research.Pathological and physiological changes resulting from disease onset are expected to serve as identifying factors for targeted delivery or targets for gene editing.Diseases are both varied and complex,and the choice of appropriate gene-editing methods and delivery vectors for different diseases is important.Meanwhile,there are still many potential challenges identified when targeting delivery of CRISPR/Cas9 technology for disease treatment.This paper reviews the current developments in three aspects,namely,gene-editing type,delivery vector,and disease characteristics.Additionally,this paper summarizes successful examples of clinical trials and finally describes possible problems associated with current CRISPR applications.Tianxiang Li Yanyan Yang Hongzhao Qi Weigang Cui Lin Zhang Xiuxiu Fu Xiangqin He Meixin Liu Pei-feng Li Tao Yu 2023Signal Transduction and Targeted Therapy2023,8,2:0
