1m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. 3Role of fractional flow reserve in guiding intervention for borderline coronary lesions显示文摘Objective:This study investigated the clinical efficacy and value of fractional flow reserve(FFR)in guiding the treatment of borderline coronary lesions.Methods:Forty-three patients with borderline coronary lesions,as demonstrated by coronary angiography,and who had FFR measurements were selected.The patients were grouped accord-ing to FFR values.All patients were evaluated 6 months after surgery to record major adverse cardiac events(MACE[sudden cardiac death,non-fatal myocardial infarction,or revasculariza-tion])and recurrence of angina pectoris.Results:After the 6-month follow-up,no sudden cardiac deaths or myocardial infarctions occurred in either group,and there were no statistically significant differences(P>0.05).Inter-group comparisons showed that in the groups with a FFR<0.75,the recurrence rate of angina pectoris in the PCI group was significantly lower than the drug therapy group(0.08%vs.0.27%,P<0.05).In contrast,the recurrence rate of angina pectoris in the PCI group among the groups with a FFR<0.75 revealed no statistical significance when compared to the groups with a FFR≥0.75(0.08%vs.0.05%,P>0.05).The recurrence rate of angina pectoris in the simple drug therapy group among the groups with a FFR<0.75 was higher than the same groups with a FFR≥0.75(0.27%vs.0.05%,P<0.05).Conclusion:When coronary intervention is used to treat borderline lesions,guiding inter-ventional therapy with measurement of FFR does not increase the incidence of adverse cardiovas-cular events in the short term and can better guide PCI therapy.Xuekun Shi Lu Cheng 2014Family Medicine and Community Health2014,2,3:0
4Influence of Virtual Synchronous Generators on Low Frequency Oscillations显示文摘By simulating the operating dynamics of synchronous generators(SGs),the use of virtual synchronous gen-rators(VSGs)can help overcome inverter-based generators'shortcomings of low inertia and minimal damping for gridforming applications.VSGs'stability are very important for their solar and wind electricity applications.Currently,the related research primarily focuses on VSGs and their applications for microgrids.There has been little research to explore how VSGs effect low frequency oscillations in power transmission systems.This paper describes a small-signal model of a VSGSG interconnected system,which is suitable for studying low frequency oscillation damping in a power transmission grid.Based on this model,the effects of VSGs on low frequency oscillations are compared with the effects of SGs to reveal the mechanism of how VSGs infuence damping characteristics.The influence of each VSG control loop on oscillations is also analyzed in this paper.Then,the low frequency oscillation risks with different types of VSGs are described.Finally,experiments on a real-time laboratory(RT-LAB)platform are conducted to verify the small-signal analysis results.Hui Liu Dawei Sun Feng Zhao Yunfeng Tian Peng Song Xuekun Cheng 2022CSEE Journal of Power and Energy Systems2022,8,4:0
