| 1 | Dissimilatory Fe(Ⅲ) reduction characteristics of paddy soil extract cultures treated with glucose or fatty acids显示文摘Dissimilatory Fe(Ⅲ) reduction is a universal process with irreplaceable biological and environmental importance in anoxic environments. Our knowledge about Fe(Ⅲ) reduction predominantly comes from pure cultures of dissimilatory Fe(Ⅲ) reducing bacteria (DFRB). The objective of this study was to compare the effects of glucose and a selection of short organic acids (citrate, succinate, pyruvate, propionate, acetate, and formate) on Fe(Ⅲ) reduction via the anaerobic culture of three paddy soil solutions with Fe(OH)3 as the sole electron acceptor. The results showed significant differences in Fe(Ⅲ) reduction among the three paddy soil solutions and substrate types. Bacteria from the Sichuan paddy soil responded quickly to substrate supply and showed higher Fe(Ⅲ) reducing activity than the other two soil types. Bacteria in the Jiangxi paddy soil culture solution could not use propionate as a source of electrons for Fe(Ⅲ) reduction. Similarly, bacteria in the Jilin paddy soil culture could not use succinate effectively. Pyruvate was readily used by cultures from all three paddy soil solutions, thus indicating that there were some similarities in substrate utilization by bacteria for Fe(Ⅲ) reduction. The use of glucose and citrate as substrate for dissimilatory Fe(Ⅲ) reduction indicates important ecological implications for this type of anoxic respiration. | HE Jiangzhou1,2, QU Dong1, 1. | 2 | The EST Analysis of A Suppressive Subtraction cDNA Library of Chinese Wild Vitis pseudoreticulata Inoculated with Uncinula necator显示文摘Uncinula necator is a worldwide serious fungus disease causing annual heavy lost on grapevine production. In order to get more informations on defense related EST sequences and help breeding program, we constructed and characterized a suppression subtractive hybridization cDNA library with artificially inoculated leaves and control Chinese wild Vitis pseudoreticulata clone Baihe-35-1, which is highly resistant to powdery mildew. In the library, the length of 58 EST fragments known as putative functions varied from 130 to 800 bp, and 60% of the ESTs exhibited high similarity to known sequences in database of GenBank with BLASTX analysis. These genes were involved in stress/defense response, detoxification, signal transduction, disease defense, and etc., and 14 ESTs remained unknown or hypothetical proteins, which may be new genes. | 5 | Effects of Microtubule Polymerization Inhibitor on the Hypersensitive Response of Wheat Induced by the Non-Host Pathogen Sphaerotheca fuliginea显示文摘The plant cytoskeleton is a highly dynamic and versatile intracellular scaffold composed of microtubules and microfilaments, serving a multiplicity of functions in plant cells. To reveal the relationship between the cytoskeleton in wheat (Triticum aestivum L.) cv. Suwon 11 attacked by the non-host pathogen Sphaerotheca fuliginea and the initiation of the hypersensitive response, the microtubule inhibitor oryzalin was injected into the wheat leaves immediately prior to inoculation. The incidence of hypersensitive cell death was significantly lower than that in water-treated control. In addition, the occurrence of hypersensitive cell death was also delayed and S. fuliginea was able to penetrate and form haustoria in epidermal tissues of wheat. | 11 | cDNA Cloning of Goat DNA Methyltransferase 1,Screening of shRNA Vectors and Influences to Development of Nuclear Transfer Embryos显示文摘This study was designed to clone cDNA of goat DNA methyltransferase 1(DNMT1) gene,to screen an effective shRNAproducing vector targeting goat DNA methyltransferase 1 and to improve the developmental competence of goat nuclear transfer embryos by decreasing the DNMT1 expression in donor cells.In this study,PCR primers were designed against regions of high homology between bovine and sheep sequences and then used to amplify the larger portions of the coding regions.Next,3 RNAi oligonucleotides were designed based on the cloned sequences and inserted into pRNAT-U6.1/Neo vector,acquiring 3 new vectors,respectively termed pRNAD1,pRNAD2 and pRNAD3.Then the positive cells were sorted by flow cytometry after transfection and detected by real-time PCR analysis and sodium bisulfite genomic sequencing.Finally,the developmental rates of nuclear transfer(NT) embryos generated using donor cells with and without the effective shRNA vector respectively,as well as in vitro fertilization(IVF) embryos were observed and recorded.The results showed that the coding regions of goat DNA methyltransferase 1 gene was successfully cloned(GenBank no.FJ617538).Furthermore,an effective interfering shRNA(pRNAD2) was obtained,with its interference effect being 47.88%.Finally,NT embryos with shRNA vector harbored better developmental competence during morula and blastocyst stage compared to controls(P < 0.05),reaching the similar rates to IVF embryos(P > 0.05).In conclusion,goat DNA methyltransferase 1 gene cDNA was cloned and sequenced,an effective shRNA vector responsible for inhibiting DNA methyltransferase 1 expression was developed and the developmental competence of goat nuclear transfer morulae and blastcysts was significantly improved,which provided a feasible pathway for improving goat nuclear transfer embryo development competence by decreasing the methylation level in donor cells through RNAi-mediated manner. | LAN Jie,SONG Yong-li,HUA Song,LIU Yong-gang,LIU Jun,ZHANG Hai-lin and ZHANG Yong Key Laboratory of Animal Reproductive Physiology & Embryo Technology,Institution of Biotechnology,College of Veterinary Medicine,Northwest A&F University,Yangling 712100,P.R.China | 2010 | Agricultural Sciences in China2010,9,7: | 1 |

| 17 | Expression of EGF and EGFR in Developing Skin of the Goat Fetus显示文摘The histological characteristics,distribution and variability of epidermal growth factor(EGF) and epidermal growth factor receptor(EGFR) during development of skin in the goat fetus were studied with histological and immunohistochemical methods.The results showed that the epidermis formed at the 6th gestational week,the thickness increased gradually because of transition from simple epithelium to stratified epithelium,and then decreased after the 15th week.The dermis was present at the 10th week,and the dermal derivatives appeared after the 11th week.All structures of the skin matured gradually during prenatal development.Low expression of EGF and EGFR could be observed at the 6th week and increased thereafter.Before the 11th week,EGF was mainly located in the cytoplasm of the basal layer of cells of epidermis,hair follicular epithelial cells and fibroblasts,and EGFR was mainly located on cell membranes of these cells.From the 11th to 16th week,the expression of EGF and EGFR extended from basal layer cells,prickle cells,hair follicular epithelial cells and fibroblasts to vascular endothelial cells,epithelial cells of sweat glands and arrector pili muscle.EGF was mainly detected cytoplasmically,and EGFR was located on membranes of these cells.From the 17th week to birth,EGF was mainly located in cells of the basal layer and hair follicular epithelial cells as the skin thinned and staining intensity continued to increase.The expressed amount of EGF and EGFR tended to increase throughout the whole development stage encompassed by gestational weeks 6 to 21 and the two components of the EGF signaling system were highly correlated.The results suggest that EGF and EGFR play important roles in the development of skin and its derivatives. | QING Su-zhu,LIN Ji-mao,ZHANG Ling-zhi,WANG Lei-lei,LUO Shi-shuang (College of Veterinary Medicine,Northwest A & F University,Yangling 712100,China) | 2010 | 畜牧兽医学报2010,41,S1: | 0 |

| 19 | Factors Influencing Willingness and Ability of Farmers to Adopt New Technologies:A Case Study of Guanzhong Area显示文摘Based on data of 248 rural households in Pucheng County and Huxian County,we established the Two-Level Logit Model to analyze the willingness of farmers to adopt new technologies,its influence factors,and probability of successful adoption of new technologies.Results show that the willingness has positive correlation with whether the farmer is head of household,the educational level,occupation,agricultural loan,the number of family labor,and information dissemination channel,while it has negative correlation with non-agricultural employment proportion and whether the farmer is village cadre.In the model of the probability of farmers'successfully adopting new technologies,occupation,agricultural loan,planting area,gender and educational level are positively correlated,while age and non-agricultural employment proportion are negatively correlated.Largescale flow of rural labor plays a negative role in popularization of technologies in rural areas through influencing factors,including number of family labor,non-agricultural employment proportion,educational level,gender,and whether the farmer is village cadre.Finally,on the basis of results of empirical study,we put forward countermeasures and suggestions for strengthening ability of farmers to adopt new technologies. | TIAN Jing-na,LI Lu-tang College of Economics and Management,Northwest A&F University,Yangling 712100,China | 2012 | Asian Agricultural Research2012,4,7: | 0 |
