| 1 | Phosphorus forms in sediments of the East China Sea and its environmental significance显示文摘By means of SEDEX, ASPILA and XRF, depth-dependent changes of different phosphorus forms in sediment cores from specific areas of the offshore Changjiang Estuary (Yangtze Estuary) in 1998 were analyzed. Results show that contents of total phosphorus (TP), organic-phosphorus (OP) and iron-phosphorus (Fe-P) decreased down-core, while those of absorbed-phosphorus (Ad-P) and calcium-phosphorus (Ca-P) increased. The distribution tendency of detritus-phosphorus (De-P) is not obvious. Results also show that TP, Fe-P and OP contents at Meso station of the Changjiang Estuary and Hangzhou Bay are higher than that of the other stations. This suggests that the pollutants carried by the Changjiang and the Qiantang rivers from inland have affected the natural environment in offshore area. TP, Fe-P and OP contents of each station become higher from bottom to top, indicating the amount of the terrestrial pollutants carried by the two rivers has been enhanced since the last 30-50 years. | 2 | Lewis blood genotypes of peptic ulcer and gastric cancer patients in Taiwan显示文摘AIM: The Lewis b (Leb) antigen has been implicated as a possible binding site for attachment of Helicobacter pylori (H pylori)to gastric mucosa. However, studies both supporting and denying this association have been reported in the literature. Differences in secretor (Se)genotype have been suggested as a possible reason for previous discrepancies. Therefore, we investigated the relationship between Le and Se genotypes and H pylori infection rates in people with peptic ulcer or gastric cancer.METHODS: Peripheral blood samples were obtained from 347 patients with endoscopic evidence of peptic ulcer disease (235 cases of duodenal ulcer, 62 of gastric ulcer,and 50 of combined duodenal ulcer/gastric ulcer) and 51patients with gastric cancer on endoscopy. Peripheral blood specimens from 101 unrelated normal volunteers were used as controls. Lewis phenotype was determined using an antibody method, whereas Le and Se genotypes were determined by DNA amplification and restriction enzyme analysis. Gastric or duodenal biopsies taken from patients with endoscopic evidence of peptic ulcer or gastric cancer were cultured for H pylori. Isolates were identified as H pylori by morphology and production of urease and catalase. The H pylori infection status was also evaluated by rapid urease test (CLO test), and urea breath test (13C-UBT). Results of studies were analyzed by chi-square test (taken as significant).RESULTS: H pyloriwas isolated from 83.7% (303/347)of patients with peptic ulcer disease. Statistical analysis did not show any significant difference in Lewis phenotype or genotype between patients with and without H pylori infection. | 3 | Investigation of physico-chemical properties and microbial community during poultry manure co-composting process显示文摘Co-composting of poultry manure and rubber wood sawdust was performed with the ratio of 2:1(V/V) for a period of 60 days. An investigation was carried out to study the extracellular enzymatic activities and structural degradation utilizing Fourier transform infrared spectroscopy(FT-IR), thermogravimetry and differential thermal analysis(TG/DTA)and scanning electron microscopy(SEM). The microbial succession was also determined by using denaturing gel gradient electrophoresis(DGGE). The compost was able to reach its highest temperature of 71°C at day 3 and stabilized between 30 and 40°C for 8 weeks.CMCase, FPase and β-glucosidase acted synergistically in order to degrade the cellulosic substrate. The xylanase activities increased gradually during the composting and reached the peak value of 11.637 U/g on day 35, followed by a sharp decline. Both Li P and Mn P activities reached their peak values on day 35 with 0.431 and 0.132 U/g respectively. The FT-IR spectra revealed an increase in aromaticity and a decrease in aliphatic compounds such as carbohydrates as decomposition proceeded. TGA/DTG data exhibited significant changes in weight loss in compost samples, indicating degradation of organic matter. SEM micrographs showed higher amounts of parenchyma exposed on the surface of rubber wood sawdust at day 60, showing significant degradation. DGGE and 16 S r DNA analyses showed that Burkholderia sp., Pandoraea sp., and Pseudomonas sp. were present throughout the composting process. | 4 | Identification of differential genes in the ovary relative to the testis and their expression patterns in half-smooth tongue sole (Cynoglossus semilaevis)显示文摘Half-smooth tongue sole (Cynoglossus semilaevis) is a rare marine flatfish whose mature ovary and testis greatly differ in volume and weight. The length and weight of mature females are over twice greater than those of mature males. To obtain sufficient information on gonad differentiation and the relationship between gonad development and growth in the fish, we compared gene expression between the ovary and testis using suppression subtractive hybridization (SSH). Testis cDNAs are subtracted from ovary cDNAs and are used to es- tablish an ovary testis-subtracted cDNA library. A total of 41 nonredundant clones are identified, including 20 known cDNAs, 9 unchar-acterized cDNAs (EST clones), and 12 novel sequences. For selected genes such as ZPC, RacGAP, survivin, aquaporin, CPEB, O5, O15, and O18, gene expression patterns are analyzed by reverse transcriptase-polymerase chain reaction (RT-PCR). The results confirm that these genes are only expressed in the ovary and not in the testis, or at higher levels in the ovary than in the testis. At the same time, expressions of certain genes such as ZPC, survivin, aquaporin, CPEB, and O15 are demonstrated to possess sexual dimorphism in the kidney or muscle, and spleen. | 5 | Study of the Gene Expression Profile of Human Ovarian Carcinoma by a Gene Chip显示文摘OBJECTIVE To study the difference in gene expression between human ovarian carcinoma and normal ovarian tissues, and screen the novel associated genes by cDNA microarrays. METHODS Total RNA from 10 cases of ovarian cancer and from normal ovarian tissues were extracted by a single step method. The cDNA was retro-transcribed from an equal quantity of mRNA derived from the 10 cases of ovarian carcinoma and normal ovarian tissues, followed by labeling the cDNA strands with Cy5 and Cy3 fluorescence as probes. The mixed probes were hybridized with BiostarH 8464 dot human somatic cell genes. Fluorescence signals were assessed by a ScanArray 4000 laser scanner and the images analyzed by Gene Pix Pro 3.0 software with a digital computer. RESULTS By applying the cDNA microarray we found a total of 185 genes for which expression levels differed more than 5 times comparing human ovarian carcinoma with normal ovarian epithelium. Among these genes 86 were up-regulated >5 times and 99 were down regulated <0.2. CONCLUSION The cDNA microarray technique is effective in screening the differential gene expression between human ovarian cancers and normal ovarian epithelium. | 6 | Discovery and Genetic Mechanism of Basic Granulite in the Altay Orogenic Belt, Xinjiang, NW China显示文摘Altay granulite (AG), which represents the product of high-grade metamorphism in the lower crust, was newly found in thc Wuqiagou area, Fuyun County in the Altay orogenic belt, Northwest China. It is composed mainly of hypersthene, augite, basic plagioclase, amphibole and brown biotite. Its mineral compositions of amphibole and biotite are rich in Mg/(Mg+Fe2+) and Ti. Geochemically, the AG is enriched in Mg/(Mg+Fe2+) and A12O3, and poor in CaO, with depletion of U, Th, K and Rb contents. Furthermore, geochemical data reflect that the protolith of the AG is igneousgenetic calc-alkalinc basalt formed under an island arc environment. The AG has ∑REE of 92.38-96.58 ppm and enriched LREE model with weak positive Eu anomaly of 1.09-1.15. In the MORB normalized spider diagram, the AG shows tridoming pattern with a strong negative Nb anomaly and medium negative P and Ti anomalies, reflecting that the AG has tectonic relation with subduction or subduction-related materials. The P-Tconditions of peak metamorphism of the AG arc 750-780°C and >0.6-0.7 GPa. Retrograde metamorphism implies that the protolith of the Altay granulite might undergo a metamorphic process along a clockwise P-T trajectory.
