| 2 | Mechanical microenvironments as key cellular regulator in the liver显示文摘Tissue stiffness, shear stress, and interstitial pressure make up major factors of liver mechanical microenvironment, which play the key regulatory role to control cell behaviors in the liver and progress of liver diseases. In this review, we focus on the characteristics of liver mechanical microenvironments and summarize cellular responses to mechanobiological changes during liver pathogenesis, especially in hepatic fibrosis and cirrhosis. | 3 | Spotlight SAR sparse sampling and imaging method based on compressive sensing显示文摘Spotlight synthetic aperture radar(SAR)emits a chirp signal and the echo bandwidth can be reduced through dechirp processing,where the A/D sampling rate decreases accordingly at the receiver.Compressive sensing allows the compressible signal to be reconstructed with a high probability using only a few samples by solving a linear program problem.This paper presents a novel signal sampling and imaging method for application to spotlight SAR based on compressive sensing.The signal is randomly sampled after dechirp processing to form a low-dimensional sample set,and the dechirp basis is imported to reconstruct the dechirp signal.Matching pursuit(MP)is used as a reconstruction algorithm.The reconstructed signal uses polar format algorithm(PFA)for imaging.Although our novel mechanism increases the system complexity to an extent,the data storage requirements can be compressed considerably.Several simulations verify the feasibility and accuracy of spotlight SAR signal processing via compressive sensing,and the method still obtains acceptable imaging results with 10%of the original echo data. | XU HuaPing YOU YaNan LI ChunSheng ZHANG LvQian | 2012 | Science China(Information Sciences)2012,55,8: | 3 |
| 4 | Freeze substitution Hi-C,a convenient and cost-effective method for capturing the natural 3D chromatin conformation from frozen samples显示文摘Chromatin interactions functionally affect genome architecture and gene regulation,but to date,only fresh samples must be used in High-through chromosome conformation capture(Hi-C)to keep natural chromatin conformation intact.This requirement has impeded the advancement of 3 D genome research by limiting sample collection and storage options for researchers and severely limiting the number of samples that can be processed in a short time.Here,we develop a freeze substitution Hi-C(FS-Hi-C)technique that overcomes the need for fresh samples.FS-Hi-C can be used with samples stored in liquid nitrogen(LN2):the water in a vitreous form in the sample cells is replaced with ethanol via automated freeze substitution.After confirming that the FS step preserves the natural chromosome conformation during sample thawing,we tested the performance of FS-Hi-C with Drosophila melanogaster and Gossypium hirsutum.Beyond allowing the use of frozen samples and confirming that FS-Hi-C delivers robust data for generating contact heat maps and delineating A/B compartments and topologically associating domains,we found that FS-HiC outperforms the in situ Hi-C in terms of library quality,reproducibility,and valid interactions.Thus,FS-HiC will probably extend the application of 3D genome structure analysis to the vast number of experimental contexts in biological and medical research for which Hi-C methods have been unfeasible to date. | Wu Zheng Zhaoen Yang Xiaoyang Ge Yijia Feng Ye Wang Chengwei Liu Yanan Luan Kun Cai Serhii Vakal Feng You Wei Guo Wei Wang Zhenhua Feng Fuguang Li | 2021 | Journal of Genetics and Genomics2021,48,3: | 0 |
| 5 | Integrated analyses of transcriptomics and network pharmacology reveal leukocyte characteristics and functional changes in subthreshold depression,elucidating the curative mechanism of Danzhi Xiaoyao powder显示文摘Objective:To investigate the molecular mechanism and identify potential drugs for subthreshold depression(SD),and elucidate the detalied mechanism of Danzhi Xiaoyao powder(DZXY)in SD.Methods:Using RNA-sequencing,we identified differentially expressed genes(DEGs)in leukocytes of SD compared to healthy controls,deciphered their functions and pathways,and identified the hub genes of SD.We also assessed changes in leukocyte transcription factor activity in patients with SD using the TELis platform.The Connectivity Map database was retrieved to screen candidate drugs for SD.Based on network pharmacology,we elucidated the'multi-component,multi-target,and multi-pathway'mechanism of DZXY in the treatment of SD.Results:We identified 1080 DEGs(padj<0.05 and|log2(fold change)l≥1&protein coding)in the leukocytes of patients with SD.These DEGs,including hub genes,were primarily involved in immune and inflammatory response-related processes.Transcription factor activity analysis revealed similarities between the leukocyte transcriptome profile in SD and the conserved transcriptional response to adversities in immune cells.Connectivity Map analysis identified 28 potential drugs for SD treatment,particularly SB-202190 and TWS-119.Constructing the'Direct Compounds-Direct Targets-Pathways'network for DZXY and SD revealed the curative mechanisms of DZXY in SD,primarily including inflammatory response,lipid metabolism,immune response,and other processes.Conclusion:These results provide new insights into the characteristics and functional changes of leukocytes in SD,partially illustrate the pathogenesis of SD,and suggest potential drugs for SD.The curative mechanisms of DZXY in SD are also partially elucidated. | Kunyu Li Leiming You Jianhua Zhen Guangrui Huang Ting Wang Yanan Cai Yunan Zhang Anlong Xu | 2024 | Journal of Traditional Chinese Medical Sciences2024,11,1: | 0 |
