| 8 | Inhibition of the α-mannosidase Man2c1 gene expression enhances adhesion of Jurkat cells显示文摘Protein N-glycosylation plays very important roles in immunity and α-mannosidase is one of the key enzymes in N-glycosylation.This paper reports that inhibition of α-mannosidase Man2cl gene expression enhances adhesion of JurkatT cells.In comparison to the controls with normal expression of the enzyme,Jurkat cells with the inhibition of Man2clgene expression(AS cell)formed larger aggregates in culture,indicating an enhancement of adhesion between the cells.mRNA differential display analysis discovered up-regulation of several adhesion molecule genes in the AS cell.Becauseof the pivotal role played by CD54-LFA-1 interaction in immune cell interaction,this study focused on the contributionof enhanced expression of CD54 and LFA-1 to the enhanced adhesion of AS Jurkat cells.These facts,including increasedbinding of AS cells to ICAM-1-Fc,Mg^(2+)activation of the binding of AS cells to ICAM-1-Fc and enhanced aggregationof AS cells,together with the inhibiting effect of a blocking CD 11a mAb on the binding to ICAM-1-Fc and aggregationof the cells demonstrate an important contribution of enhanced CD54-LFA-1 interaction to increased adhesion betweenAS cells.The enhanced CD54-LFA-1 interaction also resulted in increased adhesion between AS Jurkat T cells and RajiB cells.In addition,AS cells showed cytoskeletal rearrangement.The data imply a biological significance of MAN2C1in T-cell functioning. | Li Qu Ji Yu Ju Shuang Ling Chen Yah Shi Zhi Guang Xiang Yi Qun Zhou Yun Tian Yin Liu Li Ping Zhu | 2006 | Cell Research2006，16，7： | 1 |
| 9 | Development of an enzyme-linked immunosorbent assay using specific monoclonal antibodies against theacrine and its application显示文摘Theacrine(1,3,7,9-tetramethyluric acid),a purine alkaloid similar to caffeine in its chemical structure,is isolated from edible Camellia assamica var.kucha and has various pharmacological activities including hypnotic effects,anti-depressant effects,anti-inflammatory and analgesic effects,and a protective effect against stress-provoked liver damage.A rapid and simple assay is required to quantify theacrine in biological samples for pharmacokinetic studies in small animals.This study aimed to establish an enzyme-linked immunosorbent assay(ELISA) for theacrine quantification in blood.Herein,we successfully obtained monoclonal antibodies(MAbs) against theacrine,MAbs C11B5,and developed an ELISA method for the fast determination of theacrine in mouse blood.The range for calibration of theacrine by ELISA was 0.156-100 μg mL^1.The half maximum inhibitory concentration(IC_(50)) value was1.55 μg mL^1.The ELISA method lays a good foundation for the further research. | Zhuang Li Jun He Yue Zhang Qing-Guo Wang Yah Zhao Hui-Hua Qu Xue-Qian Wang Hui Kong Wei-Ku Zhang Jie-Kun Xu | 2016 | Chinese Chemical Letters2016，27，12： | 1 |
