2Osteoarthritis:toward a comprehensive understanding of pathological mechanism显示文摘Osteoarthritis(OA) is the most common degenerative joint disease and a major cause of pain and disability in adult individuals. The etiology of OA includes joint injury, obesity, aging, and heredity. However, the detailed molecular mechanisms of OA initiation and progression remain poorly understood and, currently,there are no interventions available to restore degraded cartilage or decelerate disease progression. The diathrodial joint is a complicated organ and its function is to bear weight, perform physical activity and exhibit a joint-specific range of motion during movement. During OA development, the entire joint organ is affected, including articular cartilage, subchondral bone, synovial tissue and meniscus. 4A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 5塔里木盆地寒武纪—奥陶纪优质烃源岩沉积与古环境变化的关系:碳氧同位素新证据显示文摘对塔里木盆地东部塔东2井上震旦统和寒武系—奥陶系样品进行TOC和碳、氧同位素及微量元素分析,发现该井稳定碳、氧同位素值在寒武系—中奥陶统发生明显变化,该层还赋存高有机质丰度烃源岩,其TOC值的高低可反映当时的生物产率和埋藏率。碳和氧同位素值在寒武系/上震旦统分界处正向漂移,说明正好在进入寒武纪前气候明显变冷,海平面下降。接着,下寒武统底部以碳和氧同位素值的快速负向漂移为标志,表明古气候迅速变暖和海平面大幅度上升,烃源岩TOC值也达到全井柱最高值。古气候显著波动、海平面最高及其频繁变化和沉积物高TOC是早寒武世的显著特征;而晚寒武世—早奥陶世,同位素正向漂移,暗示海平面下降,沉积物TOC也随之下降并降至最低;到了早奥陶世末,即早、中奥陶世的过渡时期,同位素又开始负向漂移,TOC又开始升高;中奥陶世,同位素强烈负向漂移,TOC又升至新高。稳定碳、氧同位素值的变化及其所指示的古气候与海平面变化,以及与TOC的响应关系,指示冰期、冰后期之交,古气候迅速变暖和海平面大幅度上升有利于烃源岩发育;同时暗示,δ13C、δ18O与生烃母质生物的有机生产率、有机埋藏率之间存在某种内在联系。即高TOC含量反映高的有机生产率和高的有机埋藏率;而与其同步反方向变化的δ13C、δ18O则暗示海平面较高,沉积环境属远陆海域,表层水中生烃母质生物的光合作用很强。而有机质的高产率和高埋藏率,导致海水中δ12C和δ16O被大量地固定在沉积物中。这样,就使海水中相对富集13C和18O重同位素而使海水变“重”;而这种“重”海水,又导致了同时期浅水区碳酸盐沉积物的δ13C和δ18O值明显增高。张水昌 R L WANG 金之钧 张宝民 王大锐 边立曾 2006地质学报2006,80,3:65
6Relocation of the mainshock and aftershock sequences of M_S7.0 Sichuan Lushan earthquake显示文摘The mainshock of April 20,2013 Sichuan Lushan MS7.0 earthquake was relocated using a 3-D velocity model.Double difference algorithm was applied to relocate aftershock sequences of Lushan earthquake.The locations of 2405 aftershocks were determined.The location errors in E-W,N-S and U-D direction were 0.30,0.29 and 0.59 km on average,respectively.The location of the mainshock is 102.983°E,30.291°N and the focal depth is 17.6 km.The relocation results show that the aftershocks spread approximately 35 km in length and 16 km in width.The dominant distribution of the focal depth ranges from 10 to 20 km.A few earthquakes occurred in the shallow crust.Focal depth profiles show fault planes dip to the northwest,manifested itself as a listric thrust fault.The dip angle is steep in the shallow crust and gentle in the deep crust.Although the epicenters of aftershocks distributed mainly along both sides of the Shuangshi-Dachuan fault,the seismogenic fault may be a blind thrust fault on the eastern side of the Shuangshi-Dachuan fault.Earthquake relocation results reveal that there is a southeastward tilt aftershock belt intersecting with the seismogenic fault with y-shape.We speculate it is a back thrust fault that often appears in a thrust fault system.Lushan earthquake triggered the seismic activity of the back thrust fault.FANG LiHua WU JianPing WANG WeiLai Lü ZuoYong WANG ChangZai YANG Ting CAI Yan 2013Chinese Science Bulletin2013,58,28:74
7Molecular mechanism about lymphogenous metastasis of hepatocarcinoma cells in mice显示文摘AIM To investigate the correlation between lymphogenous metastasis and matrix metalloproteinases (MMPs) activity and the expression of Fas Iigand of tumor cells in lymph nodes.METHODES Fifty-six inbred 615-mice were equally divided into 2 groups and inoculated with Hca-F and Hca-P cells. Their lymph node metastatic rates were examined.Growth fraction of lymphocytes in host lymph nodes was detected by flow cytometry. The Hca-F and Hca-P cells were cultured with extract of lymph node, liver or spleen.The quantity of MMPs in these supernatants was examined by zymographic analysis. The expression of Fas ligand,PCNA, Bcl-2 protein of Hca-F and Hca-P cells in the mice were examined by immunohistochemistry. The apoptosis signals of macrophages in lymph nodes were observed with in situ DNA fragmentation.RESULTS On the 28th day post-inoculation, the lymph node metastatic rate of Hca-F was 80% (16/20), whereas that of Hca-P was 25% (5/20). The growth fraction of lymphocytes was as follows: in the Hca-F cells, the proliferating peak of lymphocytes appeared on the 14th day post-inoculation and then decreased rapidly, while in HcaP cells, the peak appeared on the 7th day post-inoculation and then kept at a high level. With the extract of lymph node, the quantity of the MMP-9 activity increased (P<0.01) and active MMP-9 and MMP-2 were produced by both Hca-F and Hca-P tumor cells, which did not produce MMPs without the extract of lymph node or with the extracts of the liver and spleen. The expression of Fas Ligand of Hca-F cells was stronger than that of Hca-P cells (P<0.01). The expressions of PCNA and Bcl-2 protein of Hca-F cells in the tumors of inoculated area were the same as that of Hca-P cells. In situ DNA fragmentation showed that the positive signals of macrophages were around HcaF cells.CONCLUSION Secretion of MMPs which was associated with metastatic ability of Hca-F and Hca-P tumor cells depends on the environment of lymph nodes. 10The crustal structure under Sanjiang and its dynamic implications:Revealed by seismic reflection/refraction profile between Zhefang and Binchuan,Yunnan显示文摘The fault belts in Sanjiang mainly include Jinshajiang-Honghe fault, Lancangjiang fault and Nujiang fault (called Sanjiang faults) in western Yunnan Province, China. By interpreting the wide-angle seismic reflection/refraction profile between Zhefang and Binchuan, which crosses Tengchong and Baoshan blocks in Dianxi (western Yunnan) tectonic zone, we recon- struct the crustal structure with seismic traveltime tomography for crustal P-wave velocity and the seismic scattering image for crustal seismic reflection structure. In this paper, we firstly present the crustal structure images of P-wave velocity and seismic reflection under the wide-angle seismic profile. These results demonstrate that, the crustal velocity structure and seismic reflec- tion structure along the profile can be divided into 3 segments, and there is an obvious difference of crustal structure among the eastern, the western and the middle segment. Generally, crustal P-wave velocities in the Baoshan segment are 0.1―0.2 km/s slower and seismic reflection am- plitudes from Moho discontinuity are stronger than the other 2 segments. In the studied area, crustal thickness is about 40 km, and shows the thickening tendency from west to east along the profile. Additionally, it can be seen that there is one strong-amplitude seismic reflection event as bright points at the depths of 8―10 km, along the segment of 80―115 km of the profile (south- ward of Tengchong); and seismic reflection wave-field from Moho discontinuity varies obviously along the lateral direction. 15Periodicity of Holocene climatic variations in the Huguangyan Maar Lake显示文摘There exist five primary periods of 2 930, 1 140, 490, 250 and 220 a in the Holocene climatic variations in the Huguangyan Maar Lake, according to the energy-spectrum and filter analyses of high-resolution time sequences (10-15 a) of the sediment dry density. The peak values of the three temperature-decreasing periods with the 2 930 a cycle occur at about 7 300, 4 250 and 1 200 Cal. aBP. There are 7-8 temperature-decreasing periods with the 1 140 a cycle, and the climate fluctuation range is largest in the early Holocene, and reduces gradually in the middle and late Holocene. 16Estrogen enhances the functions of CD4^(+)CD25^(+)Foxp3^(+) regulatory T cells that suppress osteoclast differentiation and bone resorption in vitro显示文摘Cross-talk has been shown to occur between the immune system and bone metabolism pathways.In the present study,we investigated the impact of CD4^(+)CD25^(+)Foxp3^(+) regulatory T(Treg)cells on osteoclastogenesis and bone resorption.Treg cells that were isolated and purified from peripheral blood mononuclear cells(PBMCs)of healthy adults inhibited both the differentiation of osteoclasts(OCs)from human embryo bone marrow cells(BMCs)and the pit formation in a dose-dependent manner.In cell cocultures,the production levels of both interleukin-10(IL-10)and transforming growth factor-beta 1(TGF-β1)were proportionally upregulated as the ratio of Treg cells to BMCs was increased,and the inhibition of OC differentiation and bone resorption by Treg cells was completely reversed by anti-IL-10 and anti-TGF-β1 antibodies.Treatment of BMC and Treg cell cocultures with 17β-estradiol(E2)at concentrations between 10^(-7) and 10^(-9) mol/l suppressed OC differentiation and bone resorption more efficiently than it did in cultures of BMCs alone;this enhanced suppression occurred via the stimulation of Treg cell IL-10 and TGF-b1 expression.These data suggest that Treg cells suppress OC differentiation and bone resorption by secreting IL-10 and TGF-β1.E2 enhances the suppressive effects of Treg cells on OC differentiation and bone resorption by stimulating IL-10 and TGF-β1 secretion from these cells.Therefore,Treg cell-derived IL-10 and TGF-b1 are likely involved in the regulation of E2 on bone metabolism and represent potential therapeutic targets for the treatment of postmenopausal osteoporosis(PMO).CY Luo L Wang C Sun DJ Li 2011Cellular & Molecular Immunology2011,8,1:33
17Crustal structure of Gondwana-and Yangtze-typed blocks:An example by wide-angle seismic profile from Menglian to Malong in western Yunnan显示文摘The wide-angle seismic profile between Menglian and Malong crosses the Baoshan block (Gondwana-typed), and Simao and southwestern Yangtze blocks (Yangtze-typed). By in-terpreting the wide-angle seismic data, we obtained the seismic crust/upper mantle structure of P-wave velocities together with the seismic reflections of these three blocks, Changning- Menglian and Mojiang suture zones among the mentioned three blocks. Our interpreting results demonstrate that the P-wave crustal velocity of Simao block is slower than that of Baoshan and southwestern Yangtze block and the crustal thickness gradually thickens from the Baoshan block, Simao to southwestern Yangtze block. Crustal reflection patterns of these three blocks have dis-tinct differences too. For the Gondwana-typed blocks, seismic reflections in the upper crust are well developed while in middle-lower crust they are very weak. The crustal reflections in the Yangtze block are very well developed. The crustal reflection patterns in Simao and southwest-ern Yangtze blocks are distinguishable. The average thickness of the crust in the studied area is about 40 km.
