| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |

| 4 | High-throughput genome editing in rice with a virus-based surrogate system显示文摘With the widespread use of clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated nuclease(Cas) technologies in plants, large-scale genome editing is increasingly needed. Here, we developed a geminivirus-mediated surrogate system, called Wheat Dwarf Virus-Gate(WDV-surrogate), to facilitate high-throughput genome editing.WDV-Gate has two parts: one is the recipient callus from a transgenic rice line expressing Cas9 and a mutated hygromycin-resistant gene(HygM) for surrogate selection;the other is a WDV-based construct expressing two single guide RNAs(sgRNAs) targeting HygM and a gene of interest, respectively. We evaluated WDV-Gate on six rice loci by producing a total of 874 T_0 plants. Compared with the conventional method, the WDV-Gate system, which was characterized by a transient and high level of sgRNA expression, significantly increased editing frequency(66.8% vs. 90.1%), plantlet regeneration efficiency(2.31-fold increase), and numbers of homozygous-edited plants(36.3%vs. 70.7%). Large-scale editing using pooled sg RNAs targeting the SLR1 gene resulted in a high editing frequency of 94.4%, further demonstrating its feasibility. We also tested WDVGate on sequence knock-in for protein tagging.By co-delivering a chemically modified donor DNA with the WDV-Gate plasmid, 3xFLAG peptides were successfully fused to three loci with an efficiency of up to 13%. | 5 | Adjoint-based robust optimization design of laminar flow wing under flight condition uncertainties显示文摘It is an inherent uncertainty problem that the application of laminar flow technology to the wing of large passenger aircraft is affected by flight conditions.In order to seek a more robust natural laminar flow control effect,it is necessary to develop an effective optimization design method.Meanwhile,attention must be given to the impact of crossflow(CF)instability brought on by the sweep angle.This paper constructs a robust optimization design framework based on discrete adjoint methods and non-intrusive polynomial chaos.Transition prediction is implemented by coupled Reynolds-Averaged Navier-Stokes(RANS)and simplified e^(N)method,which can consider both Tollmien-Schlichting(TS)wave and crossflow vortex instability.We have performed gradient enhancement processing on the general Polynomial Chaos Expansion(PCE),which is advantageous to reduce the computational cost of single uncertainty propagation.This processing takes advantage of the gradient information obtained by solving the coupled adjoint equations considering transition.The statistical moment gradient solution used for the robust optimization design also uses the derivatives of coupled adjoint equations.The framework is applied to the robust design of a 25°swept wing with infinite span in transonic flow.The uncertainty quantification and sensitivity analysis on the baseline wing shows that the uncertainty quantification method in this paper has high accuracy,and qualitatively reveals the factors that dominate in different flow field regions.By the robust optimization design,the mean and standard deviation of the drag coefficient can be reduced by 29%and 45%,respectively,and compared with the deterministic optimization design results,there is less possibility of forming shock waves under flight condition uncertainties.Robust optimization results illustrate the trade-off between the transition delay and the wave drag reduction. | Yifu CHEN Hanyue RAO Yiju DENG Tihao YANG Yayun SHI Junqiang BAI | 2023 | Chinese Journal of Aeronautics2023,36,6: | 0 |
| 6 | Modulation of Microstructure and Charge Transport in Polymer Monolayer Transistors by Solution Aging显示文摘A few monolayers of organic semiconductors adjacent to the dielectric layer are of vital importance in organic field-effect transistors due to their dominant role in charge transport.In this report,the 2-nm-thick polymer monolayers based on poly(3-hexylthiophene)with different molecular weights(M_(n))were fabricated using dip-coating technique.During the monolayer(solid state)formation from the solution,a disorder-to-order transition of polymer conformation is observed through UV-vis absorption measurement.Meanwhile,high Mn polymer monolayer generates higher crystalline fibrillar microstructure than the low Mn one due to the strongerπ–πintermolecular packing between polymers.More importantly,the solution aging procedure is utilized to further improve the morphology of polymer monolayers.It is obvious that after aging for 6 d,both fiber dimension and density as well as conjugation length are significantly increased under the same processing conditions in comparison to the fresh solution,and consequently the field-effect mobilities are remarkably enhanced by 2—4 times.Note that the maximum mobility of 0.027 cm2·V^(–1)·s^(–1)is among the highest reported values for poly(3-hexylthiophene)monolayer transistors.These results demonstrate a simple but powerful strategy for boosting the device performance of polymer monolayer transistors. | Xuemei Lin Ruochen Liu Chenming Ding Junyang Deng Yifu Guo Shibing Long Ling Li Mengmeng Li | 2021 | Chinese Journal of Chemistry2021,39,11: | 0 |
