1TNF-α suppresses sweat gland differentiation of MSCs by reducing FTO-mediated m^6 A-demethylation of Nanog mRNA显示文摘An effect of inhibition of tumor necrosis factor-α(TNF-α)on differentiation of mesenchymal stromal cells(MSCs)has been demonstrated,but the exact mechanisms that govern MSCs differentiation remain to be further elucidated.Here,we show that TNF-αinhibits the differentiation of MSCs to sweat glands in a specific sweat gland-inducing environment,accompanied with reduced expression of Nanog,a core pluripotency factor.We elucidated that fat mass and obesity-associated protein(FTO)-mediated m^6 A demethylation is involved in the regulation of MSCs differentiation potential.Exposure of MSCs to TNF-αreduced expression of FTO,which demethylated Nanog m RNA.Reduced expression of FTO increased Nanog m RNA methylation,decreased Nanog m RNA and protein expression,and significantly inhibited MSCs capacity for differentiation to sweat gland cells.Our finding is the first to elucidate the functional importance of m^6 A modification in MSCs,providing new insights that the microenvironment can regulate the multipotency of MSCs at the post-transcriptional level.Moreover,to maintain differentiation capacity of MSCs by regulating m^6 A modification suggested a novel potential therapeutic target for stem cellmediated regenerative medicine.Yihui Wang Rui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu 2020Science China(Life Sciences)2020,63,1:9
2Methods of computed tomography screening and management of lung cancer in Tianjin: design of a population-based cohort study显示文摘Objective: European lung cancer screening studies using computed tomography(CT) have shown that a management protocol based on measuring lung nodule volume and volume doubling time(VDT) is more specific for early lung cancer detection than a diameter-based protocol. However, whether this also applies to a Chinese population is unclear. The aim of this study is to compare the diagnostic performance of a volume-based protocol with a diameter-based protocol for lung cancer detection and optimize the nodule management criteria for a Chinese population.Methods: This study has a population-based, prospective cohort design and includes 4000 participants from the Hexi district of Tianjin, China. Participants will undergo low-dose chest CT at baseline and after 1 year. Initially, detected lung nodules will be evaluated for diameter and managed according to a routine diameter-based protocol(Clinical Practice Guideline in Oncology for Lung Cancer Screening, Version 2.2018). Subsequently, lung nodules will be evaluated for volume and management will be simulated according to a volume-based protocol and VDT(a European lung nodule management protocol). Participants will be followed up for 4 years to evaluate lung cancer incidence and mortality. 3Clinical characteristics and work-up of small to intermediate-sized pulmonary nodules in a Chinese dedicated cancer hospital显示文摘Objectives:To evaluate the characteristics and work-up of small to intermediate-sized pulmonary nodules in a Chinese dedicated cancer hospital.Methods:Patients with pulmonary nodules 4–25 mm in diameter detected via computed tomography(CT)in 2013 were consecutively included.The analysis was restricted to patients with a histological nodule diagnosis or a 2-year follow-up period without nodule growth confirming benign disease.Patient information was collected from hospital records.Results:Among the 314 nodules examined in 299 patients,212(67.5%)nodules in 206(68.9%)patients were malignant.Compared to benign nodules,malignant nodules were larger(18.0 mm vs.12.5 mm,P<0.001),more often partly solid(16.0%vs.4.7%,P<0.001)and more often spiculated(72.2%vs.41.2%,P<0.001),with higher density in contrast-enhanced CT(67.0 HU vs.57.5 HU,P=0.015).Final diagnosis was based on surgery in 232 out of 314(73.9%)nodules,166 of which were identified as malignant[30(18.1%)stage III or IV]and 66 as benign.In 36 nodules(11.5%),diagnosis was confirmed by biopsy and the remainder verified based on stability of nodule size at follow-up imaging(n=46,14.6%).Among 65 nodules subjected to gene(EGFR)mutation analyses,28(43.1%)cases(EGFR19 n=13;EGFR21 n=15)were identified as EGFR mutant and 37(56.9%)as EGFR wild-type.Prior to surgery,the majority of patients[n=194(83.6%)]received a contrast-enhanced CT scan for staging of both malignant[n=140(84.3%)]and benign[n=54(81.8%)]nodules.Usage of positron emission tomography(PET)-CT was relatively uncommon[n=38(16.4%)].Conclusions:CT-derived nodule assessment assists in diagnosis of small to intermediate-sized malignant pulmonary nodules.Currently,contrast-enhanced CT is commonly used as the sole diagnostic confirmation technique for pre-surgical staging,often resulting in surgery for late-stage disease and unnecessary surgery in cases of benign nodules.Xiaonan Cui Daiwei Han Marjolein AHeuvelmans Yihui Du Yingru Zhao Lei Zhang Harry JMGroen Geertruida Hde Bock Monique DDorrius Matthijs Oudkerk Rozemarijn Vliegenthart Zhaoxiang Ye 2020Cancer Biology & Medicine2020,17,1:5
4Redirecting differentiation of mammary progenitor cells by 3D bioprinted sweat gland microenvironment显示文摘Background:Mammary progenitor cells(MPCs)maintain their reproductive potency through life,and their specific microenvironments exert a deterministic control over these cells.MPCs provides one kind of ideal tools for studying engineered microenvironmental influence because of its accessibility and continually undergoes postnatal developmental changes.The aim of our study is to explore the critical role of the engineered sweat gland(SG)microenvironment in reprogramming MPCs into functional SG cells.Methods:We have utilized a three-dimensional(3D)SG microenvironment composed of gelatin-alginate hydrogels and components from mouse SG extracellular matrix(SG-ECM)proteins to reroute the differentiation of MPCs to study the functions of this microenvironment.MPCs were encapsulated into the artificial SG microenvironment and were printed into a 3D cell-laden construct.The expression of specific markers at the protein and gene levels was detected after cultured 14 days.Results:Compared with the control group,immunofluorescence and gene expression assay demonstrated that MPCs encapsulated in the bioprinted 3D-SG microenvironment could significantly express the functional marker of mouse SG,sodium/potassium channel protein ATP1a1,and tend to express the specific marker of luminal epithelial cells,keratin-8.When the Shh pathway is inhibited,the expression of SG-associated proteins in MPCs under the same induction environment is significantly reduced.Conclusions:Our evidence proved the ability of differentiated mouse MPCs to regenerate SG cells by engineered SG microenvironment in vitro and Shh pathway was found to be correlated with the changes in the differentiation.These results provide insights into regeneration of damaged SG by MPCs and the role of the engineered microenvironment in reprogramming cell fate.Rui Wang Yihui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu 2019Burns & Trauma2019,7,1:5

6Integrative Monsoon Frontal Rainfall Experiment(IMFRE-I):A Mid-Term Review显示文摘The mei-yu season,typically occurring from mid-June to mid-July,on average,contributes to 32%of the annual precipitation over the Yangtze-Huai River Valley(YHRV)and represents one of the three heavy-rainfall periods in China.Here,we briefly review the large-scale background,synoptic pattern,moisture transport,and cloud and precipitation characteristics of the mei-yu frontal systems in the context of the ongoing Integrative Monsoon Frontal Rainfall Experiment(IMFRE)field campaign.Phase one of the campaign,IMFRE-I,was conducted from 10 June to 10 July 2018 in the middle reaches of the YHRV.Led by the Wuhan Institute of Heavy Rain(IHR)with primary support from the National Natural Science Foundation of China,IMFRE-I maximizes the use of our observational capacity enabled by a suite of ground-based and remote sensing instruments,most notably the IHR Mesoscale Heavy Rainfall Observing System(MHROS),including different wavelengths of radars,microwave radiometers,and disdrometers.The KA350(Shanxi King-Air)aircraft participating in the campaign is equipped with Ka-band cloud radar and different probes.The comprehensive datasets from both the MHROS and aircraft instruments are combined with available satellite observations and model simulations to answer the three scientific questions of IMFRE-I.Some highlights from a previously published special issue are included in this review,and we also briefly introduce the IMFRE-II field campaign,conducted during June-July 2020,where the focus was on the spatiotemporal evolutions of the mei-yu frontal systems over the middle and lower reaches of the YHRV.Chunguang CUI Xiquan DONG Bin WANG Baike XI Yi DENG Yihui DING 2021Advances in Atmospheric Sciences2021,38,3:2

8Public integrity verification for data sharing in cloud with asynchronous revocation显示文摘Cloud data sharing service,which allows a group of people to access and modify the shared data,is one of the most popular and efficient working styles in enterprises.Recently,there is an uprising trend that enterprises tend to move their IT service from local to cloud to ease the management and reduce the cost.Under the new cloud environment,the cloud users require the data integrity verification to inspect the data service at the cloud side.Several recent studies have focused on this application scenario.In these studies,each user within a group is required to sign a data block created or modified by him.While a user is revoked,all the data previously signed by him should be resigned.In the existing research,the resigning process is dependent on the revoked user.However,cloud users are autonomous.They may exit the system at any time without notifying the system admin and even are revoked due to misbehaviors.As the developers in the cloud-based software development platform,they are voluntary and not strictly controlled by the system.Due to this feature,cloud users may not always follow the cloud service protocol.They may not participate in generating the resigning key and may even expose their secret keys after being revoked.If the signature is not resigned in time,the subsequent verification will be affected.And if the secret key is exposed,the shared data will be maliciously modified by the attacker who grasps the key.Therefore,forcing a revoked user to participate in the revocation process will lead to efficiency and security problems.As a result,designing a practical and efficient integrity verification scheme that supports this scenario is highly desirable.In this paper,we identify this challenging problem as the asynchronous revocation,in which the revocation operations(i.e.,re-signing key generation and resigning process)and the user's revocation are asynchronous.All the revocation operations must be able to be performed without the participation of the revoked user.Even more ambitiously,the revocation process should not rely on any special entity,such as the data owner or a trusted agency.To address this problem,we propose a novel public data integrity verification mechanism in which the data blocks signed by the revoked user will be resigned by another valid user.From the perspectives of security and practicality,the revoked user does not participate in the resigning process and the re-signing key generation.Our scheme allows anyone in the cloud computing system to act as the verifier to publicly and efficiently verify the integrity of the shared data using Homomorphic Verifiable Tags(HVTs).Moreover,the proposed scheme resists the collusion attack between the cloud server and the malicious revoked users.The numerical analysis and experimental results further validate the high efficiency and scalability of the proposed scheme.The experimental results manifest that re-signing 10,000 data blocks only takes 3.815 s and a user can finish the verification in 300 ms with a 99% error detection probability.Wei Song Yu Wu Yihui Cui Qilie Liu Yuan Shen Zicheng Qiu Jianjun Yao Zhiyong Peng 2022Digital Communications and Networks2022,8,1:0
9Tensile Properties and Microstructure of DS NiAl-28Cr-5.8Mo-0.2Hf Alloy显示文摘A multiphase alloy NiAI-28Cr-5.8Mo-0.2Hf was directionally solidified in Ar atmosphere in an Al2O3-SiC2 mold by standard Bridgman method. The fracture toughness and tensile properties at 980℃ as well as tensile creep at 1050℃ were studied. It was found that the strength of the present alloy is higher than that of many NiAI-based alloy and the stress exponent n for creep is about 6.69. 10Bias Analysis and Correction of Ground Surface Temperature Observations across China显示文摘Based on the ground surface temperature(GST)and snow surface temperature(SST)measurements during the period of adjustment from manual to automatic observation systems in China,the influence of observation methods on GST and its relationship with snow cover is analyzed.GST is corrected by SST,and the correction effect is evaluated.The results show that,during the parallel observation period,the winter GSTs from automatic observations are generally higher than those from manual observations,with the automatically observed national daily GST 1.18°C higher.The adjustment has a greater impact on GSTs at 0200,0800,and 2000 Beijing Time(BT)than at 1400 BT,and it has the greatest impact in Northeast and Northwest China,with deviations of 5.24 and 2.09°C,respectively.The GST deviation is closely related to the snow depth and annual snow totals.The average daily GST deviation increases at the rate of 0.66°C per 1-cm increase of snow depth when it is<15 cm,while it tends to be stable at around 10°C for snow depth over 15 cm.The GST deviation at a station is affected by its winter snow totals in Northeast and Northwest China,where the largest deviations are found where snow totals are all above 1000 cm.After the correction with SST,the mean deviation between the automatic and manual observations as well as the false trend can be effectively reduced.Following the correction,the mean deviation of daily GST decreases by 5.8°C,and its trend decreases from 1.87 to 0.65°C decade-1.Yan CUI Wenhui XU Zijiang ZHOU Chunyu ZHAO Yihui DING Xue AO Xiaoyu ZHOU 2020Journal of Meteorological Research2020,34,6:0
