| 1 | Fast-track program vs traditional care in surgery for gastric cancer显示文摘AIM:To systematically review the evidence for the effectiveness of fast-track program vs traditional care in laparoscopic or open surgery for gastric cancer.METHODS:PubMed,Embase and the Cochrane library databases were electronically searched for published studies between January 1995 and April 2013,and only randomized trials were included.The references of relevant studies were manually searched for further studies that may have been missed.Search terms included'gastric cancer','fast track'and'enhanced recovery'.Five outcome variables were considered most suitable for analysis:postoperative hospital stay,medical cost,duration to first flatus,C-reactive protein(CRP)level and complications.Postoperative hospital stay was calculated from the date of operation to the date of discharge.Fixed effects model was used for meta-analysis.RESULTS:Compared with traditional care,fasttrack program could significantly decrease the postoperative hospital stay[weighted mean difference(WMD)=-1.19,95%CI:-1.79--0.60,P=0.0001,fixed model],duration to first flatus(WMD=-6.82,95%CI:-11.51--2.13,P=0.004),medical costs(WMD=-2590,95%CI:-4054--1126,P=0.001),and the level of CRP(WMD=-17.78,95%CI:-32.22--3.35,P=0.0001)in laparoscopic surgery for gastric cancer.In open surgery for gastric cancer,fast-track program could also significantly decrease the postoperative hospital stay(WMD=-1.99,95%CI:-2.09--1.89,P=0.0001),duration to first flatus(WMD=-12.0,95%CI:-18.89--5.11,P=0.001),medical cost(WMD=-3674,95%CI:-5025--2323,P=0.0001),and the level of CRP(WMD=-27.34,95%CI:-35.42--19.26,P=0.0001).Furthermore,fast-track program did not significantly increase the incidence of complication(RR=1.39,95%CI:0.77-2.51,P=0.27,for laparoscopic surgery;and RR=1.52,95%CI:0.90-2.56,P=0.12,for open surgery).CONCLUSION:Our overall results suggested that compared with traditional care,fast-track program could result in shorter postoperative hospital stay,less medical costs,and lower level of CRP,with no more complications occurring in both laparoscopic and open surgery for gastric cancer. | Zhi-Xing Chen Ae-Huey Jennifer Liu Ying Cen | 2014 | World Journal of Gastroenterology2014,20,2: | 41 |

| 5 | BMSC-derived exosomes ameliorate sulfur mustard-induced acute lung injury by regulating the GPRC5A-YAP axis显示文摘Sulfur mustard(SM)is a highly toxic chemical warfare agent that causes acute lung injury(ALI)and/or acute respiratory distress syndrome(ARDS).There are no effective therapeutic treatments or antidotes available currently to counteract its toxic effects.Our previous study shows that bone marrow-derived mesenchymal stromal cells(BMSCs)could exert therapeutic effects against SM-induced lung injury.In this study,we explored the therapeutic potential of BMSC-derived exosomes(BMSC-Exs)against ALI and the underlying mechanisms.ALI was induced in mice by injection of SM(30 mg/kg,sc)at their medial and dorsal surfaces.BMSC-Exs(20μg/kg in 200μL PBS,iv)were injected for a 5-day period after SM exposure.We showed that BMSC-Exs administration caused a protective effect against pulmonary edema.Using a lung epithelial cell barrier model,BMSC-Exs(10,20,40μg)dose-dependently inhibited SM-induced cell apoptosis and promoted the recovery of epithelial barrier function by facilitating the expression and relocalization of junction proteins(E-cadherin,claudin-1,occludin,and ZO-1).We further demonstrated that BMSC-Exs protected against apoptosis and promoted the restoration of barrier function against SM through upregulating G protein-coupled receptor family C group 5 type A(GPRC5A),a retinoic acid target gene predominately expressed in the epithelial cells of the lung.Knockdown of GPRC5A reduced the antiapoptotic and barrier regeneration abilities of BMSC-Exs and diminished their therapeutic effects in vitro and in vivo.BMSC-Exs-caused upregulation of GPRC5A promoted the expression of Bcl-2 and junction proteins via regulating the YAP pathway.In summary,BMSC-Exs treatment exerts protective effects against SM-induced ALI by promoting alveolar epithelial barrier repair and may be an alternative approach to stem cell-based therapy. | Guan-chao Mao Chu-chu Gong Zhen Wang Ming-xue Sun Zhi-peng Pei Wen-qi Meng Jin-feng Cen Xiao-wen He Ying Lu Qing-qiang Xu Kai Xiao | 2021 | Acta Pharmacologica Sinica2021,42,12: | 4 |

| 10 | Facile scalable one-step wet-spinning of surgical sutures with shape memory function and antibacterial activity for wound healing显示文摘Surgical suture is commonly used in clinic due to its action in accelerating the process of wound healing.However,difficultly handling in minimally invasive surgery and bacteria-induced infection usually limit its use in a wide range of applications.Here,we report a facile scalable strategy to fabricate surgical sutures with shape memory function and antibacterial activity for wound healing.Specifically,a shape memory polyurethane(SMPU)with a transition temperature(Ttrans)at 41.3℃was synthesized by adjusting the mole ratio of the hard/soft segment,and then the shape memory surgical sutures containing polyhexamethylene biguanide hydrochloride(PHMB)as a model drug for antibacterial activity were fabricated by a facile scalable one-step wet-spinning approach,in which PHMB was directly dissolved in the coagulation bath that enable its loading into the sutures through the dual diffusion during the phase separation.The prepared sutures were characterized by their morphology,mechanical properties,shape memory,antibacterial activity,as well as biocompatibility before the wound healing capability was tested in a mouse skin suture-wound model.It was demonstrated that the optimized suture is capable of both shape memory function and antibacterial activity,and promote wound healing,suggesting that the facile scalable one-step wet-spinning strategy provides a promising tool to fabricate surgical sutures for wound healing. | Xinghan Chen Pengfei Tan Ya Wen Wencheng Zhou Ying Cen Chao You Lin Tan Meng Tian | 2020 | Chinese Chemical Letters2020,31,6: | 1 |
| 11 | mTOR regulates cocaine-induced behavioural sensitization through the SynDIG1–GluA2 interaction in the nucleus accumbens显示文摘Behavioral sensitization is a progressive increase in locomotor or stereotypic behaviours in response to drugs. It is believed to contribute to the reinforcing properties of drugs and to play an important role in relapse after cessation of drug abuse. However, the mechanism underlying this behaviour remains poorly understood. In this study, we showed that mTOR signaling was activated during the expression of behavioral sensitization to cocaine and that intraperitoneal or intra-nucleus accumbens (NAc) treatment with rapamycin, a specific mTOR inhibitor, attenuated cocaine-induced behavioural sensitization. Cocaine significantly modified brain lipid profiles in the NAc of cocaine-sensitized mice and markedly elevated the levels of phosphatidylinositol-4-monophosphates (PIPs), including PIP, PIP2, and PIP3. The behavioural effect of cocaine was attenuated by intra-NAc administration of LY294002, an AKT-specific inhibitor, suggesting that PIPs may contribute to mTOR activation in response to cocaine. An RNA-sequencing analysis of the downstream effectors of mTOR signalling revealed that cocaine significantly decreased the expression of SynDIG1, a known substrate of mTOR signalling, and decreased the surface expression of GluA2. In contrast, AAV-mediated SynDIG1 overexpression in NAc attenuated intracellular GluA2 internalization by promoting the SynDIG1–GluA2 interaction, thus maintaining GluA2 surface expression and repressing cocaine-induced behaviours. | 16 | Study of the decorporation efficacy and toxicity of tetradentate 3-hydroxy- 2-pyridinone ligands at the cellular level显示文摘Objective:A series of experiments were carried out to study the comparative decorporation efficacy and toxicity of three tetradentate hydroxypyridinone ligands(3LI-1-Cm-3,2-HOPO,5LIO-1-Cm-3,2-HOPO,and 7LIO-1-Cm-3,2-HOPO)with the variation of the linker length and the substituents.Methods:Three ligands were obtained from a 4-step synthesis via the amidation of HOPO unit and different backbones.Potentiometric titrations were carried out to evaluate the formation constants of their corresponding uranyl complexes.CCK-8 and colony formation assays were used to compare the cytotoxicity of the three ligands and uranyl using renal proximal tubular epithelial(NRK-52E)cells.In vitro uranium removal assays were performed to assess decorporation efficiencies of those ligands by co-incubating uranium and chelators with NRK-52E cells for 48 h.Results:Among the three ligands,5LIO-1-Cm-3,2-HOPO exhibits the highest uranyl affinity(logβ110?18.6(7))in comparison to the other two ligands(logβ110?14.9(2)for 3LI-1-Cm-3,2-HOPO and logβ110?16.7(1)for 7LIO-1-Cm-3,2-HOPO).The results of CCK-8 tests and colony formation assays further elucidate that 5LIO-1-Cm-3,2-HOPO and 7LIO-1-Cm-3,2-HOPO both show a similar level of cytotoxicity compared with ZnNa3-DTPA at relatively low ligand concentrations.In contrast,in the 3LI-1-Cm-3,2-HOPO treated group,the cell viability decreased markedly with the increase of the ligand concentrations,and the colony formation capability of NRK-52E cells was inhibited.Furthermore,the comparative decorporation efficacy of the three ligands was obtained from in vitro uranium removal assays,suggesting that these ligands could significantly inhibit the cellular uptake and prompt the cellular release of uranium from NRK-52E cells by the level of 81.9%,91.8%,and 87.1%,respectively.Conclusion:Our current results demonstrate that the linker/backbone species would significantly affect the decorporation efficacy and toxicity of the tetradentate 3-hydroxy-2-pyridinone ligands,which is informative for the future design and modification of novel decorporation ligands. | Xiaomei Wang Cen Shi Manni Gao Ying Xu Yang Jiao Jianmei Wan Jianping Cao Zhifang Chai Juan Diwu | 2020 | Radiation Medicine and Protection2020,1,4: | 1 |

Antiviral mechanism of two samples was briefly studied by four different ways. The cytotoxicity of samples on vero cells was not detected (CC>5000 μg/mL). All samples except PSJ had obviously HSV-1 inhibitory effect. Antiviral activities of them were increasingly strengthened with raising their purities.Antiviral activities of F1, F2 and F4 were better than that of ACV, and antiviral activities of all samples on CVB3 were better than ribavirin injection. Fi, F2 and F4 had remarkable anti-CVB3 activity.
