| 1 | Expression,purification and characterization of Mycobacterium tuberculosis RpfE protein显示文摘Resuscitation promoting factor E(RpfE)is one of the five Rpf-like proteins in Mycobacterium tuberculosis(M.tuberculosis).These Rpf-like proteins are secretory,which make them candidates for recognition by the host immune system.In this study,the RpfE gene was amplified from M.tuberculosis,cloned into the expression vectors pDE22 and pPRO EXHT,and were expressed in Mycobacterium vaccae(M.vaccae)and Escherichia coli DH5α,respectively.Both recombinant RpfE proteins were purified by Ni-Sepharose affinity chromatography,and were given to C57BL/6 mice.The RpfE proteins elicited T cell proliferation,and stimulated the production of gamma interferon(IFN-γ),interleukin-10(IL-10)and IL-12.Our results indicated that the RpfE protein expressed in M.vaccae could more efficiently stimulate cellular immune response,making it a promising candidate as a subunit vaccine. | Ying Xue Yinlan Bai Xue Gao Hong Jiang Limei Wang Hui Gao Zhikai Xu | 2012 | The Journal of Biomedical Research2012,26,1: | 2 |
| 2 | Validation of different personalized risk models of chemotherapy-induced nausea and vomiting:results of a randomized,double-blind,phase III trial of fosaprepitant for cancer patients treated with high-dose cisplatin显示文摘Background:Highly emetogenic chemotherapy induces emesis in cancer patients without prophylaxis.The purpose of this study was to evaluate the efficacy and safety of a fosaprepitant-based triple antiemetic regimen for the prevention of chemotherapy-induced nausea and vomiting(CINV)in patients with solid malignant tumors,determine risk factors and externally validate different personalized risk models for CINV.Methods:This phase III trial was designed to test the non-inferiority of fosaprepitant toward aprepitant in cancer patients who were to receive the first cycle of single-day cisplatin chemotherapy.The primary endpoint was complete response(CR)during the overall phase(OP)with a non-inferiority margin of 10.0%.Logistic regression modelswere used to assess the risk factors ofCRand no nausea.To validate the personalized risk models,the accuracy of the risk scoring systems was determined by measuring the specificity,sensitivity and area under the receiver operating characteristic(ROC)curve(AUC),while the predictive accuracy of the nomogram was measured using concordance index(C-index).Results:A total of 720 patients were randomly assigned.CR during the OP in the fosaprepitant group was not inferior to that in the aprepitant group(78.1%vs.77.7%,P=0.765)with a between-group difference of 0.4%(95%CI,-5.7%to 6.6%).Female sex,higher cisplatin dose(≥70 mg/m2),no history of drinking and larger body surface area(BSA)were significantly associated with nausea.The AUC for the acute and delayed CINV risk indexes was 0.68(95%CI:0.66-0.71)and 0.66(95%CI:0.61-0.70),respectively,and the C-index for nomogram CINV prediction was 0.59(95%CI,0.54-0.64).Using appropriate cutoff points,the three models could stratify patients with high-or low-risk CINV.No nausea and CR rate were significantly higher in the low-risk group than in the high-risk group(P<0.001).Conclusions:Fosaprepitant-based triple prophylaxis demonstrated non-inferior control for preventing CINV in patients treated with cisplatin-base chemotherapy.Female cancer patients without a history of alcohol consumption,with larger BSA and received high-dose cisplatin might be more vulnerable to CINV.Three personalized prediction models were well-validated and could be used to optimize antiemetic therapy for individual patients. | Yuanyuan Zhao Bing Zhao Gang Chen Yinlan Chen Zijun Liao Haiming Zhang Weineng Feng Yinyin Li Heng Weng Weidong Li Yuefen Zhou Biyong Ren Yanda Lu Jianhua Chen Zhenteng Liu Zhenzhong Su Wenliang Wang Li Zhang | 2023 | Cancer Communications2023,43,2: | 2 |

| 11 | Puerarin inhibits NHE1 activity by interfering with the p38 pathway and attenuates mitochondrial damage induced by myocardial calcium overload in heart failure rats显示文摘Previous studies have shown that puerarin plays a key role in protecting humans and animals from cardiovascular diseases.The exact mechanism of the therapeutic effect of puerarin on various cardiovascular diseases(protective effect on cardiomyocytes)is still unclear.In the present study,we identify the role of puerarin in an animal model of experimental heart failure(HF)and explore its underlying mechanisms.The HF rat model is induced by in-traperitoneal injection of adriamycin(ADR),and puerarin is administered intragastrically at low,medium,and high concentrations.We demonstrate that puerarin significantly improves myocardial fibrosis and inflammatory in-filtration and,as a result,improves cardiac function in ADR-induced HF rats.Mechanistically,we find for the first time that puerarin inhibits overactivated Na+/H+exchange isoform 1(NHE1)in HF,which may improve HF by decreasing Na+and Ca2+ion concentrations and attenuating mitochondrial damage caused by calcium overload;on the other hand,puerarin inhibits the activation of the p38 pathway in HF,reduces the expressions of TGF-βand proinflammatory cytokines,and suppresses myocardial fibrosis.In conclusion,our results suggest that Puerarin is an effective drug against HF and may play a protective role in the myocardium by inhibiting the activation of p38 and its downstream NHE1. | Guopin Pan Baoyue Cui Mingming Han Laibiao Lin Yinlan Li Ling Wang Shuang Guo Yaling Yin Heqin Zhan Peng Li | 2024 | Acta Biochimica et Biophysica Sinica2024,56,2: | 0 |

| 14 | Dynamic changes in butyrate levels regulate satellite cell homeostasis by preventing spontaneous activation during aging显示文摘The gut microbiota plays a pivotal role in systemic metabolic processes and in particular functions,such as developing and preserving the skeletal muscle system.However,the interplay between gut microbiota/metabolites and the regulation of satellite cell(SC)homeostasis,particularly during aging,remains elusive.We propose that gut microbiota and its metabolites modulate SC physiology and homeostasis throughout skeletal muscle development,regeneration,and aging process.Our investigation reveals that microbial dysbiosis manipulated by either antibiotic treatment or fecal microbiota transplantation from aged to adult mice,leads to the activation of SCs or a significant reduction in the total number.Furthermore,employing multi-omics(e.g.,RNA-seq,16S r RNA gene sequencing,and metabolomics)and bioinformatic analysis,we demonstrate that the reduced butyrate levels,alongside the gut microbial dysbiosis,could be the primary factor contributing to the reduction in the number of SCs and subsequent impairments during skeletal muscle aging.Meanwhile,butyrate supplementation can mitigate the antibiotics-induced SC activation irrespective of gut microbiota,potentially by inhibiting the proliferation and differentiation of SCs/myoblasts.The butyrate effect is likely facilitated through the monocarboxylate transporter 1(Mct1),a lactate transporter enriched on membranes of SCs and myoblasts.As a result,butyrate could serve as an alternative strategy to enhance SC homeostasis and function during skeletal muscle aging.Our findings shed light on the potential application of microbial metabolites in maintaining SC homeostasis and preventing skeletal muscle aging. | Shujie Chen Liujing Huang Bingdong Liu Huimin Duan Ze Li Yifan Liu Hu Li Xiang Fu Jingchao Lin Yinlan Xu Li Liu Dan Wan Yulong Yin Liwei Xie | 2024 | Science China(Life Sciences)2024,67,4: | 0 |
