| 1 | Hypocrellin A-based photodynamic action induces apoptosis in A549 cells through ROS-mediated mitochondrial signaling pathway显示文摘Over recent decades, many studies have reported that hypocrellin A(HA) can eliminate cancer cells with proper irradiation in several cancer cell lines. However, the precise molecular mechanism underlying its anticancer effect has not been fully defined. HA-mediated cytotoxicity and apoptosis in human lung adenocarcinoma A549 cells were evaluated after photodynamic therapy(PDT). A temporal quantitative proteomics approach by isobaric tag for relative and absolute quantitation(iTRAQ) 2 D liquid chromatography with tandem mass spectrometric(LC–MS/MS) was introduced to help clarify molecular cytotoxic mechanisms and identify candidate targets of HA-induced apoptotic cell death. Specific caspaseinhibitors were used to further elucidate the molecular pathway underlying apoptosis in PDT-treated A549 cells. Finally, down-stream apoptosis-related protein was evaluated. Apoptosis induced by HA was associated with cell shrinkage, externalization of cell membrane phosphatidylserine, DNA fragmentation,and mitochondrial disruption, which were preceded by increased intracellular reactive oxygen species(ROS) generations. | 2 | The responses of East Asian Summer monsoon to the North Atlantic Meridional Overturning Circulation in an enhanced freshwater input simulation显示文摘We investigated the response of the East Asian Summer Monsoon (EASM) to a weakened Atlantic Meridional Overturning Circulation (AMOC) and its mechanism in an enhanced freshwater input experiment (FW) by using a fully-coupled climate model. The response was a weakened EASM and the mechanisms can be explained as follows. The simulated weakened AMOC resulted in a drop in sea surface temperature (SST) in the North Atlantic (NA) and, correspondingly, an anomalous high sea level pressure (SLP) over the North American regions, which in turn increased the northeast surface winds across the equator in the eastern tropical Pacific (ETP). The anomalous northeast winds then induced further upwelling in the ETP and stronger air/sea heat exchange, therefore leading to an anomalous cooling of the eastern tropical sea surface. As a result, the climatologic Hadley Circulation (HC) was weakened due to an anomalous stronger sinking of air in the ETP north of the equator, whereas the Walker Circulation (WC) in the western tropical Pacific (WTP) north of the equator was strengthened with an eastward-shifted upwelling branch. This feature was in agreement with the anomalous convergent winds in the WTP, and led to a weakened EASM and less East Asian summer precipitation (EASP). | 3 | Physiological and ultralstructural changes of Chlorella sp. induced by UV-B radiation显示文摘In order to investigate the mechanisms of enhanced UV-B radiation on algae, the effects of UV-B radiation on the physiological and ultrastructural changes of Chlorella sp. were examined. The results showed that UV-B radiation could inhibit the growth and photosynthesis of microalgae. UV-B radiation at lower doses increased the photosynthetic pigment (chlorophyll a (Chla) and carotenoid (Car)) contents, while at higher doses of UV-B radiation Chla and Car contents were decreased. The ultrastructure of Chlorella sp. without exposure to UV-B showed that the thylakoid lamellae were clear and regular, the stroma of its chloroplast was apparent and clear. The globules with photosynthetic pigments and the cristae of mitochondria were clearly seen. After exposure to UV-B radiation at dose of 2.88 kJ/m2, the thylakoid lamellae of Chlorella sp. were lost and dissolved, the globules which contained photosynthetic pigments in chloroplast were bleached; some mitochondria cristae were dissolved; slight plasmolysis was found in some Chlorella sp. cells. After exposure to 5.76 kJ/m2 UV-B radiation, the thylakoid was in disarray and disintegration, plasmolysis was found in most cells, and the cell wall was broken and began to fall out. Many blank areas were observed in cells, mitochondria were seriously deformed and most of the mitochondria cristae were dissolved. Also, globules containing photosynthetic pigments in chloroplast were bleached and some empty globules were found in chloroplast. | 7 | Effect of Co-Culture of Chinese Shrimp(Fenneropenaeus chinensis) and Sea Cucumber(Apostichopus japonicus Selenka) on Pond Environment显示文摘Monoculture of sea cucumber(pond S) and polyculture of shrimp with sea cucumber(pond SS) were established to evaluate the effect of shrimp on the environmental conditions of sea cucumber farming pond. Contributions of sediment organic matter(SOM2) resuspended from benthic sediment and the suspended particulate organic matter(SPOM) deposited from the water column to the precipitated organic matter(SOM1) collected with sediment traps were estimated with carbon stable isotope analysis. The results showed that the levels of SPOM and SOM2 in pond SS significantly decreased in comparison with those in pond S at the end of experiment(P < 0.05), indicating that co-culturing shrimp in sea cucumber farming pond could purify the farming water. Carbon stable isotope analysis showed that the proportion of SOM2 in SOM1 in pond SS(84.97% ± 0.38%) was significantly lower than that in pond S(95.20% ± 0.30%)(P < 0.05), suggesting that the resuspension of organic matter from benthic sediment into overlying water was reduced in polyculture pond. | 10 | The effects of enriched C0_(2) and enhanced UV-B radiation on ultrastructure of Dunaliella salina, singly and in combination显示文摘The effects of ambient CO2/ambient UV- B, enriched CO2/ambient UV- B, ambient CO2/enhanced UV- B, and enriched CO2/ enhanced UV- B on the ultrastructure of Dunaliella salina were investigated. (1) The ultrastructure of D. salina cell in the control experiment showed that the arrangement of thylakoid lamellae was regular, and there were many large starch grains among the thylakoid lamellae. A prominent well-developed pyrenoid was found in the middle of the chloroplast. Nucleus envelope and nucleolus were clearly ob- served. The Golgi apparatus accompanied by numerous vesicles with a compact arrangement of cisternae and the peripheral tips of the cisternae were swollen to a size comparable to that of some of the associated vesicles. (2) The ultrastructure of D. salina cell in en- riched CO2 showed that the arrangement of thylakoid was regular and the lamellae were vivid. Developed pyrenoids were found in the low-CO2-grown cells, but not in the high-CO2-grown cells. The mitochondria cristae were vivid. The arrangement of Golgi apparatus was compact. (3) The ultrastructure of D. salina cell in enhanced UV- B showed that the thylakoid was dissolved and the cells had a less developed pyrenoid or no detectable pyrenoid. Part of the nucleus envelope was dissolved. The number of mitochondria was in- creased and some mitochondria cristae were disintegrated. The starch grains were broken apart into many small starch grains. The Gol- gi apparatus with a loose arrangement of cisternae and the peripheral tips of the Golgi cisternae were not especially swollen, with sev- eral large associated vesicles. (4) The ultrastructure of D. salina cell in the enriched CO2/enhanced UV- B showed that part of the thy- lakoid and nucleus envelopes of some cells were dissolved. The pyrenoid was larger than that of the enhanced UV- B. There were many mitochondria between stroma and chloroplast membrane, but mitochondria cristae were partly dissolved. Many small starch grains were accumulated in cells. The starch sheath was broken into several discontinuous starch grains with different sizes. The ar- rangement of Golgi apparatus was loose. | 12 | Microwear analysis of a bone-handled microblade excavated at Shangzhai site in Beijing: Evidences from ESEM显示文摘The conventional wisdom has been challenged recently that microblades with handles are only tools for hunting and gathering in the Neolithic Age. Some scholars argued that microblades actually played an important part in the early agricultural development in the Neolithic. However, both of these opinions lacked direct evidence. Environmental scanning electron microscopy (ESEM) is used to analyze the residues of the bone-handled microblade excavated at the Shangzhai site in Pinggu, Beijing. The ESEM observation and EDX analysis show that at the edge of this bone-handled microblade are Ca, P, C, O or C, N, O, etc, which provide direct evidence for determining its function, namely that this microblade may have been dealt with animals. | 14 | Change of Digestive Physiology in Sea Cucumber Apostichopus japonicus(Selenka) Induced by Corn Kernels Meal and Soybean Meal in Diets显示文摘The present study was conducted to determine the change of digestive physiology in sea cucumber Apostichopus japonicus(Selenka) induced by corn kernels meal and soybean meal in diets. Four experimental diets were tested, in which Sargassum thunbergii was proportionally replaced by the mixture of corn kernels meal and soybean meal. The growth performance, body composition and intestinal digestive enzyme activities in A. japonicus fed these 4 diets were examined. Results showed that the sea cucumber exhibited the maximum growth rate when 20% of S. thunbergii in the diet was replaced by corn kernels meal and soybean meal, while 40% of S. thunbergii in the diet can be replaced by the mixture of corn kernels meal and soybean meal without adversely affecting growth performance of A. japonicus. The activities of intestinal trypsin and amylase in A. japonicus can be significantly altered by corn kernels meal and soybean meal in diets. Trypsin activity in the intestine of A. japonicus significantly increased in the treatment groups compared to the control, suggesting that the supplement of corn kernels meal and soybean meal in the diets might increase the intestinal trypsin activity of A. japonicus.
