| 1 | HIF-1α-induced expression of m6A reader YTHDF1 drives hypoxia-induced autophagy and malignancy of hepatocellular carcinoma by promoting ATG2A and ATG14 translation显示文摘N6-methyladenosine(m6A),and its reader protein YTHDF1,play a pivotal role in human tumorigenesis by affecting nearly everystage of RNA metabolism.Autophagy activation is one of the ways by which cancer cells survive hypoxia.However,the possibleinvolvement of m6A modification of mRNA in hypoxia-induced autophagy was unexplored in human hepatocellular carcinoma(HCO).In this study,specific variations in YTHDF1 expression were detected in YTHDF1-overexpressing,knockout,and-knockdownHCC cells,HCC organoids,and HCC patient-derived xenograft(PDX)murine models.YTHDF1 expression and hypoxia inducedautophagy were significantly correlated in vitro;signifhcant overexpression of YTHDF1 in HCC tissues was associated with poorprognosis,Multivariate cox regression analysis identihed YTHDF1 expression as an independent prognostic factor in patients withHCC.Multiple HC models conhrmed that YTHDF1 deficiency inhibited HCC autophagy,growth,and metastasis.Luciferase reporterassays and chromatin immunoprecipitation demonstrated that HlIF-1a regulated YTHDF1 transcription by directly binding to itspromoter region under hypoxia.The results of methylated RNA immunoprecipitation sequencing,proteomics,and polysomeprofling indicated that YTHDF1 contibuted to the translation of autophagy-related genes ATG2A and ATG14 by binding to m6A-modifhed ATG2A and ATG14 mRNA,thus facilitating autophagy and autophagy-related malignancy of HCC.Taken together,HlE-1d-induced YTHDF1 expression was associated with hypoxia-induced autophagy and autophagy-related HCC progression via promoting translation of autophagy-related genes ATG2A and ATG14 in a m6A-dependent manner.Our fndings suggest thatYTHDF1 is a potential prognostic biomarker and therapeutic target for patients with HCC. | Qing Li Yong Ni Liren Zhang Runqiu Jiang Jing Xu Hong Yang Yuanchang Hu Jiannan Qiu Liyong Pu Jinhai Tang Xuehao Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,3: | 24 |
| 2 | Characteristics of global cerebral ischemia models constructed by modified four-vessel occlusion in rats显示文摘BACKGROUND: Pulsinelli et al developed a kind of rat models with four-vessel occlusion-induced global cerebral ischemia. Because the histo-pathological changes and severe cerebral ischemia reproducibility of this model are good and the stability of this model in circulation respiration is superior to that of other models, so four-vessel occlusion method has become a classic modeling method for global cerebral ischemia model. This model has been improved in some laboratories to meet different requirements in different studies. OBJECTIVE: To establish a highly reproducible rat model of reversible forebrain ischemia by modifying four-vessel occlusion model introduced by Pulsinelli et al, and to investigate its neurophysiological and pathological changes and the characteristics of modified operation. DESIGN:Completely randomized grouping, controlled trial. SETTING:Department of Anesthesiology,Changhai Hospital, Second Military Medical University of Chinese PLA. MATERIALS: A total of 65 male healthy SD rats, weighing 250-300 g, were provided by the Experimental Animal Center of Second Military Medical University of Chinese PLA. VSM hemodynamic monitor and temperature monitor (Thermal ert TH-5,U.S.A) were used. METHODS: The trial was conducted in the Department of Anesthesiology, Changhai Hospital, Second Military Medical University of Chinese PLA from January 2005 to March 2006. ① Experimental grouping: Sixty-five rats were randomly divided into the following 7 groups: sham-operation group (n =9): given the same operation, without occlusion of vessels; 5 minutes ischemia group (n =9): ischemia 5 minutes and reperfusion 72 hours; 10 minutes ischemia group (n =8): ischemia 10 minutes and reperfusion 72 hours; 15 minutes ischemia group (n =9): ischemia 15 minutes and reperfusion 72 hours; 20 minutes ischemia group (n =8): ischemia 20 minutes and reperfusion 72 hours; 30 minutes ischemia group (n =7); ischemia 30 minutes and reperfusion 72 hours; ischemia control group (n =15): ischemia 15 minutes and reperfusion 180 minutes. ② Preparation of the model of global cerebral ischemia: Four-vessel occlusion- induced global cerebral ischemia rat models were modified, i.e. bilateral vertebral arteries could be electrocauterized and blocked, and bilateral common carotid arteries were enclosed with 10-0 suture loosely. On the second day, keeping the animal awake, the suture was tightened and kept tense to block the blood flow of bilateral common carotid artery. After certain duration of ischemia, the suture was cut off and drawn out, thus the reperfusion of bilateral common carotid artery was resumed. ③Observation of physiological indexes: VSM hemodynamic monitor and temperature monitor (Thermal ert TH-5,U.S.A) were used to record and measure the changes of blood pressure, rectal temperature, brain temperature ,arterial blood gas and other physiological indexes of the rats in the control group before, 5 and 15 minutes after ischemia and 10,30,60,120 and 180 minutes after reperfusion. ④Preparation of brain tissue pathological samples: Except for ischemia control group, rats of other groups were anesthetized and their brain tissues were harvested and subjected to haematoxylin and eosin staining at 72 hours after reperfusion. ⑤Evaluation of pathological change of brain tissue: The coronal plane of anterior commissure of cerebrum was used to evaluate corpora striatum, and the coronal plane of anterior hippocampus was used to evaluate hippocampal CA1/2 region, CA3 region and CA4 region, subiculum, superior pyramidal lobe and inferior pyramidal lobe of dentate gyrus as well as neocortex. Irreversible neuronal damage included pyknotic cells with eosinophilic cytoplasm and trachychromatic nucleus, homogenous cytoplasm and naked nucleus. Neurons without the above changes were considered to be normal. The number of normal neurons in the above-mentioned brain regions was counted under the microscope. MAIN OUTCOME MEASURES:① The changes of blood pressure, rectal temperature, brain temperature, arterial blood gas and other physiological indexes of rats before, 5 and 15 minutes after ischemia and 10,30,60,120 and 182 minutes after reperfusion. ②The number of normal neurons in hippocampal CA1/2 region, CA3 region, CA4 region, subiculum, superior pyramidal lobe and inferior pyramidal lobe of dentate gyrus as well as neocortex at 72 hours of ischemia. RESULTS: Sixty-five SD rats were involved in the result analysis. ①Changes in the physiological indexes of rats during cerebral ischemia/reperfusion: Changes in arterial blood pressure: arterial blood pressure at 5 minutes after ischemia was significantly higher than that before ischemia [(132.82±12.38) mm Hg vs. (118.58±5.85) mm Hg,1 mm Hg=0.133 kPa, P < 0.01], and there was no significant difference in arterial blood pressure between at ischemia 15 minutes, reperfusion 10,30,60,120 and 180 minutes and before ischemia (P > 0.05); Changes in arterial blood gas: pH value at 15 minutes after ischemia was significantly higher than that before ischemia (7.56±0.05 vs. 7.47±0.09,P < 0.01), PaO2 value at 15 minutes after ischemia was significantly higher than that beischemia [(1.71±0.04) kPa vs. (1.56±0.08) kPa, P < 0.01], and PaCO2 value at 5 and 15 minutes after ischemia was significantly lower than that before ischemia [(0.44±0.04),(0.37±0.04),(0.57±0.04) kPa,P < 0.01];Changes in rectal temperature and brain temperature: Brain temperature at 5 and 10 minutes after ischemia and at 10 and 30 minutes after reperfusion was significantly lower than that before ischemia [(37.01±0.43),(36.44±0.35),(36.81±0.45),(37.23±0.24),(37.74±0.28) ℃,P < 0.01], and rectal temperature at 10 minutes after reperfusion was significantly lower than that before ischemia [(36.76±0.21),(37.37±0.19) ℃,P < 0.01]. ② Changes in the normal neurons in different encephalic regions of rats at different time after ischemia and 72 hours after reperfusion: At 5,10,15,20 and 30 minutes after ischemia, the number of normal neurons in subiculum was 86%,73%,11%,8%,6% of that in the sham-operation, respectively (P < 0.05-0.01), the number of normal neurons in internal segment of hippocampal CA1/2 region was 74%,53%,8%,5%,5% of that in the sham-operation group (P < 0.01),the number of normal neurons in middle segment of hippocampal CA1/2 was 73%,48%,11%,8%,6% of that in the sham-operation group, respectively (P < 0.01),and the number of normal neurons in external segment of hippocampal CA1/2 region was 90%,72%,37%,31%,22% of that in the sham-operation group, respectively (P < 0.01); At 10,15,20 and 30 minutes after ischemia, the number of normal neurons in hippocampal CA3 region was 80%,67%,47%,40% of that in the sham-operation group respectively (P < 0.01), the number of normal neurons in hippocampal CA4 region was 79%,70%,60%,43% of that in the sham-operation group , respectively (P < 0.01) and the number of normal neurons in superior pyramidal lobe of dentate gyrus was 85%,75%,59%,52% of that in the sham-operation group, respectively (P < 0.05,0.01). At 15,20 and 30 minutes after ischemia, the number of normal neurons in inferior pyramidal lobe of dentate gyrus was 80%,70%,59% of that in the sham-operation group (P < 0.01) and the number of normal neurons in the neocortex was 79%,72%,61% of that in the sham-operation group (P < 0.05,0.01). | 3 | Genetic analysis and fine mapping of an enclosed panicle mutant esp2 in rice (Oryza sativa L.)显示文摘The phenomenon of panicle enclosure in rice is mainly caused by the shortening of uppermost internode.Elucidating the molecular mechanism of panicle enclosure will be helpful for solving the problem of panicle enclosure in male sterile lines and creating new germplasms in rice.We acquired a monogenic recessive enclosed panicle mutant,named as esp2 (enclosed shorter panicle 2),from the tissue culture progeny of indica rice cultivar Minghui-86.In the mutant,panicles were entirely enclosed by flag leaf sheaths and the uppermost internode was almost completely degenerated,but the other internodes did not have obvious changes in length.Genetic analysis indicated that the mutant phenotype was controlled by a recessive gene,which could be steadily inherited and was not affected by genetic background.Apparently,ESP2 is a key gene for the development of uppermost internode in rice.Using an F 2 population of a cross between esp2 and a japonica rice cultivar Xiushui-13 as well as SSR and InDel markers,we fine mapped ESP2 to a 14-kb region on the end of the short arm of chromosome 1.According to the rice genome sequence annotation,only one intact gene exists in this region,namely,a putative phosphatidylserine synthase gene.Sequencing analysis on the mutant and the wild type indicated that this gene was inserted by a 5287-bp retrotransposon sequence.Hence,we took this gene as a candidate of ESP2.The results of this study will facilitate the cloning and functional analysis of ESP2 gene. | GUAN HuaZhong DUAN YuanLin LIU HuaQing CHEN ZhiWei ZHUO Ming ZHUANG LiJun QI WenMing PAN RunSen MAO DaMei ZHOU YuanChang WANG Feng WU WeiRen | 2011 | Chinese Science Bulletin2011,56,14: | 6 |

| 5 | Silvicultural alternatives to conventional even-aged forest man-agement-what limits global adoption?显示文摘Background: The development of forestry as a scientific and management discipline over the last two centuries has mainly emphasized intensive management operations focused on increased commodity production, mostly wood. This 'conventional' forest management approach has typically favored production of even-aged, single-species stands. While alternative management regimes have generally received less attention, this has been changing over the last three decades, especially in countries with developed economies. Reasons for this change include a combination of new information and concerns about the ecological consequences of intensive forestry practices and a willingness on the part of many forest owners and society to embrace a wider set of management objectives. Alternative silvicultural approaches are characterized by a set of fundamental principles, including avoidance of clearcutting, an emphasis on structural diversity and small-scale variability, deployment of mixed species with natural regeneration,and avoidance of intensive site-preparation methods.Methods: Our compilation of the authors' experiences and perspectives from various parts of the world aims to initiate a larger discussion concerning the constraints to and the potential of adopting alternative silvicultural practices.Results: The results suggest that a wider adoption of alternative silvicultural practices is currently hindered by a suite of ecological, economic, logistical, informational, cultural, and historical constraints. Individual contexts display their own unique combinations and relative significance of these constraints, and accordingly, targeted efforts, such as regulations and incentives, may help to overcome specific challenges.Conclusions: In a broader context, we propose that less emphases on strict applications of principles and on stand structures might provide additional flexibility and facilitate the adoption of alternative silvicultural regimes in a broader set of circumstances. At the same time, the acceptance of alternative silvicultural systems as the 'preferred or default mode of management' wil necessitate and benefit from the continued development of the scientific basis and valuation of a variety of ecosystem goods and services. | 8 | Terminal sulfur atoms formation via defect engineering strategy to promote the conversion of lithium polysulfides显示文摘The defect engineering shows great potential in boosting the conversion of lithium polysulfides intermediates for high energy density lithium-sulfur batteries(LSBs),yet the catalytic mechanisms remain unclear.Herein,the oxygen-defective Li_(4)Ti_(5)O_(12)-xhollow microspheres uniformly encapsulated by N-doped carbon layer(OD-LTO@NC)is delicately designed as an intrinsically polar inorganic sulfur host for the research on the catalytic mechanism.Theoretical simulations have demonstrated that the existence of oxygen deficiencies enhances the adsorption capability of spinel Li_(4)Ti_(5)O_(12)towards soluble lithium polysulfides.Some-S-S-bonds of the Li2S6on the defective Li_(4)Ti_(5)O_(12)surface are fractured by the strong adsorption force,which allows the inert bridging sulfur atoms to be converted into the susceptible terminal sulfur atoms,and reduces the activation energy of the polysulfide conversion in some degree.In addition,with the N-doped carbon layer,secondary hollow microspheres architecture built with primary ultrathin nanosheets provide a large amount of void space and active sites for sulfur storage,adsorption and conversion.The as-designed sulfur host exhibits a remarkable rate capability of 547 m Ah g^(-1)at 4C(1 C=1675 m A g^(-1))and an outstanding long-term cyclability(519 m Ah g^(-1)after 1000 cycles at 3 C).Besides,a high specific capacity of 832 m Ah g^(-1)is delivered even after 100 cycles under a high sulfur mass loading of 3.2 mg cm^(-2),indicating its superior electrochemical performances.This work not only provides a strong proof for the application of oxygen defect in the adsorption and catalytic conversion of lithium polysulfides,but offers a promising avenue to achieve high performance LSBs with the material design concept of incorporating oxygen-deficient spinel structure with hierarchical hollow frameworks. | Yuanchang Li Wenda Li Xiujuan Yan Zhenfang Zhou Xiaosong Guo Jing Liu Changming Mao Zhonghua Zhang Guicun Li | 2022 | Journal of Materials Science & Technology2022,,8: | 2 |

| 10 | Differential response of multiple zebrafish hepatic F-box protein genes to 17α-ethinylestradiol treatment显示文摘Estrogens are accumulating in environment and their effects on a variety of reproductive processes and tumorigenesis were reported by previous study, but the mechanism of estrogen promoting neoplasia was still not clear. F-box protein (FBP) is the component of E3 ubiquitin ligase which takes part in a variety of key biological processes. In this study, using mature male zebrafish, which are more sensitive to estrogen treatment, we examined influence of 17α-ethinylestradiol (EE2) exposure on the expression of a series of hepatic FBP genes, which take part in a variety of biological processes, including tumorigenesis. The influence of EE2 on the expression of hepatic mRNA concentrations of FBP genes were quantified based on the expression of the optimal internal control gene in male zebrafish after 7-day exposure to EE2, from a low-dose concentration (1 ng/L) to environmentally relevant concentrations (10, 100 ng/L). Our results showed that EE2 exposure reduced the expression of fbxl14a, fbxl14b, fbxo25 and β-TRCP2b, but enchanced the expression of skp2. While the alterations in fbxl2, fbxw7, fbxo9, β-TRCP2a, fbxl18 and fbxo45 mRNA levels were not observed after EE2 exposure. Thus, our results showed that the expression of hepatic FBP genes exhibited differentially in male zebrafish exposed EE2. | 16 | Machine Learning in Predicting Printable Biomaterial Formulations for Direct Ink Writing显示文摘Three-dimensional(3D)printing is emerging as a transformative technology for biomedical engineering.The 3D printed product can be patient-specific by allowing customizability and direct control of the architecture.The trial-and-error approach currently used for developing the composition of printable inks is time-and resource-consuming due to the increasing number of variables requiring expert knowledge.Artificial intelligence has the potential to reshape the ink development process by forming a predictive model for printability from experimental data.In this paper,we constructed machine learning(ML)algorithms including decision tree,random forest(RF),and deep learning(DL)to predict the printability of biomaterials.A total of 210 formulations including 16 different bioactive and smart materials and 4 solvents were 3D printed,and their printability was assessed.All ML methods were able to learn and predict the printability of a variety of inks based on their biomaterial formulations.In particular,the RF algorithm has achieved the highest accuracy(88.1%),precision(90.6%),and F1 score(87.0%),indicating the best overall performance out of the 3 algorithms,while DL has the highest recall(87.3%).Furthermore,the ML algorithms have predicted the printability window of biomaterials to guide the ink development.The printability map generated with DL has finer granularity than other algorithms.ML has proven to be an effective and novel strategy for developing biomaterial formulations with desired 3D printability for biomedical engineering applications. | Hongyi Chen Yuanchang Liu Stavroula Balabani Ryuji Hirayama Jie Huang | 2023 | Research2023,,4: | 1 |
