| 1 | Lactobacillus reuteri-derived extracellular vesicles maintain intestinal immune homeostasis against lipopolysaccharideinduced inflammatory responses in broilers显示文摘Background:Lactobacillus reuteri strains are widely used as probiotics to prevent and treat inflammatory bowel disease by modulating the host’s immune system.However,the underlying mechanisms by which they communicate with the host have not been clearly understood.Bacterial extracellular vesicles(EVs)have been considered as important mediators of host-pathogen interactions,but their potential role in commensals-host crosstalk has not been widely studied.Here,we investigated the regulatory actions of EVs produced by L.reuteri BBC3,a gut-associated commensal bacterium of Black-Bone chicken,in the development of lipopolysaccharide(LPS)-induced intestinal inflammation in a chicken model using both in vivo and in vitro experiments.Results:L.reuteri BBC3 produced nano-scale membrane vesicles with the size range of 60-250 nm.Biochemical and proteomic analyses showed that L.reuteri BBC3-derived EVs(LrEVs)carried DNA,RNA and several bioactive proteins previously described as mediators of other probiotics’beneficial effects such as glucosyltransferase,serine protease and elongation factor Tu.In vivo broiler experiments showed that administration of LrEVs exerted similar effects as L.reuteri BBC3 in attenuating LPS-induced inflammation by improving growth performance,reducing mortality and decreasing intestinal injury.LrEVs suppressed the LPS-induced expression of pro-inflammatory genes(TNF-α,IL-1β,IL-6,IL-17 and IL-8),and improved the expression of anti-inflammatory genes(IL-10 and TGF-β)in the jejunum.LrEVs could be internalized by chicken macrophages.In vitro pretreatment with LrEVs reduced the gene expression of TNF-α,IL-1βand IL-6 by suppressing the NF-κB activity,and enhanced the gene expression of IL-10 and TGF-βin LPS-activated chicken macrophages.Additionally,LrEVs could inhibit Th1-and Th17-mediated inflammatory responses and enhance the immunoregulatory cells-mediated immunosuppression in splenic lymphocytes of LPS-challenged chickens through the activation of macrophages.Finally,we revealed that the reduced content of both vesicular proteins and nucleic acids attenuated the suppression of LrEVs on LPS-induced inflammatory responses in ex vivo experiments,suggesting that they are essential for the LrEVs-mediated immunoregulation.Conclusions:We revealed that LrEVs participated in maintaining intestinal immune homeostasis against LPSinduced inflammatory responses in a chicken model.Our findings provide mechanistic insight into how commensal and probiotic Lactobacillus species modulate the host’s immune system in pathogens-induced inflammation. | Rujiu Hu Hua Lin Mimi Wang Yuezhen Zhao Haojing Liu Yuna Min Xiaojun Yang Yupeng Gao Mingming Yang | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 5 |

| 3 | FGFR/RACK1 interacts with MDM2, promotes P53 degradation, and inhibits cell senescence in lung squamous cell carcinoma显示文摘Objective:FGFR is considered an important driver gene of lung squamous cell carcinoma(LSCC).Thus,identification of the biological events downstream of FGFR is important for the treatment of this malignancy.Our previous study has shown that the FGFR/RACK1 complex interacts with PKM2 and consequently promotes glycolysis in LSCC cells.However,the biological functions of the FGFR/RACK1 complex remain poorly understood.Methods:Anchorage-independent assays and in vivo tumorigenesis assays were performed to evaluate cancer cell malignancy.Distant seeding assays were performed to evaluate cancer cell metastasis.β-gal staining was used to examine cell senescence,and immunoprecipitation assays were performed to examine the interactions among FGFR,RACK1,and MDM2.Results:FGFR/RACK1 was found to regulate the senescence of LSCC cells.Treatment with PD166866,an inhibitor of FGFR,or knockdown of RACK1 induced senescence in LSCC cells(P<0.01).A molecular mechanistic study showed that FGFR/RACK1/MDM2 form a complex that promotes the degradation of p53 and thus inhibits cell senescence.PD166866 and RG7112,an MDM2/p53 inhibitor,cooperatively inhibited the colony formation and distal seeding of LSCC cells(P<0.01),and upregulated the expression of p53 and p21.Conclusions:Together,our findings revealed the regulatory roles and mechanisms of FGFR/RACK1 in cell senescence.This understanding should be important in the treatment of LSCC. | Tao Chen Fei Wang Shupei Wei Yingjie Nie Xiaotao Zheng Yu Deng Xubin Zhu Yuezhen Deng Nanshan Zhong Chengzhi Zhou | 2021 | Cancer Biology & Medicine2021,18,3: | 3 |

| 7 | Effect of Zibushenjing Fang on skeletal development and brain tissue antioxidation in mice with kidney essence insufficiency: a mechanistic study显示文摘OBJECTIVE: To investigate the effects of Zibushenjing Fang (formula for tonifying the kidney essence) on skeletal development and brain tissue antioxidation in mice with kidney essence insufficiency. METHODS: Fifty male Kunming mice were randomly divided into five groups: normal group, model group, Jinkuishenqi Wan group, Zibushenjing Fang high dose group, and Zibushenjing Fang low dose group, with 10 mice in each group. The model of kidney essence insufficiency syndrome was established in all the mice except the normal group by using a cat to threaten the mice and by swimming until exhaustion daily which lasted about 21 days. Mice in the model group were administered 20 mL/ kg -1 · d -1 of normal saline intragastrically. The Jinkuishenqi Wan group was given 2.7 g/kg-1 · d-1 ofa solution of Jinkuishenqi Wan. The Zibushenjing Fang high dose group was given 20 g/kg -1 · d -1 and the Zibushenjing Fang low dose group was given 10 g/kg -1 · d -1 of a solution of Zibushenjing Fang.The general condition of all the groups was observed, including the quantity of food and water intake, swimming time, length of femur, and weight of the femur and musculus quadriceps femoris. The total superoxide dismutase (T-SOD), glutathione peroxi- dase (GSH-Px), and methane dicarboxylic aldehyde (MDA) activities in the brain tissues were detected. RESULTS: Zibushenjing Fang could improve the manifestation of kidney essence insufficiency, increase the quantity of food and water intake, swimming time, femur length, and femur and musculus quadriceps femoris weight. It could also regulate the activities of T-SOD, GSH-Px, and MDA in brain tissue.
