1Evolving concepts in bone infection: redefining “biofilm”,“acute vs. chronic osteomyelitis”, “the immune proteome” and “local antibiotic therapy”显示文摘Osteomyelitis is a devastating disease caused by microbial infection of bone. While the frequency of infection following elective orthopedic surgery is low, rates of reinfection are disturbingly high. Staphylococcus aureus is responsible for the majority of chronic osteomyelitis cases and is often considered to be incurable due to bacterial persistence deep within bone. Unfortunately, there is no consensus on clinical classifications of osteomyelitis and the ensuing treatment algorithm. Given the high patient morbidity,mortality, and economic burden caused by osteomyelitis, it is important to elucidate mechanisms of bone infection to inform novel strategies for prevention and curative treatment. Recent discoveries in this field have identified three distinct reservoirs of bacterial biofilm including: Staphylococcal abscess communities in the local soft tissue and bone marrow, glycocalyx formation on implant hardware and necrotic tissue, and colonization of the osteocyte-lacuno canalicular network(OLCN) of cortical bone. In contrast, S.aureus intracellular persistence in bone cells has not been substantiated in vivo, which challenges this mode of chronic osteomyelitis. There have also been major advances in our understanding of the immune proteome against S. aureus, from clinical studies of serum antibodies and media enriched for newly synthesized antibodies(MENSA), which may provide new opportunities for osteomyelitis diagnosis, prognosis, and vaccine development. Finally, novel therapies such as antimicrobial implant coatings and antibiotic impregnated 3D-printed scaffolds represent promising strategies for preventing and managing this devastating disease. 4Mitochondrial GPX1 silencing triggers differential photosynthesis impairment in response to salinity in rice plants显示文摘The physiological role of plant mitochondrial glutathione peroxidases is scarcely known. This study attempted to elucidate the role of a rice mitochondrial isoform(GPX1) in photosynthesis under normal growth and salinity conditions. GPX1 knockdown rice lines(GPX1s) were tested in absence and presence of 100 mM NaCl for 6 d.Growth reduction of GPX1 s line under non-stressful conditions, compared with non-transformed(NT) plants occurred in parallel to increased H_2O_2 and decreased GSH contents. These changes occurred concurrently with photosynthesis impairment, particularly in Calvin cycle's reactions, since photochemical efficiency did not change.Thus, GPX1 silencing and downstream molecular/metabolic changes modulated photosynthesis differentially. In contrast, salinity induced reduction in both phases of photosynthesis, which were more impaired in silenced plants.These changes were associated with root morphology alterations but not shoot growth. Both studied lines displayed increased GPX activity but H_2O_2 content did not change in response to salinity. Transformed plants exhibited lower photorespiration, water use efficiency and root growth, indicating that GPX1 could be important to salt tolerance. 6Esophageal early basaloid squamous carcinoma with unusual narrowband imaging magnified endoscopy findings显示文摘Basaloid squamous carcinoma(BSC)is a rare variant of esophageal cancer.There are very few reports of'early'BSC.Here we report a case of early BSC with unusual findings by narrowband imaging magnified endoscopy(NBI-ME).A 70-year-old man with a middle thoracic esophageal tumor was referred to our hospital.White-light endoscopy revealed a reddish depressed lesion 5 mm in diameter having a subepithelial tumor-like prominence with a gentle rising slope.NBI-ME revealed irregular loop-shaped microvessels coexistent with thick irregularly branched non-looped vessels.Iodine staining revealed a pale brown lesion.We performed endoscopic submucosal dissection for diagnostic treatment.Histologic examination showed the proliferation of basal cell-like hyperchromatic tumor cells in the lamina propria and with slight invasion into the submucosa at a depth of 320μm.The tumor cells formed solid nests and microcystic structures,containing an Alcian blue-positive mucoid matrix.The surface was covered with squamous epithelium without cellular atypia.Thin vessels were observed in the intra-epithelial papilla and thick vessels were observed around the solid nests beneath the epithelium.Based on these findings together,we diagnosed the lesion as BSC.In this case,the NBI-ME findings differed from those of typical squamous cell carcinoma in that both non-invasive cancer-like irregular loop-shaped microvessels coexisted with massively invasive cancerlike thick non-looped vessels.We speculate that the looped and non-looped vessels observed by NBI-ME histologically corresponded to thin vessels in the intraepithelial papilla and thick vessels around the tumor nests,respectively.These NBI-ME findings might be a feature of early esophageal BSC.Yugo Kai Motohiko Kato Yoshito Hayashi Tomofumi Akasaka Shinichiro Shinzaki Tsutomu Nishida Masahiko Tsujii Eiichi Morii Tetsuo Takehara 2014World Journal of Gastroenterology2014,20,35:4
7Analysis of blasting damage in adjacent mining excavations显示文摘Following a small-scale wedge failure at Yukon Zinc's Wolverine Mine in Yukon, Canada, a vibration monitoring program was added to the existing rockbolt pull testing regime. The failure in the 1150 drift occurred after numerous successive blasts in an adjacent tunnel had loosened friction bolts passing through an unmapped fault. Analysis of blasting vibration revealed that support integrity is not compromised unless there is a geological structure to act as a failure plane. The peak particle velocity(PPV) rarely exceeded 250 mm/s with a frequency larger than 50 Hz. As expected, blasting more competent rock resulted in higher PPVs. 9地基多轴DOAS仪器的NO_2斜柱浓度对比研究显示文摘介绍了两台地基多轴DOAS仪器测量的NO2斜柱浓度的对比研究。利用日本海洋研究开发机构的一套多轴DOAS设备在2009年11月—12月31日期间16d的观测数据,与安光所自主研发的一套多轴DOAS设备测量的NO2差分斜柱浓度进行了对比。通过对比发现,采用自动调整积分时间的方法,与固定积分时间的设置相比仪器具有更高的信噪比;两套仪器的反演结果在小角度下具有比较好的一致性,相关系数高达0.995,但随着角度的增大相关性逐渐变差。在9点至16点时段内相对偏差较小,其中20°方向的结果最为接近,最小偏差仅有12%,但在9点前和16点后二者偏差增大。日方仪器在可见波段的反演结果明显好于紫外波段,反演中的剩余噪声减小了60%以上,同时在可见波段的差分斜柱浓度的计算结果与我们的多轴DOAS在紫外波段的计算结果在全天都具有非常好的一致性。徐晋 谢品华 司福祺 李昂 窦科 刘文清 Yugo Kanaya Hitoshi Irie 2012光谱学与光谱分析2012,32,2:2
