| 1 | Effects and mechanisms of Bazhen decoction,Siwu decoction,and Sijunzi decoction on 5-fluorouracil-induced anemia in mice显示文摘OBJECTIVE:To investigate the effects of Bazhen decoction(BZD),Siwu decoction(SWD) and Sijunzi decoction(SJZD) in mice with anemia induced by5-fluorouracil(5-FU) and discussed the possible pharmacological hematopoietic mechanism to provide experimental evidence for the clinical use of the three classical prescriptions in the treatment of anemia.METHODS:Anemia was induced by intravenous injection of 5-FU and 80 female Kunming mice were randomly,assigned to oral administration of SWD,SJZD,or BZD daily for 10 days.Peripheral blood cells count and bone marrow cell cycle were monitored to evaluate anti-anemia effects.Serum cytokines,interferon-γ(IFN-γ),interleukin-3(IL-3),erythropoietin(EPO),granulocyte-macrophage colonystimulating factor(GM-CSF),and tumor necrosis factor-α(TNF-α) were assayed.EPO m RNA expression was assayed in kidney and liver tissue homogenates.RESULTS:BZD and SWD significantly increased the number of red blood cells,hemoglobin concentration,and hematocrit,promoted bone marrow cells to enter the cell cycle,proliferate and differentiate,significantly increased IL-3 secretion,and significantly inhibited IFN-γ secretion.BZD stimulated transcription of EPO m RNA in the kidney and liver and enhanced serum EPO expression.A therapeutic effect of SJZD was not observed.CONCLUSION:BZD and SWD treatment specifically enhanced hematopoietic function and mediated myelopoiesis by altering serum cytokines levels and accelerating entry of bone marrow cells into the cell cycle.Better curative effects were achieved via nourishing both Qi and blood(BZD) than by enriching the blood(SWD) or invigorating Qi(SWZD)alone. | Tian Yunan Xiang Yuke Wan Guoran Wan Dong Zhu Huifeng Wang Tao Yang Xian | 2016 | Journal of Traditional Chinese Medicine2016,36,4: | 12 |

| 3 | Sequence modification of merB gene and high organomercurial resistance of transgenic tobacco plants显示文摘Mercury pollution has caused severe damage to environment and great attention has been paid to its control. Phytoremediation may become one of the most efficient measures to recover the polluted soil since it is economical, highly efficient and friendly to environment. In this report, plant genetic engineering methods were employed to modify the DNA sequence of merB genes that catalyze the conversion of organomercurals into ionic mercury. The modified mer-Bhe genes were introduced into tobacco by Agrobacterium, and the resultant transgenic plants were verified by Southern and Northern hybridization. High level of organomercu-rial resistance was detected on progenies of transgenic plants, some of which were resistant to PMA (phenyl mercury acetate) of 2.5μmol/L whereas 0.1μmol/L PMA killed the seedlings of wild-type tobacco in soiless culrure. With the increase of PMA concentration, the inhibition of the seedling growth became apparent. | 4 | Effect of pretreatment with different concertrations of morphine, fentanyl and tramadol on the differentiation of human helper T cells in vitro显示文摘Objective: To investigate and compare the effects of different concentrations of morphine, fentanyl and tramadol on the differentiation of human adult helper T cells in vitro. Methods: Twenty out-patients without immune disease were selected and their peripheral blood was collected. Then the whole blood of peripheral blood mononuclear cells (PBMCs) were pretreated with different concentration of morphine, fentanyl and tramadol for 24 h. The level of CD4+IFN-γ+IL-2+/CD4+IL-4+IL-10+ was analyzed by three-color flow cytometry, and the CD4+CCR5+ and CD4+CCR3+ cells were counted to observe the imbalance of Th2/Th2. Results: The number of Th2 increased significantly and the ratio of Th2/Th2 decreased dramatically compared with the control group, and there was a dose-dependent fashion in all drugs. | 9 | Sign-reversed anomalous Nernst effect in the ferromagnetic Weyl-semimetal Fe_(3-δ)GeTe_(2): the role of Fe vacancies显示文摘Anomalous Nernst effect, as a thermal partner of anomalous Hall effect, is particularly sensitive to the Berry curvature anomaly near the Fermi level, and has been used to probe the topological nature of quantum materials. In this work, we report the observation of both effects in the ferromagnetic Weyl-semimetal Fe_(3-δ)GeTe_(2) with tunable Fe vacancies. With decreasing Fe vacancies,the anomalous Hall conductivity evolves as a function of the longitudinal conductivity from the hopping region to the region where the intrinsic Berry curvature contribution dominates. Concomitant evolutions in the anomalous Nernst signal and the anomalous off-diagonal thermoelectric coefficient are observed below the Curie temperature, displaying a unique sign change caused by the Fe vacancies. | 10 | Intrathecal co-administration of ketamine and morphine preventing activation of astrocytes and decreasing releases of IL-1β and IL-6 from spinal cord in rats of morphine tolerance显示文摘Objective: To investigate the effects of intrathecal administration of ketamine, a non-competitive N-methy-D-aspartate receptor antagonist, combined with morphine on the activation of astrocytes and releases of IL-1β and IL-6 from spinal cord in the rats of morphine tolerance. Methods: Twenty-seven Sprague-Dawley male rats were randomly divided into sham-operated, morphine tolerance, and morphine plus ketamine group. The subarachnoid catheterization of all the rats was prepared by the method of Jianping Yang. Morphine 20 μg in 10 μl was administrated intrathecally to induce spinal morphine tolerance once daily for 5 consecutive days. Morphine and ketamine 250 μg in 10 μl total volume was given in morphine plus ketamine group. Three groups all received intrathecal morphine 5 μg in 10 μl for morphine challenge test at 24 h after last administration of the morphine. After morphine challenge test, lumbar spinal tissues were taken for measurement of glial fibrillary acidic protein (GFAP) of astrocyte in lumbar spinal horn cord by immunohistochemistry and IL-1β and IL-6 of spinal cord by ELISA. Results: The decrease of %MPE induced by chronic intrathecal morphine was inhibited by ketamine and hyperalgesia and allodynia induced by morphine-withdrawl were alleviated. The average areas, the average absorbency ([AKA-]), the integral absorbency (A) of GFAP immuno-reactive cells in the dorsal horn, and IL-1β and IL-6 of spinal cord were significantly larger in morphine tolerance group than in morphine plus ketamine group. Conclusion: Co-administration of ketamine and morphine enhance antinociceptive effect of morphine and prevent the development of morphine tolerance. | 12 | Comparative cost analysis of three different anesthesia methods in gynecological laparoscopic surgery显示文摘In the current study,we assessed and evaluated the costs and benefits of three popular methods of general anesthesia practiced in our department for gynecological laparoscopic surgery in recent years.Sixty adult female patients who underwent elective gynecological laparoscopic surgery under general anesthesia were randomly divided into three groups:group Ⅴ,group Ⅰ and group C.In group Ⅴ,anesthesia was induced intravenously with midazolam,remifentanil,propofol and vecuronium,and maintained with continuous infusion of propofol and remifentanil.In group Ⅰ,anesthesia was intravenously induced with midazolam,fentanyl,propofol and vecuronium,and maintained with inhaled isoflurane and intravenous bonus of fentanyl.In group C,anesthesia was induced as in group Ⅰ,but maintained with isoflurane inhalation combined with propofolremifentanil infusion.All patients received vecuronium for muscle relaxation.Perioperative incidences of complications and total anesthesia costs for patients in all groups were recorded.In addition,postoperative satisfaction of the patients was also noted,and similar outcomes of the satisfaction were reported in all 60 patients.Although there was no statistical significance among groups,the incidence of postoperative nausea and vomiting were higher in group C,and the rates of shivering and the needs for analgesics were higher in group Ⅴ.Anesthesia costs in group Ⅰ were the lowest.Therefore,it is concluded that the costs of anesthesia induced with midazolam,fentanyl,propofol,vecuronium,and maintained with isoflurane,fentanyl and vecuronium are cheapest,and there is no significant difference in patients’satisfaction and safety among the three above-mentioned methods of anesthesia in our department. | Xiaohui Chi Yeling Chen Mingfeng Liao Fei Cao Yuke Tian Xueren Wang | 2012 | Frontiers of Medicine2012,6,3: | 0 |
| 13 | Effects of intracerebroventricular NMDA and non-NMDA receptor agonists or antagonists on general anesthesia of propofol in mice显示文摘The effects of intracerebroventricular(icv)agonists and antagonists of N-methyl-D-aspartate(NMDA)and alpha-amino-3-hydroxy–5-methyl-4-isoxazole-propionic acid(AMPA)receptors on the general anesthesia of propofol were studied.A total of 144 Kunming mice,male and female with body mass of(22P3)g,were used.Part One of the Experiment:a total of 104 Kunming mice,male and female,were randomly divided into 13 groups.Intracerebroventricular artificial cerebral fluid(aCSF)or different doses of NMDA,AMPA,MK-801 or NBQX was injected immediately after intravenously administered propofol 25 mg/kg and the recovery time following the loss of righting reflex(LORR)was recorded.Part Two of the Experiment:a total of 40 Kunming female mice were divided randomly into 5 groups and injected with icv aCSF or NMDA,AMPA,MK-801 or NBQX after intraperitoneally administered propofol 50 mg/kg.The pain threshold of the mice was then investigated by hot-plate test(HPPT).NMDA(0.05 or 0.075μg,icv)or AMPA(0.05μg,icv)exhibited no effects on the LORR,but NMDA(0.1μg,icv)or AMPA(0.075 or 0.1μg,icv)prolonged the LORR significantly compared with the aCSF group(P<0.05,P<0.01).The LORR of the 2μg MK-801 group had no changes,while those of the 4 or 8μg MK-801 groups were prolonged significantly.The LORR of the 0.5,2 or 4μg NBQX groups were all prolonged significantly.NMDA 0.05μg or AMPA 0.05μg decreased the pain threshold slightly but did not differ in effect compared with the aCSF group;2μg MK-801 or 0.5μg NBQX both increased the pain threshold significantly.Our results indicate that propofol produces general anesthesia partly through an interaction with brain NMDA and AMPA receptors in mice. | XU Aijun DUAN Shiming TIAN Yuke | 2007 | Frontiers of Medicine2007,1,2: | 0 |
