3High-purity magnesium screws modulate macrophage polarization during the tendon–bone healing process in the anterior cruciate ligament reconstruction rabbit model显示文摘Magnesium(Mg)screws perform clinical potential in anterior cruciate ligament(ACL)reconstruction,and promote fibrocartilaginous entheses regeneration at the femoral entrance.We aim to prove that high-purity Magnesium(HP Mg)screws modulate macrophage polarization in fibrocartilage interface regeneration both in vitro and in vivo.HP Mg extracts performed good cytocompatibility and significantly promoted M2 macrophage polarization in the flow cytometry and ELISA assays.M2 macrophages stimulated fibrochondrocyte differentiation of cocultured hBMSCs,and HP Mg extracts had synergistic effect on the process.Then we applied HP Mg screws,with Ti screws as control,in the ACL reconstruction rabbit model.In the histological and immunofluorescence analysis,HP Mg screws inhibited M1 polarization at 2 weeks and highly promoted M2 polarization at 2 and 4 weeks at the tendon–bone interface.Furthermore,regeneration of fibrocartilaginous entheses,rather than the fibrovascular scar interface,was detected in the HP Mg group at 12 weeks.For further mechanism study via RNA-seq detection and WB assays,we found that AKT1 was highly activated in M2 polarization,and HP Mg could stimulate AKT1 expression,rather than AKT2,in the early phase of tendon–bone healing.Our study elucidated macrophage polarization during tendon–bone healing process and emphasized HP Mg on M2 polarization and fibrocartilage interface regeneration via the selective activation of AKT1 and PI3K/AKT pathway.PengFei Cheng ZhenJun Weng Musha Hamushan Weijie Cai Yubo Zhang Zun Ren Yunchu Sun XiaoNong Zhang Hao Shen Pei Han 2022Regenerative Biomaterials2022,9,1:0
4Immunostimulatory gene therapy combined with checkpoint blockade reshapes tumor microenvironment and enhances ovarian cancer immunotherapy显示文摘Immune evasion has made ovarian cancer notorious for its refractory features, making the development of immunotherapy highly appealing to ovarian cancer treatment. The immune-stimulating cytokine IL-12 exhibits excellent antitumor activities. However, IL-12 can induce IFN-γ release and subsequently upregulate PDL-1 expression on tumor cells. Therefore, the tumor-targeting folate-modified delivery system F-DPC is constructed for concurrent delivery of IL-12 encoding gene and small molecular PDL-1 inhibitor(i PDL-1) to reduce immune escape and boost anti-tumor immunity. The physicochemical characteristics, gene transfection efficiency of the F-DPC nanoparticles in ovarian cancer cells are analyzed. The immune-modulation effects of combination therapy on different immune cells are also studied. Results show that compared with non-folate-modified vector, folate-modified F-DPC can improve the targeting of ovarian cancer and enhance the transfection efficiency of p IL-12. The underlying anti-tumor mechanisms include the regulation of T cells proliferation and activation, NK activation,macrophage polarization and DC maturation. The F-DPC/p IL-12/i PDL-1 complexes have shown outstanding antitumor effects and low toxicity in peritoneal model of ovarian cancer in mice. Taken together, our work provides new insights into ovarian cancer immunotherapy. 5Dispersion and Interface Characteristics of Nano-TiO_2 Based Wood Preservatives显示文摘The key problems of preparing water-based nano TiO2 wood preservatives are the particle’s agglomeration and dispersion.The influences of adding titanate NDZ-105 and sodium hexamet aphosphate on transmittance,effects of dispersion modification on crystal type,distribution and interface characteristics of modified TiO2 in wood were studied by photometric,XRD,SEM and FTIR analysis,respectively.The results show that the dispersion of nano TiO2 suspension firstly increased, then decreased as the amount of the two kinds of dispersed agent increased,when addition amount of titanate N DZ-105,six partial sodium were separately 5%and 3%-5%,the nano TiO2 wood preservatives were obtained that their dispersive property were stabilized.After dispersion modification,the TiO2 maintained the crystal diffraction characteristics of anatase;the nano TiO2 distributed in wood successfully,showing a granule or ball-shaped,and the hydrogen bond and covalent bond were combined between wood and nano-TiO2,but only hydrogen bond was combined between wood cellulose and IPBC.Wang Min Wu Yiqiang Hu Yunchu Zhang Xinli Yang Shoulu 2012Chinese Forestry Science and Technology2012,11,3:0
6Co-delivery of doxorubicin and curcumin via cRGD-peptide modified PEG-PLA self-assembly nanomicelles for lung cancer therapy显示文摘Lung cancer is the most common malignancy in the world, with a high mortality rate. Nevertheless,therapies to act effectively against lung cancer remain elusive. So far, chemotherapy is still the frontline treatment of lung cancer. Doxorubicin(DOX) is a broad-spectrum anti-tumor drug. However, DOX often has serious side effects and causes multi-drug resistance, which greatly limits its clinical application.In this work, biodegradable methoxy poly(ethylene glycol)-poly(lactic acid)(MPEG-PLA) and cyclo(ArgGly-Asp-D-Phe-Lys)(c RGD) polypeptide modified PEG-PLA(c RGD-PEG-PLA) copolymers were used for the co-delivery of curcumin(CUR) and DOX(CUR-DOX/c RGD-M). The particle size of the self-assembled drugloaded nanomicelle approximately was 27.4 nm and the zeta potential was -2.7mV. Interestingly, CUR can enhance the uptake of DOX by Lewis lung carcinoma(LL/2) cells. The experimental results in vivo and in vitro showed that CUR-DOX/c RGD-M combination therapy could promote apoptosis of lung cancer cells, and conspicuously inhibit the tumor growth.
