1Inulin Can Alleviate Metabolism Disorders in ob/ob Mice by Partially Restoring Leptinrelated Pathways Mediated by Gut Microbiota显示文摘Inulin has been used as a prebiotic to alleviate glucose and lipid metabolism disorders in mice and humans by modulating the gut microbiota. However, the mechanism underlying the alleviation of metabolic disorders by inulin through interactions between the gut microbiota and host cells is unclear. We use ob/ob mice as a model to study the effect of inulin on the cecal microbiota by16 S rRNA gene amplicon sequencing and its interaction with host cells by transcriptomics. The inulin-supplemented diet improved glucose and lipid metabolism disorder parameters in ob/ob mice,alleviating fat accumulation and glucose intolerance. The a diversity of gut microbial community of ob/ob mice was reduced after inulin treatment, while the b diversity tended to return to the level of wild type mice. Interestingly, Prevotellaceae UCG 001(family Prevotellaceae) was obviously enriched after inulin treatment. A comparative analysis of the gene expression profile showed that the cecal transcriptome was changed in leptin gene deficiency mice, whereas the inulin-supplemented diet partially reversed the changes in leptin gene-related signaling pathways, especially AMPK signaling pathway, where the levels of gene expression became comparable to those in wild type mice.Further analysis indicated that Prevotellaceae UCG 001 was positively correlated with the AMPK signaling pathway, which was negatively correlated with markers of glycolipid metabolism disorders. 2A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 4Benthic Habitat Quality Assessment Based on Biological Indices in Xiaoqing River Estuary and Its Adjacent Sea of Laizhou Bay,China显示文摘Based on the macrofauna data(2008-2011) in Xiaoqing River estuary and its adjacent sea, Laizhou Bay of Bohai Sea, China, the AZTI Marine Biotic Index(AMBI) and Multivariate AMBI(M-AMBI) were used for benthic habitat quality(BHQ) assessment. Results showed that BHQ presented an obvious trend of improvement along the direction of stream channel and river mouth, and in the coastal areas. AMBI and M-AMBI were significantly related to environmental pressure gradient data. Therefore, the two indices can well indicate BHQ in the studied area. However, there were significant differences between results of the two indices. In the cases of low taxa number and high abundance of single species, AMBI might overestimate BHQ. We thus adjusted its thresholds to solve this problem. And M-AMBI might overestimate BHQ when benthic assemblage was dominated by the opportunistic species. 6Effects of complement inhibition on the ABC phenomenon in rats显示文摘Researchers reported that intravenously injected PEGylated colloidal drug carriers lose their long-circulating characteristic and accumulated extensively in liver when they are administrated twice in the same animal with certain intervals. This phenomenon was referred to as the 'accelerated blood clearance(ABC) phenomenon'. Some former studies had found that complement-mediated phagocytosis, activated by antigen–antibody complex, was responsible for inducing the phenomenon. According to the theory, we have used cobra venom factor to deplete complement in vivo and to investigate the effect of complement inhibition on the ABC phenomenon. Rats were administered by injection of cobra venom factor solution to build up the model of complement exhaustion/inhibition, and the effect of the inhibition of complement on ABC phenomenon was carried out. It seemed that inhibition of complement didn’t affect the pharmacokinetic of the first infection. By contrast, in rats of which complement had been depleted, the second dose of PEGylated nanoemulsions showed enhanced circulation time compared with normal rats in a complement inhibition-independent manner, but the ABC phenomenon was not completely eliminated. 7Multimarker Detection of MAGE-1,MAGE-3 and AFP mRNAs by a Real-time Quantitative PCR Assay:a Possible Predictor of Hematogenous Micrometastasis of Hepatocellular Carcinoma显示文摘OBJECTIVE To explore the relationship between multimarker detection of MAGE-1,MAGE-3 and AFP mRNAs in the peripheral blood of patients with hepatocellular carcinoma and micrometastasis using a realtime quantitative-PCR(real-time Q-PCR)assay. METHODS Peripheral blood samples were obtained from control subjects and 86 patients with hepatocellular carcinoma (HCC).Real-time Q-PCR was used to detect MAGE-1,MAGE-3, and AFP mRNAs in the blood cells. RESULTS In 86 tumor specimens,the positivity for MAGE-1, MAGE-3,and AFP genes was respectively 34.9%(30/86),60.5% (52/86)and 69.8%(60/86).All specimens expressed at least one marker.MAGE-1,MAGE-3,and AFP transcripts were detected respectively in 12(14.0%),18(20.1%)and 29(33.7%)of the 86 blood specimens from hepatocellular carcinoma patients,while 45 specimens(52.3%)were positive for at least one marker.In addition,MAGE-1,MAGE-3 and AFP gene transcripts were not detected in any peripheral blood specimens from 25 chronic liver disease patients and 28 normal healthy volunteers.The positive rate correlated with the TNM clinical stages,extrahepatic metastasis and portal vein carcinothrombosis(P<0.05).No correlation was found between tumor size,tumor number, differentiation,serum a-fetoprotein(AFP)and the positive rate. 14Thermo-kinetic connectivity by integrating thermo-kinetic correlation and generalized stability显示文摘Designing structured materials with optimized mechanical properties generally focuses on engineering microstructures,which are closely determined by the processing routes,such as phase transformations(PTs)and plastic deformations(PDs).Both PTs and PDs follow inherent trade-off relation between thermodynamic driving force ΔG and kinetic energy barrier Q,i.e.,so-called thermo-kinetic correlation.By analyzing nucleation and growth and proposing a conception of negative driving force integrating strain energy,interface energy and any kind of energy that equivalently inhibits the PT itself,ΔG^(S),unified expressions for the thermo-kinetic correlation and generalized stability(GS)were derived for three kinds of PTs,i.e.,diffusive PTs with simultaneously decreasedΔG and increased Q,diffusive PTs with simultaneously increasedΔG and decreased Q,and displacive PTs with simultaneously increased ΔG and decreased Q.This leads to so-called thermo-kinetic connectivity by integrating the thermo-kinetic correlation and the GS,where,by application in typical PTs,it was clearly shown,a criterion of high ΔG-high GS can be predicted by modulating chemical driving force,negative driving force and kinetic energy barrier for diffusion or nucleation.Following thermo-kinetic connectivity,analogous procedure for dislocation evolution upon PDs was performed,and materials design in terms of the highΔG-high GS criterion was discussed and prospected.Yuqing He Shaojie Song Jinglian Du Haoran Peng Zhigang Ding Huaiyu Hou Linke Huang Yongchang Liu Feng Liu 2022Journal of Materials Science & Technology2022,,32:1
15Novel Approach for Automatic Region of Interest and Seed Point Detection in CT Images Based on Temporal and Spatial Data显示文摘Accurately finding the region of interest is a very vital step for segmenting organs in medical image processing.We propose a novel approach of automatically identifying region of interest in Computed Tomography Image(CT)images based on temporal and spatial data.Our method is a 3 stages approach,1)We extract organ features from the CT images by adopting the Hounsfield filter.2)We use these filtered features and introduce our novel approach of selecting observable feature candidates by calculating contours’area and automatically detect a seed point.3)We use a novel approach to track the growing region changes across the CT image sequence in detecting region of interest,given a seed point as our input.We used quantitative and qualitative analysis to measure the accuracy against the given ground truth and our results presented a better performance than other generic approaches for automatic region of interest detection of organs in abdominal CT images.With the results presented in this research work,our proposed novel sequence approach method has been proven to be superior in terms of accuracy,automation and robustness.Zhe Liu Charlie Maere Yuqing Song 2019Computers, Materials & Continua2019,,5:1
16The accelerated blood clearance phenomenon of PEGylated nanoemulsion upon cross administration with nanoemulsions modified with polyglycerin显示文摘For investigating the accelerated blood clearance(ABC) phenomenon of polyglycerin modified nanoemulsions upon cross administration with polyethylene glycol(PEG) covered nanoemulsion, we used the 1,2-distea-royl-sn-glycero-3-phosphoethanolamine-npolyglycerine-610 and the 1,2-distearoyl-n-glycero-3-phosphoethanolamine-n-[methoxy(polyethylene glycol)-2000] as modify materials, the dialkylcarbocyanines as fluorescence indicator. Exhausted macrophages rat model was established and new material containing polycarboxyl structure was synthesized. The microplate reader and the in vivo optical imaging system were applied to measure the concentration of nanoemulsions in tissues.The results show that the first dose of polyglycerin modified nanoemulsion can induce the ABC phenomenon of the second dose of PEGylated nanoemulsion. With the increase in the amount of the surface polyglycerin, the extent of the ABC phenomenon decreases. Liver accumulation has positive relationship with the ABC phenomenon. Furthermore, kupffer cells in liver can get more immune information from polyhydroxy structure than polycarboxyl group in the modify compound. 18Molding of Molecular Sieve Residues and Their Application in Cleaning Oily Wastewater显示文摘To deeply clean oily wastewater,molecular sieve residues(MSRs)were sufficiently recycled and utilized due to their high specific surface area,porous structure,and outstanding adsorption property.Molding MSRs(MMSRs)were prepared by adding additives(starch,citric acid,and soluble glass)to MSRs and were then filled into a fixed bed for adsorbing and separating the oil in wastewater.Sodium dodecylbenzenesulfonate was used to modify the MMSRs,and their adsorption property was also investigated.In addition,the MSRs were characterized by scanning electron microscopy,energy-dispersive X-ray spectroscopy,Brunauer–Emmett–Teller analysis,and Fourier transform infrared spectroscopy.The results indicated that MMSRs satisfied the filling requirement of fixed bed,and their dynamic adsorption capacity could reach 0.1854 mg g^?1.Furthermore,the static adsorption capacity of MMSRs achieved 1.7346 mg g^?1 in the optimum conditions,and the oil adsorption performance of modified MMSRs was further enhanced.Therefore,this work suggests that MSRs are promising alternatives in cleaning oily wastewater.Zilu Jin Yuqing Zhang Song Wei Shuai Sun 2019Transactions of Tianjin University2019,25,6:1
19The As-surface of an iron-based superconductor CaKFe_(4)As_(4)显示文摘As a new type of iron-based superconductor, CaKFe_(4)As_(4) has recently been demonstrated to be a promising platform for observing Majorana zero modes (MZMs). The surface of CaKFe_(4)As_(4) plays an important role in realizing the MZM since it hosts superconducting topological surface states. However, due to the complicated crystal structure, the terminal surface of CaKFe_(4)As_(4) has not been determined yet. Here, by using scanning tunneling microscopy/spectroscopy (STM/S), we find that there are two types of surface structure in CaKFe_(4)As_(4). Bias-dependent atomic resolution images show an evolvement from 2–√×2–√ superstructure with respect to the As lattice into 1 × 1 when the tip is brought close to the surface, revealing the sublattice of missing As atoms. Together with the first-principles calculations, we show that the surface As layer has a buckled structure. 20The OX40-TRAF6 axis promotes CTLA-4 degradation to augment antitumor CD8^(+)T-cell immunity显示文摘Immune checkpoint blockade(ICB),including anti-cytotoxic T-lymphocyte associated protein 4(CTLA-4),benefits only a limited number of patients with cancer.Understanding the in-depth regulatory mechanism of CTLA-4 protein stability and its functional significance may help identify ICB resistance mechanisms and assist in the development of novel immunotherapeutic modalities to improve ICB efficacy.Here,we identified that TNF receptor-associated factor 6(TRAF6)mediates Lys63-linked ubiquitination and subsequent lysosomal degradation of CTLA-4.Moreover,by using TRAF6-deficient mice and retroviral overexpression experiments,we demonstrated that TRAF6 promotes CTLA-4 degradation in a T-cell-intrinsic manner,which is dependent on the RING domain of TRAF6.This intrinsic regulatory mechanism contributes to CD8+T-cell-mediated antitumor immunity in vivo.Additionally,by using an OX40 agonist,we demonstrated that the OX40-TRAF6 axis is responsible for CTLA-4 degradation,thereby controlling antitumor immunity in both tumor-bearing mice and patients with cancer.Overall,our findings demonstrate that the OX40-TRAF6 axis promotes CTLA-4 degradation and is a potential therapeutic target for the improvement of T-cell-based immunotherapies.Jizhang Yu Jikai Cui Xi Zhang Heng Xu Zhang Chen Yuan Li Yuqing Niu Song Wang Shuan Ran Yanqiang Zou Weicong Ye Dan Zhang Cheng Zhou Jiahong Xia Jie Wu 2023Cellular & Molecular Immunology2023,20,12:0
