1Gene expression of fibrinolytic factors urokinase plasminogen activator and plasminogen activator inhibitor-1 in rabbit temporo-mandibular joint cartilage with disc displacement显示文摘Background The urokinase plasminogen activator system is believed to play an important role in degradation of the extracellular matrix associated with cartilage and bone destruction; however its precise roles in temporomandibular disorders have not yet been clarified. The aims of this study were to investigate the gene expression of fibrinolytic factors urokinase plasminogen activator (uPA) and plasminogen activator inhibitor-1 (PAI-1) in the articular cartilage of rabbit temporomandibular joint (TMJ) with disc displacement (DD) and to probe the relationship between fibrinolytic activity and cartilage remodeling. Methods Disc displacement of right joints was performed in 36 of 78 rabbits under investigation. The animals were sacrificed at 4 days and 1, 2, 4, 8 and 12 weeks after surgery, respectively. The right joints of these animals were harvested and processed for the examination of mRNA expression of uPA and PAI-1 in articular cartilage using in situ hybridization techniques. Results The expression of uPA and PAI-1 was co-expressed weakly in the chondrocytes from transitive zone to hypertrophic zone and mineralized zone, while no hybridizing signals were shown in proliferative zone and superficial zone in control rabbits. The most striking was the up-regulation of uPA and PAI-1 mRNA in 4-day rabbits postoperatively at the onset of cartilage degeneration. The strongest hybridizing signals for uPA and PAI-1 were seen in 2-week rabbits postoperatively. After 2 weeks, the expression of uPA and PAI-1 began to decrease and reached nearly normal level at 12 weeks. Conclusions The expression of the uPA/PAI-1 system coincides with the pathological changes in condylar cartilage after DD. 2Significance of MD-2 and MD-2B expression in rat liver during acute cholangitis显示文摘AIM: To investigate the expression of myeloid differentiation protein-2 (MD-2), MD-2B (a splicing isoform of MD-2 that can block Toll-like receptor 4 (TLR4)/MD-2 LPS-mediated signal transduction) and TLR4 in the liver of acute cholangitis rats. METHODS: Male Sprague-Dawley rats (SPF level) were randomly divided into four groups: (A) sham-operated group; (B) simple common bile duct ligation group; (C)acute cholangitis group; and (D) acute cholangitis anti- TLR4 intervention group (n = 25 per group). Rat liver tissue samples were used to detect TLR4, MD-2 and MD-2B mRNA expression by fluorescence quantitative PCR in parallel with pathological changes. RESULTS: In acute cholangitis, liver TLR4 and MD-2 mRNA expression levels at 6, 12, 24, 48 and 72 h were gradually up-regulated but MD-2B mRNA expression gradually down-regulated (P < 0.05). After TLR4 antibody treatment, TLR4 and MD-2 mRNA expression were lower compared with the acute cholangitis group (P < 0.05). However, MD-2B mRNA expression was higher than in the acute cholangitis group (P < 0.05). MD-2 and TLR4 mRNA expressions were positively correlated (r = 0.94981, P < 0.05) and MD-2B mRNA expression was negatively correlated with MD-2 and TLR4 mRNA (r = -0.89031, -0.88997, P< 0.05). 14A New Fifth-Order Finite Volume Central WENO Scheme for Hyperbolic Conservation Laws on Staggered Meshes显示文摘In this paper,a new fifth-order finite volume central weighted essentially non-oscillatory(CWENO)scheme is proposed for solving hyperbolic conservation laws on staggered meshes.The high-order spatial reconstruction procedure using a convex combination of a fourth degree polynomial with two linear polynomials(in one dimension)or four linear polynomials(in two dimensions)in a traditional WENO fashion and a time discretization method using the natural continuous extension(NCE)of the Runge-Kutta method are applied to design this new fifth-order CWENO scheme.This new finite volume CWENO scheme uses the information defined on the same largest spatial stencil as that of the same order classical CWENO schemes[37,46]with the application of smaller number of unequal-sized spatial stencils.Since the new nonlinear weights are adopted,the new finite volume CWENO scheme could obtain the same order of accuracy and get smaller truncation errors in L1 and L¥norms in smooth regions,and control the spurious oscillations near strong shocks or contact discontinuities.The new CWENO scheme has advantages over the classical CWENO schemes[37,46]on staggered meshes in its simplicity and easy extension to multi-dimensions.Some one-dimensional and two-dimensional benchmark numerical examples are provided to illustrate the good performance of this new fifthorder finite volume CWENO scheme.Shengzhu Cui Zhanjing Tao Jun Zhu 2022Advances in Applied Mathematics and Mechanics2022,14,5:0
