1The Qitai radio telescope显示文摘This study presents a general outline of the Qitai radio telescope(QTT)project.Qitai,the site of the telescope,is a county of Xinjiang Uygur Autonomous Region of China,located in the east Tianshan Mountains at an elevation of about 1800 m.The QTT is a fully steerable,Gregorian-type telescope with a standard parabolic main reflector of 110 m diameter.The QTT has adopted an umbrella support,homology-symmetric lightweight design.The main reflector is active so that the deformation caused by gravity can be corrected.The structural design aims to ultimately allow high-sensitivity observations from 150 MHz up to115 GHz.To satisfy the requirements for early scientific goals,the QTTwill be equipped with ultra-wideband receivers and large field-of-view multi-beam receivers.A multi-function signal-processing system based on RFSo C and GPU processor chips will be developed.These will enable the QTT to operate in pulsar,spectral line,continuum and Very Long Baseline Interferometer(VLBI)observing modes.Electromagnetic compatibility(EMC)and radio frequency interference(RFI)control techniques are adopted throughout the system design.The QTT will form a world-class observational platform for the detection of lowfrequency(nano Hertz)gravitational waves through pulsar timing array(PTA)techniques,pulsar surveys,the discovery of binary black-hole systems,and exploring dark matter and the origin of life in the universe.The QTT will also play an important role in improving the Chinese and international VLBI networks,allowing high-sensitivity and high-resolution observations of the nuclei of distant galaxies and gravitational lensing systems.Deep astrometric observations will also contribute to improving the accuracy of the celestial reference frame.Potentially,the QTT will be able to support future space activities such as planetary exploration in the solar system and to contribute to the search for extraterrestrial intelligence.Na Wang Qian Xu Jun Ma Zhiyong Liu Qi Liu Hailong Zhang Xin Pei Maozheng Chen Richard N.Manchester Kejia Lee Xingwu Zheng Hans J.Kärcher Wulin Zhao Hongwei Li Dongwei Li Martin Süss Matthias Reichert Zhongyi Zhu Congsi Wang Mingshuai Li Rui Li Ning Li Guljaina Kazezkhan Wenming Yan Gang Wu Lang Cui Ming Zhang Haitao Li 2023Science China(Physics,Mechanics & Astronomy)2023,66,8:14
2Novel functional proteins interact with midkine in hepatic cancer cells显示文摘BACKGROUND: Midkine is a heparin-binding growth factor that promotes the proliferation, survival, migration and differentiation of various target cells. Midkine plays an important role in tumorigenesis and tumor progression, and is overexpressed in many human malignant tumors. Patients with high tumor midkine expression frequently have a worse prognosis than those with low expression. The present study was designed to investigate the interaction network of midkine in hepatic cancer cells, and to elucidate its role in hepatocellular carcinoma. METHODS: DNA encoding full-length midkine was cloned into pDBLeu vector to serve as bait in yeast two-hybrid screening to identify interacting proteins. Candidate proteins were examined on SC-Leu-Trp-His+3-AT (20 mmol/L) plates and assayed for X-gal activity, then sequenced and classified according to the GenBank. Finally, identified proteins were expressed by the in vitro expression system pCMVTnT, and protein interactions were confirmed by co-immunoprecipitation. RESULTS: Using the yeast two-hybrid system, we found 6 proteins that interacted with midkine: NK-kappa-B inhibitor alpha (I-κ-B-α), Dvl-binding protein naked cuticle 2, granulin, latent active TGF-β binding protein 3, latent active TGF-β binding protein 4, and phospholipid scramblase 1. 3Expression of co-stimulator 4-1BB molecule in hepatocellular carcinoma and adjacent non-tumor liver tissue,and its possible role in tumor immunity显示文摘AIM:To investigate the expression of 4-1BB molecule in hepatocellular carcinoma (HCC) and its adjacent tissues.METHODS:Reverse transcription-polymerase chain reaction (RT-PCR) was used to determine the gene expression of 4-1BB in hepatocarcinoma and its adjacent tissues,and peripheral blood mononuclear cells (PBMCs) from both HCC and health control groups.Flow cytometry was used to analyse the phenotypes of T cell subsets from the blood of HCC patients and healthy volunteers,and further to determine whether 4-1BB molecules were also expressed on the surface of CD4+ and CD8+ T cells. The localization of 4-1BB proteins on tumor infiltrating T cells was determined by direct immunofluorescence cytochemical staining and detected by confocal microscopy.RESULTS:4-1BB mRNA, which was not detectable in normal liver,was found in 19 liver tissues adjacent to tumor edge(<1.0cm).Low expression of 4-1BB mRNA was shown in 8 tumor tissues and 6 liver tissues located within 1 to 5cm away from tumor edge. In PBMCs, 4-1BB mRNA was almost not detected. Percentage of CD4^+, CD8^+ and CD3^+/CD25^+ T cells, as well as ratio of CD4 to CD8 revealed no difference between groups (P>0.05,respectively),while a significant lower percentage of CD3^+ T cell was found in HCC group as compared to healthy control group (P<0.05).However, 4-1BB molecules were almost not found on the surface of CD4+ and CD8+ T cells in HCC and healthy control group.Double-staining of 4-1BB^+/CD4^+ and 4-1BB^+/CD8^+ immunofluorescence on tumor infiltrating T cells was detected in 13 liver tissues adjacent to tumor edge (<1.0cm) by confocal microscopy.CONCLUSION: Although HCC may escape from immune attack by weak immunogenicity or downregulated expression of MHC-1 molecules on the tumor cell surface,tumor infiltrating T cells can be activated via other costimulatory signal pathways to exert a limited antitumor effect on local microenvironment. 4Survival factors after resection of small hepatocellular carcinoma显示文摘Early resection of hepatocellular carcinoma is a key measure to prolong the survival of patients. This study was designed to summarize our experience in surgical resection of small hepatocellular carcinoma (HCC), and to analyze the factors influencing the postoperative survival of patients. METHODS:The clinicopathologic data of 105 patients with small HCC after resection from 1986 through 2003 were analyzed ; the patients had been followed up for more than half a year (median 33 months). Nine clinicopathologic factors, preoperative α-fetoprotein (AFP) level, liver cirrhosis, Child-Pugh score, tumor size (>2cm vs.≤2 cm) and number (single vs. multiple), capsule formation, portal vein tumor thrombi (PVTT), Edmondson tumor grade and surgical method, were analyzed by the log-rank test and the Cox proportional harzards model analysis. RESULTS:The cumulative 1-,3- and 5-year survival rates after the operation were 86.5%, 70.3% and 55.2%, respectively, and the 1-, 3- and 5-year disease-free survival rates were 78%, 58. 9% and 45. 6%, respectively. One patient died from esophagogastric varices hemorrhage in 2 weeks after reoperation. Thirty-six patients had intrahepatic recurrence or metastasis postoperatively and 34 patients died. The Kaplan-Meier method and the Cox proportional harzards model analysis indicated that poor Child-Pugh score, tumor more than 2 cm in diameter, PVTT and multiple lesions (including satellitic lesions) were adverse factors affecting postoperative survival. The Cox proportional harzards model analysis indicated that tumor size, PVTT and multiple lesions were the factors influencing postoperative disease-free survival. CONCLUSIONS:Limited hepatectomy with a margin more than 1 cm is an appropriate surgical approach.
