1Multiple transgenes Populus xeuramericana 'Guariento' plants obtained by biolistic bombardment显示文摘A JERF36 regulation gene, a selection marker gene (NPT-II), and the foreign genes levansucrase (SacB), Vitreoscilla hemoglobin (vgb), and Binary coleopterus insect resistance (BtCry3A+OC-I) were co-transferred into Populus xeuramericana 'Guariento' using biolistic bombardment; 25 kanamycin resistant plants were obtained. The results of PCR and Southern hybridization showed that the foreign genes had been integrated into the genome of P. xeuramericana 'Guariento' and 5 genes were all transferred into 7 poplar plants. 2Microdissection and amplification of the chromosome arm 5S in a rice telo-tetrasomic显示文摘The rice aneuploids with telochromosomes are ideal genetic stocks for chromosome arm identification and microdissection. A rice telo-tetrasomic line with two extra short arms of chromosome 5 (5S) was used in the present study. The arms of 5S were microdissected from the prometaphase cells in mitosis and amplified with linker adaptor PCR (LA-PCR). 4Construction of single-chromosome DNA library from Lilium regale Wilson显示文摘A protocol of simple rapid microdissection of single-chromosome, amplification and cloning of its DNA from Lilium regale Wilson is described. Single-chromosome, microdissected by micromanipulator, was put into a 0 5 mL Eppendorf tube and digested with Sau3A, and then the Sau3A linker adaptors were ligated to the ends of DNA fragments. After 2 rounds of PCR amplification with one chain of linker adaptor as primer, the PCR products thus obtained have a length of 300-2 500 base pairs (bp) with predominant fragments at about 1 000 bp. Southern blot analysis confirmed that the PCR products originated from the genome of Lilium regale Wilson. By cloning the amplification products from the second round of PCR, single-chromosome DNA library was constructed, in which about as many as 100 000 recombinant clones were produced. A total number of 84 clones were analysed, and it was revealed that the inserts ranged in size from 300 to 1 800 bp, with an average of 780 bp. Compared with the methods described in other literature, this protocol, eliminating the need for enzymatic digestion and ligating micromanipulation of chromosomal DNA in nanoliter volumes, permits the efficient amplification of single chromosome (not tens of chromosomes as reported before) and the fragments (780 bp in average) cloned in this study are longer than those reported before (650 bp in average).Benyuan Dang Zanmin Hu Yihua Zhou Lihua Cui Lanlan Wang Liangcai Li Zhenghua Chen 1998Chinese Science Bulletin1998,43,5:2

7Chromosome microdissection by laser microbeam, chromosomal fragment isolation and amplification in vitro in barley (Hordeum vulgare L.)显示文摘A method for microdissection, isolation and amplification of plant chromosomal fragments using laser microbeam and a glass microneedle was established. Firstly, 7H chromosome of barley (Hordeum vulgare L.) was dissected by Nd: YAG laserbeam with suitable parameters and the fragment comprising a satellite was isolated with a glass microneedle which was fixed on a micromanipulator. Then, the chromosomal fragment DNA was amplified by LA_PCR (linker adaptor PCR) for two rounds. The size of the DNA fragments of PCR products varied from 500-3 000 bp and the PCR products originated from the genome of barley were verified by Southern hybridization. Compared with previous reports, there are some advantages in this research. The performance is easier, the dissection is more precise and the cost is low. It also permits efficient amplification with only one single chromosome fragment. 8Effectiveness of Post-Traumatic Growth Intervention to Promote Positive Post-Traumatic Traits in Chinese Breast Cancer Patients:A Randomized Controlled Trial显示文摘Objective:The purpose of this study was to evaluate the effectiveness of post-traumatic growth(PTG)model-based intervention to improve positive psychological traits in Chinese breast cancer patients.Design:A randomized control trial of a psychological group intervention based on PTG model.Methods:The Clinical Trial was registered on 17 August 2019 at Chinese Clinical Trials.gov with Registration number ChiCTR1900025264.A total of 92 patients with breast cancer were recruited.The participants were randomly assigned to the experimental group(n=46)and the control group(n=46).A six-session psychological group intervention based on PTG model was implemented in the experimental group,and a six-session health education was implemented in the control group.The outcomes weremeasured at baseline(pre-intervention),3 weeks,6 weeks after the intervention.The primary outcome was posttraumatic growth assessed by the Chinese version of the Post-Traumatic Growth Inventory(PTGI);Secondary outcomes included psychological resilience,family resilience,rumination,and self-disclosure.Results:A total of 87 patients with breast cancer completed this study,including 44 patients in the experimental group and 43 patients in the control group.There was no significant difference in baseline data of breast cancer patients between the two groups except for the treatment regimen(p>0.05).The two groups were compared after the intervention;the interaction effects between the total scores of posttraumatic growth,family resilience,and self-disclosure and the time term were statistically significant(p<0.05),indicating that the trend of change in total scores of post-traumatic growth,family resilience,and self-disclosure differed between the experimental and control groups over time,and the scores improved in the experimental group were significantly higher than those in the control group.The comparison of psychological resilience and total score of rumination at each time point was statistically significant(p<0.05),indicating that group intervention based on the PTG model could improve the psychological recovery ability and rumination level of the experimental group.Conclusion:The psychological group intervention based on the PTG model significantly improved post-traumatic growth,family resilience,and self-disclosure in patients with breast cancer.However,the impact on psychological resilience and rumination was relatively small.Long-term intervention is needed to further test the effect of the PTG model on psychological resilience and rumination.Cuirong Xu Han Yan Dawei Xu Guanjie Chen Qian Xu Kunpeng Li Yehao Rui Zanmin Song Neeraj Gill Jing Sun 2023International Journal of Mental Health Promotion2023,25,10:0
