1Single-cell RNA-seq data analysis on the receptor ACE2 expression reveals the potential risk of different human organs vulnerable to 2019-nCoV infection显示文摘It has been known that,the novel coronavirus,2019-nCoV,which is considered similar to SARS-CoV,invades human cells via the receptor angiotensin converting enzyme II(ACE2).Moreover,lung cells that have ACE2 expression may be the main target cells during 2019-nCoV infection.However,some patients also exhibit non-respiratory symptoms,such as kidney failure,implying that 2019-nCoV could also invade other organs.To construct a risk map of different human organs,we analyzed the single-cell RNA sequencing(scRNA-seq)datasets derived from major human physiological systems,including the respiratory,cardiovascular,digestive,and urinary systems.Through scRNA-seq data analyses,we identified the organs at risk,such as lung,heart,esophagus,kidney,bladder,and ileum,and located specific cell types(i.e.,type II alveolar cells(AT2),myocardial cells,proximal tubule cells of the kidney,ileum and esophagus epithelial cells,and bladder urothelial cells),which are vulnerable to 2019-nCoV infection.Based on the findings,we constructed a risk map indicating the vulnerability of different organs to 2019-nCoV infection.This study may provide potential clues for further investigation of the pathogenesis and route of 2019-nCoV infection.Xin Zou Ke Chen Jiawei Zou Peiyi Han Jie Hao Zeguang Han 2020Frontiers of Medicine2020,14,2:193
2Gene editing and its applications in biomedicine显示文摘The steady progress in genome editing, especially genome editing based on the use of clustered regularly interspaced short palindromic repeats(CRISPR) and programmable nucleases to make precise modifications to genetic material, has provided enormous opportunities to advance biomedical research and promote human health. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges. 4A transient multiphysics coupling method based on OpenFOAM for heat pipe cooled reactors显示文摘Differing from traditional pressurized water reactors(PWRs),heat pipe cooled reactors have the unique characteristics of fuel thermal expansion,expansion reactivity feedback,and thermal contact conductance.These reactors require a new multiphysics coupling method.In this paper,a transient coupling method based on OpenFOAM is proposed.The method considers power variation,thermal expansion,heat pipe operation,thermal contact conductance,and gap conductance.In particular,the reactivity feedback caused by working medium redistribution in a heat pipe is also preliminarily considered.A typical heat pipe cooled reactor KRUSTY(Kilowatt Reactor Using Stirling TechnologY)is chosen as the research object.Compared with experimental results of load following,the calculated results are in good agreement and show the validity of the proposed method.To discuss the self-adjusting capability of this type of reactor system,a hypothetical accident is simulated.It is assumed that at the beginning of this accident,loss of the heat sink occurs.After 1500 s of the transient process,the reactor system recovers immediately.During this hypothetical accident,the control rod is always out of the reactor core,and the reactor only relies on the reactivity feedback to regulate the fission power.According to the simulation,the peak temperature is only about 1112 K,which is far below the safety limit.As for system recovery,the reactor needs approximately 2500 s to return to a steady state and can realize effective power regulation by reactivity feedback.This study confirms the availability of this coupling method and that it can be an effective tool for the simulation of heat pipe cooled reactors.GUO YuChuan LI ZeGuang WANG Kan SU ZiLin 2022Science China(Technological Sciences)2022,65,1:4

7Significant flow velocity reduction at the intracranial aneurysm neck after endovascular treatment leads to favourable angiographic outcome:a prospective study显示文摘Background With widely usage of flow diverter in intracranial aneurysm treatment,some previously used predictors may not be effective in evaluating the recurrence risk.We aimed to comprehensively re-evaluate the predictors of intracranial aneurysm outcome with various endovascular treatment methods and devices.Methods This is a prospective registered study.We analysed 6-month and 18-month follow-up angiographic data from the prospective study.Data on patient demographics,aneurysm morphology and type of treatment were recorded.Patient-specific haemodynamic simulations were performed.An unfavourable angiographic outcome was defined as recurrence of aneurysm in cases with coiling or stent-assisted coiling,patency of aneurysm in cases with flow diverters or retreatment during follow-up.Results In total,165 patients(177 intracranial aneurysms)with at least one angiographic follow-up data were analysed.For the short-term(6-month)results,after univariate analysis,the demographic,morphological and treatment-related factors did not achieve significantly statistical differences.The reduction ratio(RR)of velocity at aneurysm neck after embolisation was significantly lower in the unfavourable angiographic group than the favourable angiographic outcome group(p=0.002).After the Cox regression analysis,the RR of velocity at aneurysm neck was the only independent factor associated with favourable angiographic outcome(OR 0.028;p=0.001)and had an acceptable area under the curve(0.714)with a clear cut-off value(46.14%).Similarly,for the analysis of midterm(18-month)results,the RR of velocity at the aneurysm neck was the only independent significant factor for the unfavourable angiographic outcome(OR 0.050;p=0.017).The area under the curve was 0.754 and the cut-off value was 48.20%.Conclusions The haemodynamics showed an independent effect on angiographic follow-up results and may provide helpful suggestions for clinical practice in the future.Zhongbin Tian Jian Liu Jay Kumar Wenqiang Li Yisen Zhang Ying Zhang Kun Wang Shengzhang Wang Zeguang Ren Xinjian Yang 2021Stroke & Vascular Neurology2021,6,3:1

14A study of spray granulation in vibrated fluidized bed with internal heating tubes显示文摘The granulation process in a vibratedfluidized bed with immersed horizontal heating tubes was studied in this paper,with small monoammonium phosphate particles as the initial particles and monoammonium phosphate solution sprayed on them.The concrete influences on the granulating performance of the immersed horizontal heating power,fluidizing gas velocity,vibration frequency and amplitude had been analyzed theoretically.The results show that appropriate vibration intensity is a preferred condition for the growth of partials,but thefine dust will become much more along with prolonged vibration.Increasing the heating power is beneficial to raise the growth rate.Particles will grow faster if thefluidizing gas velocity is higher,but this will cause morefine dust.The optimized condition of A=0.002 m,f=6.67 Hz,u=1.4 m/s was given after the analysis of the experimental results.In the end,a semiempirical formula was derived from the experiment data,and the calculation results show good agreement with the experimental data.Xiaoheng PAN Shichao YE Zeguang ZHANG Li ZHANG Xuejun ZHU 2009Frontiers of Chemical Science and Engineering2009,3,1:0

19Prognostic model for prostate cancer based on glycolysis-related genes and non-negative matrix factorization analysis显示文摘Background:Establishing an appropriate prognostic model for PCa is essential for its effective treatment.Glycolysis is a vital energy-harvesting mechanism for tumors.Developing a prognostic model for PCa based on glycolysis-related genes is novel and has great potential.Methods:First,gene expression and clinical data of PCa patients were downloaded from The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO),and glycolysis-related genes were obtained from the Molecular Signatures Database(MSigDB).Gene enrichment analysis was performed to verify that glycolysis functions were enriched in the genes we obtained,which were used in nonnegative matrix factorization(NMF)to identify clusters.The correlation between clusters and clinical features was discussed,and the differentially expressed genes(DEGs)between the two clusters were investigated.Based on the DEGs,we investigated the biological differences between clusters,including immune cell infiltration,mutation,tumor immune dysfunction and exclusion,immune function,and checkpoint genes.To establish the prognostic model,the genes were filtered based on univariable Cox regression,LASSO,and multivariable Cox regression.Kaplan–Meier analysis and receiver operating characteristic analysis validated the prognostic value of the model.A nomogram of the risk score calculated by the prognostic model and clinical characteristics was constructed to quantitatively estimate the survival probability for PCa patients in the clinical setting.Result:The genes obtained from MSigDB were enriched in glycolysis functions.Two clusters were identified by NMF analysis based on 272 glycolysis-related genes,and a prognostic model based on DEGs between the two clusters was finally established.The prognostic model consisted of LAMPS,SPRN,ATOH1,TANC1,ETV1,TDRD1,KLK14,MESP2,POSTN,CRIP2,NAT1,AKR7A3,PODXL,CARTPT,and PCDHGB2.All sample,training,and test cohorts from The Cancer Genome Atlas(TCGA)and the external validation cohort from GEO showed significant differences between the high-risk and low-risk groups.The area under the ROC curve showed great performance of this prognostic model.Conclusion:A prognostic model based on glycolysis-related genes was established,with great performance and potential significance to the clinical application.ZECHAO LU FUCAI TANG HAOBIN ZHOU ZEGUANG LU WANYAN CAI JIAHAO ZHANG ZHICHENG TANG YONGCHANG LAI ZHAOHUI HE 2023BIOCELL2023,47,2:0
