2Evaluation of a New Real-time PCR Assay for Detection of Mycoplasma Pneumoniae in Clinical Specimens显示文摘Objective To establish and evaluate a real-time PCR assay to detect Mycoplasma pneumoniae (M.pneumoniae) in clinical specimens.Methods By analysing the whole p1 gene sequence of 60 M.pneumoniae clinical isolates in Beijing of China,an optimized real-time PCR assay (MpP1) using p1 gene conserved region was designed.The specificity and sensitivity of this assay were evaluated and compared with other two reported assays (RepMp1 and Mp181) using 40 positive and 100 negative clinical specimens.Results The detection limit of the new assay was 8.1 fg (about 1~3CFU) M.pneumoniae DNA.The sensitivity of MpP1,RepMp1,and Mp181 assays appeared to be 100%,100%,and 85%,respectively.Conclusion MpP1 assay is suitable for the detection of M.pneumoniae in Chinese clinical specimens.ZHAO Fei CAO Bin HE Li Hua YIN Yu Dong TAO Xiao Xia SONG Shu Fan MENG Fan Liang ZHANG Jian Zhong 2012Biomedical and Environmental Sciences2012,25,1:11

4Research progress on the breeding of japonica super rice varieties in Jiangsu Province,China显示文摘In this study we report the results of a decade-long breeding program for japonica super rice made by Nanjing Branch of Chinese National Center for Rice Improvement in Jiangsu Academy of Agricultural Sciences. We concluded that selection of parents with good comprehensive traits and complementary advantages and disadvantages of both parents in the hybrid combination, and early selection of high heritability traits in earlier segregating generations could significantly improve the breeding efficiency. The use of closely-linked functional markers in pyramiding of multiple genes could greatly increase breeding efficiency, avoiding time-consuming and laborious steps that were used in traditional breeding program. 6Rapid Detection of Haemophilus influenzae and Haemophilus parainfluenzae in Nasopharyngeal Swabs by Multiplex PCR显示文摘Objective To establish multiplex PCR-based assays for detecting H.influenzae and H.parainfluenzae.And the PCR-based assays were applied to detect the carriage rates of H.influenzae and H.parainfluenzae in nasopharyngeal swab specimens which were collected from healthy children.Methods Multiplex primers for species-specific PCR were designed by using DNAstar soft based on the sequences of 16S rRNA genes from genus Haemophilus to detect H.influenzae and H.parainfluenzae.Results The sensitivity of the 16S rRNA PCR assay for detecting H.influenzae and H.parainfluenzae was 97.53% and 100% respectively,and the specificity was 95.89% and 96.63% respectively.Youden's Index on the ability to detect H.influenzae and H.parainfluenzae was 0.9342 and 0.9663 respectively.666 nasopharyngeal swab specimens were collected from healthy children.The detection rates of H.influenzae and H.parainfluenzae were 14.11% and 16.07% respectively by using isolation and culture methods.The detection rates of H.influenzae and H.parainfluenzae were 43.54% and 57.96% respectively by 16S rRNA PCR assays.The carriage rates of serotypes a,b,c,d,e,f and non-typeable isolates were 0% (0/666),0.15% (1/666),1.20% (8/666),0.15% (1/666),1.20% (8/666),1.80% (12/666),95.50% (636/666) respectively.Conclusion The multiplex PCR assays were very rapid,reliable and feasible methods for detection of H.influenzae and H.parainfluenzae in pharyngeal swab specimens which were compared to conventional isolation and culture methods.95.5% of H.influenzae strains in healthy children were nontypeable.The encapsulated or typable strains were mainly three serotypes which was c,e,and f serotype.TIAN Guo Zhong ZHANG Li Juan WANG Xiao Lei ZHANG Li LI Shu Feng GU Chang Mei SUN Jian CUI Bu Yun 2012Biomedical and Environmental Sciences2012,25,3:8
7Isolation,Characterization,and Phylogenetic Analysis of Two New Crimean-Congo Hemorrhagic Fever Virus Strains from the Northern Region of Xinjiang Province,China显示文摘Crimean-Congo hemorrhagic fever(CCHF)caused by the CCHF virus(CCHFV)is a tick-borne natural focal disease with a mortality rate of approximately 50%.CCHFV is widely prevalent in Africa,southern Asia,the Middle East,and southeast Europe.CCHF outbreaks have been reported previously in Xinjiang province,China,especially in its southern region.Epidemiological surveys conducted on ticks and animals have revealed the presence of CCHFV strains in ticks,rodents,and infected individuals from cities and counties in southern Xinjiang.Phylogenetic analyses revealed that the Chinese CCHFV strains belong to one genotype,based on complete sequences of the S segments of its negative-stranded RNA genome.The present study reports two new CCHFV strains isolated from Hyalomma asiaticum asiaticum ticks collected from Fukang City and Wujiaqu City in the northern region of Xinjiang.Viral characteristics and their evolutionary relationships were analyzed through metagenomic and reverse-transcription PCR analyses;these analyses indicated that the genotype of both strains was different from that of other Chinese strains.Furthermore,previous reports of CCHFV in Xinjiang were reviewed and phylogenetic analyses were performed.CCHFV was found to prevail in Fukang City in Junggar Basin for more than 20 years,and that Fukang City and Wujiaqu City are considered natural reservoirs of different genotypes of CCHFV strains.Our findings facilitate the understanding of CCHFV distribution in Xinjiang province and provide insights into the evolutionary relationships among Chinese CCHFV strains.Yanfang Zhang Shu Shen Yaohui Fang Jinliang Liu Zhengyuan Su Jinhao Liang Zhong Zhang Qiaoli Wu Cheng Wang Abulikemu Abudurexiti Zhihong Hu Yujiang Zhang Fei Deng 2018Virologica Sinica2018,33,1:8

14Rb-Sr chronological characteristics of Jurassic granitoid gravels in north margin of Dabie block,Central China显示文摘Granitoid gravels are discovered from alluvial fan conglomerates, the base of Jurassic Fenghuangtai Formation (J22), in the north margin of Dabie block and the south part of the Hefei Basin. These granitoid gravels make up 5%-10% of all gravels by volume percent. For the four components, K-feldspar, biotite, amphibole and whole rock divided from two selected granitoid gravel samples, their Rb-Sr isotopes are measured, which gives two formation ages of granitoids: (428±20) Ma and (430±54) Ma, and means that the Early Paleozoic granitoid intrusive rocks once existed in the Dabie Mountain provenances of Jurassic. 16GEOLOGICAL AND GEOCHEMICAL STUDY ON THE GENESIS OF THE FANKOU Pb-Zn DEPOSIT显示文摘The super large Pb Zn deposit at Fankou is very complex in metallogenic mechanism. Syngenetic brine sedimentation, hot submarine spring eruption—accumulation, syngenetic sedimentation—vadose brine replacement had been supposed as the genetic models before. Under these models, syngenetic mineralization should be dominant in ore formation. However, a series of geological and geochemical properties investigated by authors of this paper are contrary to these models. 19Phosphoribosyl-pyrophosphate synthetase 2 (PRPS2)depletion regulates spermatogenic cell apoptosis and is correlated with hypospermatogenesis显示文摘Phosphoribosyl-pyrophosphate synthetase 2(PRPS2)is a rate-limiting enzyme and plays an important role in purine and pyrimidine nucleotide synthesis.Recent studies report that PRPS2 is involved in male infertility.However,the role of PRPS2 in hypospermatogenesis is unknown.In this study,the relationship of PRPS2 with hypospermatogenesis and spermatogenic cell apoptosis was investigated.The results showed that PRPS2 depletion increased the number of apoptotic spermatogenic cells in vitro.PRPS2 was downregulated in a mouse model of hypospermatogenesis.When PRPS2 expression was knocked down in mouse testes,hypospermatogenesis and accelerated apoptosis of spermatogenic cells were noted.E2F transcription factor 1(E2F1)was confirmed as the target gene of PRPS2 and played a key role in cell apoptosis by regulating the P53/Bcl-xl/Bcl-2/Caspase 6/Caspase 9 apoptosis pathway.Therefore,these data indicate that PRPS2 depletion contributes to the apoptosis of spermatogenic cells and is associated with hypospermatogenesis,which may be helpful for the diagnosis of male infertility.Bin Lei Li-Xia Xie Shou-Bo Zhang Bo Wan Li-Ren Zhong Xu-Ming Zhou Xiang-Ming Mao Fang-Peng Shu 2020Asian Journal of Andrology2020,22,5:2
