| 1 | Engineering tropane biosynthetic pathway in Hyoscyamus niger hairy root cultures显示文摘Scopolamine is a pharmaceutically important tropane alkaloid extensively used as an anticholinergic agent.Here,we report the simultaneous introduction and overexpression of genes encoding the rate-limiting upstream enzyme pu-trescine N-methyltransferase(PMT)and the downstream enzyme hyoscyamine6β-hydroxylase(H6H)of scopolamine biosynthesis in transgenic henbane(Hyoscyamus niger)hairy root cultures.Transgenic hairy root lines expressing both pmt and h6h produced significantly higher(P<0.05)levels of scopolamine compared with the wild-type and transgenic lines har-boring a single gene(pmt or h6h).The best line(T 3 )produced411mg/liter scopolamine,which was over nine times more than that in the wild type(43mg/liter)and more than twice the amount in the highest scopolamine-producing h6h single-gene transgenic line H 11 (184mg/liter).To our knowledge,this is the highest scopolamine content achieved through genetic engi-neering of a plant.We conclude that transgenic plants harboring both pmt and h6h possessed an increased flux in the tropane alkaloid biosynthetic pathway that enhanced scopolamine yield,which was more efficient than plants harboring only one of the two genes.It seems that the pulling force of the downstream enzyme(the faucet enzyme)H6H plays a more important role in stimulating scopolamine accumulation in H.niger whereas the functioning of the upstream enzyme PMT is increased propor-tionally.This study provides an effective approach for large-scale commercial production of scopolamine by using hairy root culture systems as bioreactors. | ZHANGLei DINGRu-Xian CHAIYou-Rong MercedesBonfill ElisabeMoyano Kirsi-MarjaOksman-Caldentey XUTie-Feng PIYan WANGZi-Nan ZHANGHan-Ming KAIGuo-Yin LIAOZhi-Hua SUNXiao-Fen TANGKe-Xuan | 2004 | 第二军医大学学报2004,25,6: | 41 |
| 2 | Chronic idiopathic thrombocytopenic purpura in adult Chinese patients: a retrospective single centered analysis of 1791 cases显示文摘Background Adult chronic idiopathic thrombocytopenic purpura (ITP) is a common hematologic disease characterized by persistent thrombocytopenia. So far, there were only a few reports on adult Chinese patients with chronic ITP. This study aimed at defining the treatment outcome and prognostic factors for chronic ITP based on a large cohort of Chinese patients followed up for over 25 years at a single center Methods The medical records of 1791 patients aged 14 years or older who were diagnosed as having chronic ITP at our hospital from 1974 to 1999 were retrospectively analyzed Results The female to male ratio was 2∶1, with a median age of 34 years (ranging from 14 to 80 years), median platelet count of 38×10 9/L [range (1-99)×10 9/L], and median follow up of 36 months (range 1-220 months) Steroids were used in 689 patients, among them 209 (30.3%) achieved complete remission (CR) A splenectomy was performed in 124 patients, and response to steriod pre splenectomy was not available in 14 patients The CR rate after a splenectomy was lower in steroid nonresponders (29 of 90, 32.2%) than in those who relapsed after successful steroid treatment (12 of 20, 60.0%) ( P <0 05) In comparison with patients negative for antinuclear antibody (ANA), those who were ANA positive had similar responses to steroids, but a significantly shorter remission period after a splenectomy ( P <0 01) Conclusions Adult Chinese chronic ITP patients can have long term remission after steroid therapy and splenectomies Primary steroid refractoriness is a prognostic factor predicting poor subsequent response to a | LIHong-qiang ZHANGLei ZHAOHui JILin-xiang YANGRen-chi | 2005 | Chinese Medical Journal2005,,1: | 20 |

| 4 | Insights into the influence of temperature on the adsorption behavior of sodium oleate and its response to flotation of quartz显示文摘Temperature affects the flotation of quartz in the calcium/sodium oleate(Na OL)system,while there is a lack of understanding of its potential mechanism.Therefore,in this work,the flotation response of quartz to temperature was investigated via micro-flotation experiments,interface property analyses,and theoretical calculations.Flotation results demonstrated that increasing temperature contributed to higher flotation recovery of quartz,which enhanced the removal of quartz from hematite.Surface tension results revealed that higher temperatures lowered the critical micelle concentration(CMC)and surface tension of the Na OL solution,and thus enhanced its surface activity.Solution chemistry calculations and X-ray photoelectron spectroscopy(XPS)measurements confirmed that the increased content of Ca(OH)+achieved by increasing temperatures enhanced the adsorption amounts of calcium species(acting as activation sites)on the quartz surface.Dynamic light scattering(DLS)measurements verified that the association degree of RCOOàto form(RCOO)22àwas strengthened.Furthermore,adsorption density measurements and molecular dynamics(MD)simulations confirmed that increasing the temperature facilitated Na OL adsorption toward the surface of the quartz,which was attributed to the stronger interaction between Na OL and the calcium-activated quartz surface at higher temperatures.As a result,quartz flotation was improved by increasing temperatures.Accordingly,a possible adsorption model was proposed. | Shaohang Cao Wanzhong Yin Bin Yang Zhanglei Zhu Haoran Sun Qiuyue Sheng Keqiang Chen | 2022 | International Journal of Mining Science and Technology2022,32,2: | 4 |
| 5 | CO2 emissions and their bearing on China's economic development: the long view显示文摘Greenhouse-gas (GHG) emissions in China have aroused much interest, and not least in recent evidence of their reduction. Our intent is to place that reduction in a larger context, that of the process of industrialization. A lengthy time perspective is combined with a cross-sectional approachChina plus five other countries-and addressed through two general models. The findings are salutary. First, they suggest that a diversified economic structure is consistent with diminished intensity in energy use. Secondly, and the obverse of the first, they imply that a diversified energy structure promotes reductions in CO2 emissions. Finally, one is led inevitably to the conclusion that, together, the findings point to a path for countries to transform their economies while at the same time undertaking to drastically moderate their energy use, switching from a pattern of heavy carbon emissions to one in which lighter carbon emissions prevail. | 8 | Application of Cerebellar Model Articulation Controller (CMAC) with a Modified Algorithm in Monitoring Machine Performance Degradation显示文摘On the basis of CMAC-PDM (Pattern Discrimination Model), a novel algorithm of CMAC for monitoring machine degradation is proposed in this paper. The output of CMAC with the novel algorithm represents the state of a machine and PDM is not needed. The principle was explained by analyzing the modified mapping process of CMAC. The novel CMAC is applied to a tool condition monitoring system and two methodologies (novel CMAC and CMAC-PDM) are compared. | 10 | The VER2 promoter contains repeated sequences and requires vernalization for its activity in winter wheat (Triticum aestivum L.)显示文摘Previously, we isolated a vernalization-related gene, VER2, from winter wheat (Triticum aestivum L.) and its expression was restricted in the immature leaves of vernal-ized wheat seedlings. To further investigate the regulation of VER2 expression and the function of its promoter, we iso-lated a 41.7 kb genomic clone containing VER2 gene from a transformation-competent artificial chromosome (TAC) li-brary of wheat (Triticum aestivum-Haynaldia villosa). The sequence analysis showed that there were eleven predicted genes in the TAC. The exons of gene 3 corresponded to the cDNA sequence of VER2 gene. Analysis of VER2 promoter structure showed that there were three small repeat se-quences divided by two large repeat sequences. The putative response elements, such as abscisic acid response elements (ABRE), MeJA-response elements (Me-JARE), low-tempe- rature response elements (LTR), endosperm expression ele-ments, MYB binding sites and similar elements to GA re-sponse elements (GARE), were involved in the VER2 pro-moter region. Construct containing the VER2 promoter (-5895 to +73) driving GFP reporter gene was bombarded into vernalized or non-vernalized immature leaves in wheat. The vernalized immature leaves showed bright green fluo-rescence after incubation for 24 h, however, the green fluo-rescence was not observed in the non-vernalization leaves under the same condition.
