| 1 | The Glutamatergic Postrhinal Cortex–Ventrolateral Orbitofrontal Cortex Pathway Regulates Spatial Memory Retrieval显示文摘A deficit in spatial memory has been taken as an early predictor of Alzheimer’s disease(AD) or mild cognitive impairment(MCI). The uncinate fasciculus(UF) is a long-range white-matter tract that connects the anterior temporal lobe with the orbitofrontal cortex(OFC)in primates. Previous studies have shown that the UF impairment associated with spatial memory deficits may be an important pathological change in aging and AD, but its exact role in spatial memory is not well understood. The pathway arising from the postrhinal cortex(POR) and projecting to the ventrolateral orbitofrontal cortex(vlOFC)performs most of the functions of the UF in rodents.Although the literature suggests an association between spatial memory and the regions connected by the POR–vlOFC pathway, the function of the pathway in spatialmemory is relatively unknown. To further illuminate the function of the UF in spatial memory, we dissected the POR–vlOFC pathway in mice. We determined that the POR–vlOFC pathway is a glutamatergic structure, and that glutamatergic neurons in the POR regulate spatial memory retrieval. We also demonstrated that the POR–vlOFC pathway specifically transmits spatial information to participate in memory retrieval. | 2 | The biological effect of recombinant humanized collagen on damaged skin induced by UV-photoaging:An in vivo study显示文摘The application of medical devices to repair skin damage is clinically accepted and natural polymer enjoys an important role in this field,such as collagen or hyaluronic acid,etc.However,the biosafety and efficacy of these implants are still challenged.In this study,a skin damage animal model was prepared by UV-photoaging and recombinant humanized type Ⅲ collagen(rhCol Ⅲ)was applied as a bioactive material to implant in vivo to study its biological effect,comparing with saline and uncrosslinked hyaluronic acid(HA).Animal skin conditions were non-invasively and dynamically monitored during the 8 weeks experiment.Histological observation,specific gene expression and other molecular biological methods were applied by the end of the animal experiment.The results indicated that rhCol Ⅲ could alleviate the skin photoaging caused by UV radiation,including reduce the thickening of epidermis and dermis,increase the secretion of Collagen Ⅰ(Col Ⅰ)and Collagen Ⅲ(Col Ⅲ)and remodel of extracellular matrix(ECM).Although the cell-material interaction and mechanism need more investigation,the effect of rhCol Ⅲ on damaged skin was discussed from influence on cells,reconstruction of ECM,and stimulus of small biological molecules based on current results.In conclusion,our findings provided rigorous biosafety information of rhCol Ⅲ and approved its potential in skin repair and regeneration.Although enormous efforts still need to be made to achieve successful translation from bench to clinic,the recombinant humanized collagen showed superiorities from both safety and efficacy aspects. | Jing Wang He Qiu Yang Xu Yongli Gao Peijie Tan Rui Zhao Zhanhong Liu Yajun Tang Xiangdong Zhu Chongyun Bao Hang Wang Hai Lin Xingdong Zhang | 2022 | Bioactive Materials2022,7,5: | 7 |
| 3 | Dynamic microstructure evolution and mechanical properties of dilute Mg-Al-Ca-Mn alloy during hot rolling显示文摘The dynamic microstructure and texture of dilute Mg-0.50Al-0.71Ca-0.33Mn(wt.%) during hot rolling at two slab temperatures were investigated by electron backscattered diffraction(EBSD) and high resolution transmission electron microscopy(HRTEM). The results show that the development of the rolling microstructures is to first form {10–12} extension twins in the original grains, thereby forming the extension-twinned regions, and then to further form {10–11}-{10–12} double twins and kinks in the extension-twinned regions, and finally to form continuous dynamic recrystallized(continuous DRXed)grains in the double twins and the cross parts of the shear-deformed coarse extension-twinned regions.These extension twins, double twins and kinks show a decisive effect on the formation of rolling texture.The number of {10–11}-{10–12} double twins and resultant the continuous DRX process are strongly affected by the rolling slab temperature and the reduction thickness per pass. By optimizing the rolling conditions, texture and microstructures of the multi-pass rolled Mg-Al-Ca-Mn alloy sheets are successfully modified. | 4 | Impaired Parahippocampal Gyrus-Orbitofrontal Cortex Circuit Associated with Visuospatial Memory Deficit as a Potential Biomarker and Interventional Approach for Alzheimer Disease显示文摘The parahippocampal gyrus-orbitofrontal cortex(PHG-OFC)circuit in humans is homologous to the postrhinal cortex(POR)-ventral lateral orbitofrontal cortex(vlOFC)circuit in rodents.Both are associated with visuospatial malfunctions in Alzheimer’s disease(AD).However,the underlying mechanisms remain to be elucidated.In this study,we explored the relationship between an impaired POR-vlOFC circuit and visuospatial memory deficits through retrograde tracing and in vivo local field potential recordings in 5XFAD mice,and investigated alterations of the PHG-OFC circuit by multi-domain magnetic resonance imaging(MRI)in patients on the AD spectrum.We demonstrated that an impaired glutamatergic POR-vlOFC circuit resulted in deficient visuospatial memory in 5XFAD mice.Moreover,MRI measurements of the PHG-OFC circuit had an accuracy of 77.33%for the classification of amnestic mild cognitive impairment converters versus non-converters.Thus,the PHG-OFC circuit explains the neuroanatomical basis of visuospatial memory deficits in AD,thereby providing a potential predictor for AD progression and a promising interventional approach for AD. | Lin Zhu Zan Wang Zhanhong Du Xinyang Qi Hao Shu Duan Liu Fan Su Qing Ye Xuemei Liu Zheng Zhou Yongqiang Tang Ru Song Xiaobin Wang Li Lin Shijiang Li Ying Han Liping Wang Zhijun Zhang | 2020 | Neuroscience Bulletin2020,36,8: | 2 |
| 5 | Canine ACL reconstruction with an injectable hydroxyapatite/collagen paste for accelerated healing of tendon-bone interface显示文摘Healing of an anterior cruciate ligament(ACL)autologous graft in a bone tunnel occurs through the formation of fibrovascular scar tissue,which is structurally and compositionally inferior to normal fibrocartilaginous insertion and thus may increase the reconstruction failure and the rate of failure recurrence.In this study,an injectable hydroxyapatite/type I collagen(HAp/Col I)paste was developed to construct a suitable local microenvironment to accelerate the healing of bone-tendon interface.Physicochemical characterization demonstrated that the HAp/Col I paste was injectable,uniform and stable.The in vitro cell culture illustrated that the paste could promote MC3T3-E1 cells proliferation and osteogenic expression.The results of a canine ACL reconstruction study showed that the reconstructive ACL had similar texture and color as the native ACL.The average width of the tunnel,total bone volume,bone volume/tissue volume and trabecular number acquired from micro-CT analysis suggested that the healing of tendon-bone interface in experimental group was better than that in control group.The biomechanical test showed the maximal loads in experimental group achieved approximately half of native ACL’s maximal load at 24 weeks.According to histological examination,Sharpey fibers could be observed as early as 12 weeks postoperatively while a typical four-layer transitional structure of insertion site was regenerated at 48 weeks in the experimental group.The injectable HAp/Col I paste provided a biomimetic scaffold and microenvironment for early cell attachment and proliferation,further osteogenic expression and extracellular matrix deposition,and in vivo structural and functional regeneration of the tendon-bone interface. | Qingsong Jiang Liren Wang Zhanhong Liu Jinlei Su Yajun Tang Peijie Tan Xiangdong Zhu Kai Zhang Xing Ma Jia Jiang Jinzhong Zhao Hai Lin Xingdong Zhang | 2023 | Bioactive Materials2023,,2: | 1 |
| 6 | Characterization of recombinant humanized collagen type III and its influence on cell behavior and phenotype显示文摘Collagen made a tremendous impact in the field of regenerative medicine as a bioactive material.For decades,collagen has been used not only as a scaffolding material but also as an active component in regulating cells’biological behavior and phenotype.However,animal-derived collagen as a major source suffered from problems of immunogenicity,risk of viral infection,and the unclear relationship between bioactive sequence and function.Recombinant humanized collagen(rhCol)provided alternatives for regenerative medicine with more controllable risks.However,the characterization of rhCol and the interaction between rhCol and cells still need further investigation,including cell behavior and phenotype.The current study preliminarily demonstrated that recombinant humanized collagen type III(rhCol III)conformed to the theoretical amino acid sequence and had an advanced structure resembling bovine collagen.Furthermore,rhCol III could facilitate basal biological behaviors of human skin fibroblasts,such as adhesion,proliferation and migration.rhCol III was beneficial for some extracellular matrix-expressing cell phenotypes.The study would shed light on the mechanism research of rhCol and cell interactions and further understanding of effectiveness in tissue regeneration. | Jing Wang Hong Hu Jian Wang He Qiu Yongli Gao Yang Xu Zhanhong Liu Yajun Tang Lu Song John Ramshaw Hai Lin Xingdong Zhang | 2022 | Journal of Leather Science and Engineering2022,4,1: | 0 |
