1Rescue of premature aging defects in Cockayne syndrome stem cells by CRISPR/Cas9-mediated gene correction显示文摘Cockayne syndrome(CS)is a rare autosomal recessive inherited disorder characterized by a variety of clinical features,including increased sensitivity to sunlight,progressive neurological abnormalities,and the appearance of premature aging.However,the pathogenesis of CS remains unclear due to the limitations of current disease models.Here,we generate integration-free induced pluripotent stem cells(iPSCs)from fibroblasts from a CS patient bearing mutations in CSB/ERCC6 gene and further derive isogenic genecorrected CS-iPSCs(GC-iPSCs)using the CRISPR/Cas9 system.CS-associated phenotypic defects are recapitulated in CS-iPSC-derived mesenchymal stem cells(MSCs)and neural stem cells(NSCs),both of which display increased susceptibility to DNA damage stress.Premature aging defects in CS-MSCs are rescued by the targeted correction of mutant ERCC6.We next map the transcriptomic landscapes in CS-iPSCs and GC-iPSCs and their somatic stem cell derivatives(MSCs and NSCs)in the absence or presence of ultraviolet(UV)and replicative stresses,revealing that defects in DNA repair account for CS pathologies.Moreover,we generate autologous GC-MSCs free of pathogenic mutation under a cGMP(Current Good Manufacturing Practice)-compliant condition,which hold potential for use as improved biomaterials for future stem cell replacement therapy for CS.Collectively,our models demonstrate novel disease features and molecular mechanisms and lay a foundation for the development of novel therapeutic strategies to treat CS.Si Wang Zheying Min Qianzhao Ji Lingling Geng Yao Su Zunpeng Liu Huifang Hu Lixia Wang Weiqi Zhang Keiichiro Suzuiki Yu Huang Puyao Zhang Tie-Shan Tang Jing Qu Yang Yu Guang-Hui Liu Jie Qiao 2020Protein & Cell2020,11,1:7
2Characteristics and Outcome of Exertional Heatstroke Patients Complicated by Acute Hepatic Injury:A Cohort Study显示文摘Background and Aims:Exertional heatstroke(EHS)is associated with strenuous physical activity in hot environments.The present study aimed to investigate dynamic changes of hepatic function indices in EHS patients and determine risk factors for death.Methods:This single-center retrospective cohort study considered all patients with EHS admitted to the intensive care unit at the General Hospital of Southern Theater Command of PLA from October 2008 to May 2019.Data on general characteristics,organ function parameters,and the 90-day outcome of enrolled patients were collected.Hepatic indices were collected dynamically,and patients with acute hepatic injury(AHI)were identified by plasma total bilirubin(TBIL)≥34.2μmol/L and an international normalized ratio≥1.5,or with any grade of hepatic encephalopathy.Results:In patients who survived,TBIL,alanine aminotransferase and aspartate aminotransferase were increased at 24 h,peaked at 2–3 days,and began to decrease at 5 days.In non-survivors,TBIL continuously increased post-admission.The area under the receiver operating characteristic curve for the prediction of mortality based on sequential organ failure assessment(SOFA)scores was 89.8%,and the optimal cutoff value was 7.5.Myocardial injury and infection were identified as independent risk factors for death in EHS patients with AHI.Conclusions:In EHS patients,hepatic dysfunction usually occurred within 24 h.Patients with AHI had more severe clinical conditions,and significantly increased 90-day mortality rates.SOFA scores over 7.5,complicated with myocardial injury or infection,were found to be risk factors for death in EHS patients with AHI.Jingjing Ji Jinghua Gao Conglin Wang Leifang Ouyang Zheying Liu Zhifeng Liu 2021Journal of Clinical and Translational Hepatology2021,9,5:6

5Neuroendocrine characteristics of induced pluripotent stem cells from polycystic ovary syndrome women显示文摘Dear Editor, Polycystic ovary syn drome (PCOS) is a comm on female reproductive endocrinopathy that afflicts up to 10%-15% of women in reproductive age worldwide (Nestler, 2016). Women with PCOS exhibit hyperandrogenism, intermittent/ absent menstrual cycles, and polycystic ovaries on ultrasound (Rotterdam, 2004). The pathophysiology of PCOS extends beyond infertility and hirsutism to hypothalamic neuroendocrine dysfunotion (Goodarzi et al., 2011). Most wome n with PCOS exhibit in creased luteinizing horm one (LH) levels, resulting from high-frequency gonadotropin-releasing hormone (GnRH) secretion (Cimino et al., 2016). Pren ata I testostero ne (T) treatment in sheep results in disrupted steroid feedback on gonadotropin release, which in creases pituitary sen sitivity to GnRH and subseque ntly leads to LH hypersecretion (Sullivan and Moenter, 2004;Cardoso et al., 2016). A recent study shows that GnRHdependent LH pulsatility and secretion are elevated by anti- Mullerian hormone (AMH) in PCOS disease. The increased prenatal AMH reprograms fetus and induces PCOS in adults (Tata et al., 2018). Furthermore, the androgen receptor (AR) plays a role in hyperandrogenism and ovarian folliculoge esis in PCOS (Wang et al., 2015;Abbott, 2017). However, the disease mechanism behind PCOS remains unclear, and current management focuses on treating the symptoms but not the mechanism (Chen et al., 2016;Shi et al., 2018). 132D antimonene-integrated composite nanomedicine for augmented low-temperature photonic tumor hyperthermia by reversing cell thermoresistance显示文摘The overexpression of heat shock proteins(HSPs)in tumor cells can activate inherent defense mechanisms during hyperthermia-based treatments,inducing thermoresistance and thus diminishing the treatment efficacy.Here,we report a distinct“non-inhibitor involvement”strategy to address this issue through engineering a calcium-based nanocatalyst(G/A@CaCO_(3)-PEG).The constructed nanocatalyst consists of calcium carbonate(CaCO_(3))-supported glucose oxidase(GOD)and 2D antimonene quantum dots(AQDs),with further surface modification by lipid bilayers and polyethylene glycol(PEG).The engineered G/A@CaCO_(3)-PEG nanocatalyst features prolonged blood circulation,which is stable at neutral pH but rapidly degrades under mildly acidic tumor microenvironment,resulting in rapid release of drug cargo in the tumor microenvironment.The integrated GOD effectively catalyzes the depletion of glucose for reducing the supplies of adenosine triphosphate(ATP)and subsequent down-regulation of HSP expression.This effect then augments the therapeutic efficacy of photothermal hyperthermia induced by 2D AQDs upon irradiation with near-infrared light as assisted by reversing the cancer cells’thermoresistance.Consequently,synergistic antineoplastic effects can be achieved via low-temperature photothermal therapy.Systematic in vitro and in vivo evaluations have demonstrated that G/A@CaCO_(3)-PEG nanocatalysts feature potent antitumor activity with a high tumor-inhibition rate(83.92%).This work thus paves an effective way for augmenting the hyperthermia-based tumor treatments via restriction of the ATP supply.Jianrong Wu Xiaojun Cai Gareth R.Williams Zheying Meng Weijuan Zou Li Yao Bing Hu Yu Chen Yuanyi Zheng 2022Bioactive Materials2022,7,4:0

15Hsa_Circ_0000826 inhibits the proliferation of colorectal cancer by targeting AUF1显示文摘Many circular RNAs(circRNAs)are reported to be abnormally expressed during the progression of various tumors,and these circRNAs can be used as anti-tumor targets.Therefore,it is important to identify circRNAs that can be used effectively for the clinical diagnosis and treatment of colorectal cancer(CRC).Here,we report that hsa_Circ_0000826(Circ_0000826),a circ RNA with significantly reduced expression level in CRC tissues,is associated with a poor prognosis in patients.The silencing of Circ_0000826 promotes the proliferation of CRC cells.Conversely,the overexpression of Circ_0000826 restricted CRC cell proliferation both in vitro and in vivo.Furthermore,Circ_0000826 could target AU-rich element RNA-binding protein 1(AUF1).AUF1,known as heterogeneous nuclear ribonucleoprotein D(hnRNP D),could bind to the c-MYC 3’-UTR and promote c-MYC expression.When Circ_0000826 binds to AUF1,it competitively inhibits the binding of AUF1 to the c-MYC 3’-UTR,which inhibits the c-MYC expression and cell proliferation.These results provide novel insights into the functional mechanism of Circ_0000826 action in CRC progression and indicate its potential use as a therapeutic target in CRC.Zheying Zhang Wenyan Fan Qingzu Gao Yifei Han Jingyu Ma Wuji Gao Yuhan Hu Huifang Zhu Rui Yang Haijun Wang Baoshun Du Zuoyang Zhang Jiateng Zhong 2023Journal of Genetics and Genomics2023,50,3:0
