1Activity of the Lenglongling fault system and seismotectonics of the 2016 MS6.4 Menyuan earthquake显示文摘The MS6.4 Menyuan earthquake occurred on the northern side of the Lenglongling fault(LLLF) in the mid-western of the Qilian-Haiyuan fault zone on January 21, 2016. The earthquake epicenter was distant from the Minle-Damaying and Huangcheng-Shuangta faults, eastern of the Northern Qilian Shan fault zone. A near northwest-striking rupture plane intersects the two faults at a certain angle. The focal mechanism solution shows that this was a thrust-type earthquake, slightly different from the strike-slip movement with a thrust component of the LLLF. Field geological mapping, tectonic geomorphology analysis, trench excavation and 14 C dating reveal that(1) the LLLF has been obviously active since the Holocene, and may behave with characteristic slip behavior and produce M_W7.3–7.5 earthquakes;(2) the LLLF appears as a flower structure in terms of structure style, and dips NNE at a steep angle; and(3) the most recent earthquake event occurred after 1815–1065 a BP. An associated fault, the Northern Lenglongling fault(NLLLF), is located at the northwestern end of the LLLF. Consequently, the NLLLF was continually subject to tectonic pushing effects from the left-lateral shear at the end of the LLLF, and, accordingly, it bent and rotated outward tectonically.Subsequently, the fault deviated from the dominant rupture azimuth and activity weakened. In the late Quaternary, it behaved as a thrust fault with no obvious deformation at the surface. This is indicated by the arc shape, with a micro-protrusion northeastward,and no geologic or geomorphic signs of surface rupturing since the late Quaternary. However, such faults could still rupture at depth, producing moderate-strong earthquakes. The geometric and kinematic properties of the NLLLF are in good agreement with the occurrence and kinematic properties of nodal plane 2, and with the distribution characteristics of the aftershocks and seismic intensity. Therefore, the NLLLF is a more suitable seismogenic structure for the MS 6.4 Menyuan earthquake. In addition, the thrust movement of the NLLLF accommodates subsequent movement of the LLLF. 2the National Natural Science Foundation of China(41877029,41961130383);Royal SocietyNewton Advanced Fellowship(NAF\R1\191017);Wuhan Science and Technology Bureau(2019020701011469).显示文摘Due to the tremendous diversity of microbial organisms in topsoil,the estimation of saturated richness in a belowground ecosystem is still challenging.Here,we intensively surveyed the 16S rRNA gene in four 1 m2 sampling quadrats in a typical grassland,with 141 biological or technical replicates generating over 11 million sequences per quadrat.Through these massive data sets and using both non-asymptotic extrapolation and non-parametric asymptotic approaches,results revealed that roughly 15919±193,27193±1076 and 56985±2347 prokaryotic species inhabited in 1 m2 topsoil,classifying by DADA2,UPARSE(97%cutoff)and Deblur,respectively,and suggested a huge difference among these clustering tools.Nearly 500000 sequences were required to catch 50%species in 1 m2,while any estimator based on 500000 sequences would still lose about a third of total richness.Insufficient sequencing depth will greatly underestimate both observed and estimated richness.At least~911000,~3461000,and~1878000 sequences were needed for DADA2,UPARSE,and Deblur,respectively,to catch 80%species in 1 m2 topsoil,and the numbers of sequences would be nearly twice to three times on this basis to cover 90%richness.In contrast,α-diversity indexes characterized by higher order of Hill numbers,including Shannon entropy and inverse Simpson index,reached saturation with fewer than 100000 sequences,suggesting sequencing depth could be varied greatly when focusing on exploring differentα-diversity characteristics of a microbial community.Our findings were fundamental for microbial studies that provided benchmarks for the extending surveys in large scales of terrestrial ecosystems.Shuzhen Li Xiongfeng Du Kai Feng Yueni Wu Qing He Zhujun Wang Yangying Liu Danrui Wang Xi Peng Zhaojing Zhang Arthur Escalas Yuanyuan Qu Ye Deng 2022Soil Ecology Letters2022,4,3:1

4In vivo and in situ real-time fluorescence imaging of peripheral nerves in the NIR-II window显示文摘The peripheral nervous system(PNS)is essential for performing and maintaining various motor and sensory functions.Abnormalities can lead to a series of peripheral neurological conditions,such as paraesthesia,pain,or spasms,which are debilitating and lowering the quality of life.Thecurrent guidelines for diagnosis rely predominantly on clinical symptoms resulting from PNS dysfunction,which occur already at an advancedstage.There are currently no effective methods that visually reflect the extent of peripheral neuropathy.In our study,we present a novel in vivoand in situ real-time imaging of peripheral nerves based on the second near-infrared window(NIR-II)fluoresce nee.In NIR-II system,lead sulfidequa ntum dots(PbS Qds)with NIR-II fluoresce nee specifically bound to motor neuro rvspecific protein agrin,acting as image con trast.In micemodel,peripheral nerves were visible as soon as after 2 h post injection.We provide evidenee for the efficacy of this approach,which allows todirectly dem on strate peripheral nerves,their structure,and pote ntial damagesites and degree.Furthermore,our products were of goodbiocompatibility,while the n eural fluoresce nee signal was solid,bright and stable for 4 h in vivo.Thus,overall,our results suggest that NIR-II isan effective new method for direct imaging of peripheral nerves in vivo,opening new horizons on early,improved and more precise,targeteddiag no sis.A resulti ng more rapid installatio n of perso nalized therapy facilitates a better prognosis of clinical peripheral neuropathy.Zhujun Feng Yimeng Yang Jian Zhang Kan Wang Yunxia Li Heng Xu Zhen Wang Ewelina Biskup Shixian Dong Xing Yang Yuefeng Hao Jun Chen Yan Wo 2019Nano Research2019,12,12:1

7MDSNet:a multiscale decoupled supervision network for semantic segmentation of remote sensing images显示文摘Recent deep-learning successes have led to a new wave of semantic segmentation in remote sensing(RS)applications.However,most approaches rarely distinguish the role of the body and edge of RS ground objects;thus,our understanding of these semantic parts has been frustrated by the lack of detailed geometry and appearance.Here we present a multiscale decoupled supervision network for RS semantic segmentation.Our proposed framework extends a densely supervised encoder-decoder network with a feature decoupling module that can decouple semantic features with different scales into distinct body and edge components.We further conduct multiscale supervision of the original and decoupled body and edge features to enhance inner consistency and spatial boundaries in remote sensing image(RSl)ground objects,enabling new segmentation designs and semantic components that can learn to perform multiscale geometry,and appearance.Our results outperform the previous algorithm and are robust to different datasets.These results demonstrate that decoupled supervision is an effective solution to semantic segmentation tasks of RS images.Jiangfan Feng Panyu Chen Zhujun Gu Maimai Zeng Wei Zheng 2023International Journal of Digital Earth2023,16,1:0
8Soil microbiome mediated nutrients decline during forest degradation process显示文摘Degradation succession in forests is an important and serious land use/cover change problem in ecology,and during these processes soil microbial communities mediate the recycling of most important nutrients.To reveal the effect of degradation succession processes on soil microbial community diversity,structure,and species interrelationships,we collected abundant samples(21 per vegetation type)in broad-leaved forest,coniferous forest,and meadow to observe the microbial community dynamics.The results showed that diversity and structure of soil prokaryotic and fungal communities responded differently to different forest degradation processes,diversity of soil microbial communities increased during degradation processes.Soil microbial communities abundance changes may indicate that prokaryotic communities showed a living strategies change as an ecological adaption to harsh conditions during forest degradation process.While for fungal communities,their abundance changes may indicate that environmental selection pressure and plant selectivity during forest degradation process.Changes in soil prokaryotic communities and fungal communities were both correlated with soil carbon and nitrogen loss.The soil microbial interaction network analysis indicated more complex species interrelationships formed due to the loss of soil nutrients during degradation succession processes,suggesting soil microbial communities might form more complex and stable networks to resist the external disturbance of soil nutrient loss.All results suggested soil microorganisms,including bacteria,archaea and fungi,all involved in the soil nutrient decline during the forest degradation process.Yangying Liu Shang Wang Zhujun Wang Zhaojing Zhang Huayu Qin Ziyan Wei Kai Feng Shuzhen Li Yueni Wu Huaqun Yin Hui Li Ye Deng 2019Soil Ecology Letters2019,1,1:0
9Hexyl-pentynoic acid serves as a novel radiosensitizer for breast cancer by inhibiting UCHL3-dependent Rad51 deubiquitination显示文摘Objective:To investigate the effects and underlying mechanism of 2-hexyl-4-pentynoic acid(HPTA),a derivative of valproic acid(VPA),on radiotherapy in breast cancer.Methods:MCF7 cells and 7,12-dimethylbenz-[α]-anthracene(DMBA)-induced transformed human normal breast cells(MCF10A–DMBA cells)were irradiated with 8 Gy X-rays.For both cells there were four groups:control,valproic acid(VPA)/HPTA,IR,and VPA/HPTA+IR groups.MTT and clonogenic survival assays were performed to assess cell proliferation,and comet assay was performed to evaluate DNA damage.Protein expression ofγH2AX,53BP1,Rad51,and BRCA1 was examined via immunofluorescence and immunoblotting.Cycloheximide chase and ubiquitination experiments were conducted to determine Rad51 ubiquitination.In vivo experiments involved a rat model of DMBA-induced breast cancer,with four fractionated doses of 2 Gy.Tumor tissue pathological changes andγH2AX,Rad51,and UCHL3 expression levels were measured by hematoxylin-eosin staining,immunohistochemistry,and immunoblotting.Results:Compared with the IR group,15μmol/L HPTA reduced the cell proliferation ability of irradiated MCF7 cells(t=2.16,P<0.05).The VPA/HPTA+IR group exhibited significantly increased DNA double-strand breaks relative to those in the IR group(VPA+IR vs.IR,t=13.37,P<0.05;HPTA+IR vs.IR,t=8.48,P<0.05).Immunofluorescence and immunoblotting experiments demonstrated that the VPA/HPTA+IR group displayed signifi-cantly increased cell foci formation,γH2AX and 53BP1 protein expression levels compared to the IR group[(γH2AX:VPA+IR vs.IR,t=8.88,P<0.05;HPTA+IR vs.IR,t=8.90,P<0.05),(53BP1,VPA+IR vs.IR,t=5.73,P<0.05;HPTA+IR vs.IR,t=6.40,P<0.05)].Further,Rad51 expression was downregulated(VPA+IR vs.IR,t=3.12,P<0.05;HPTA+IR vs.IR,t=2.70,P<0.05),and Rad51 inhibition effectively counteracted HPTA-induced radiosensitization.Ubiquitination detection further verified that HPTA inhibits Rad51 expression via UCHL3-dependent Rad51 deubiquitination.In vivo study results showed that 20 mg/kg HPTA significantly enhanced the radiosensitivity of breast tumors in rats by inhibiting Rad51 expression.Conclusions:HPTA is a highly effective radiosensitizer that enhances the radiotherapeutic efficacy of breast cancer treatment through UCHL3-dependent deubiquitination of Rad51.Zuchao Cai David Lim Beidi Jia Guochao Liu Wenwen Ding Zhendong Wang Zhujun Tian Junxuan Peng Fengmei Zhang Chao Dong Zhihui Feng 2023Radiation Medicine and Protection2023,4,4:0
