| 2 | Diode-pumped power scalable Kerr-lens mode-locked Yb:CYA laser显示文摘Stable 68 fs pulses with the average power of 1.5 W is directly generated from a multimode diode-pumped Kerr-lens mode-locked Yb:CYA laser by separating the gain medium and Kerr medium. The repetition rate is about 50 MHz, resulting in a single pulse energy of 30 n J and a peak power of 0.44 MW. To the best of our knowledge, this is the highest single pulse energy ever produced from a mode-locked Yb:CYA oscillator. Our experimental results show that Yb:CYA crystal is an excellent candidate for multiwatt, sub-100 fs pulse generation in diode-pumped all-solid-state lasers. | 3 | Characterization of two monoclonal antibodies, 38F10 and 44D11, against the major envelope fusion protein of Helicoverpa armigera nucleopolyhedrovirus显示文摘The envelope fusion protein F of baculoviruses is a class I viral fusion protein which play a significant role during virus entry into insect cells. F is initially synthesized as a precursor(F_0) and then cleaved into a disulfide-linked F_1 and F_2 subunits during the process of protein maturation and secretion. To facilitate further investigation into the structure and function of F protein during virus infection, monoclonal antibodies(mAbs) against the F_2 subunit of Helicoverpa armigera nucleopolyhedrovirus(HearNPV)(Ha F) were generated. Two kinds of mAbs were obtained according to their different recognition epitopes: one kind of mAbs, as represented by 38F10,recognizes amino acid(aa) 85 to 123 of F_2 and the other kind, represented by 44D11, recognizes aa148 to 173 of F_2. Western blot and immunofluorescence assay confirmed that both of the mAbs recognized the F protein expressed in HearNPV infected cells, however, only 44D11 could neutralize HearNPV infection. The results further showed that 44D11 may not interact with a receptor binding epitope, rather it was demonstrated to inhibit syncytium formation in cells expressing the Ha F protein. | 4 | Towards high-power, high-coherence, integrated photonic mm Wave platform with microcavity solitons显示文摘Millimetre-wave(mmWave)technology continues to draw great interest due to its broad applications in wireless communications,radar,and spectroscopy.Compared to pure electronic solutions,photonic-based mmWave generation provides wide bandwidth,low power dissipation,and remoting through low-loss fibres.However,at high frequencies,two major challenges exist for the photonic system:the power roll-off of the photodiode,and the large signal linewidth derived directly from the lasers.Here,we demonstrate a new photonic mmWave platform combining integrated microresonator solitons and high-speed photodiodes to address the challenges in both power and coherence.The solitons,being inherently mode-locked,are measured to provide 5.8 dB additional gain through constructive interference among mmWave beatnotes,and the absolute mmWave power approaches the theoretical limit of conventional heterodyne detection at 100 GHz.In our free-running system,the soliton is capable of reducing the mmWave linewidth by two orders of magnitude from that of the pump laser.Our work leverages microresonator solitons and high-speed modified uni-traveling carrier photodiodes to provide a viable path to chip-scale,high-power,low-noise,high-frequency sources for mmWave applications. | Beichen Wang Jesse SMorgan Keye Sun Mandana Jahanbozorgi Zijiao Yang Madison Woodson Steven Estrella Andreas Beling Xu Yi | 2021 | Light(Science & Applications)2021,10,1: | 3 |

| 10 | Molecular characterization and expression of the SiUCP2 gene in sea urchin Strongylocentrotus intermedius显示文摘Uncoupling protein 2 (UCP2) is a proton transporter located in the inner mitochondrial membrane, and inhibits the formation of adenosine triphosphate and reactive oxygen species by uncoupling oxidative phosphorylation. To provide a theoretical basis for the role of SiUCP2 in lipid metabolism, a 2 341-bp full-length cDNA of SiUCP2 from sea urchin Strongylocentrotus intermedius , which encodes 323 amino acids (predicted MW 36.11 kDa) was obtained, and the structure and function of the SiUCP2 gene and its expression at the mRNA and protein level were studied. SiUCP2 had high homology with UCP2 of other species. Expression of SiUCP2 was detected in the order of tube feet > gonads > coelomocytes > intestines. The expression level was the highest in prismatic larvae and lowest in the two-cell stage. Moreover, using in-situ hybridization, we found that SiUCP2 protein was expressed in the gonads and intestine. | 11 | Radio-frequency line-by-line Fourier synthesis based on optical soliton microcombs显示文摘Radio-frequency(RF) waveform synthesis has broad applications in ultrawide-bandwidth wireless communications, radar systems, and electronic testing. Photonic-based approaches offer key advantages in bandwidth and phase noise thanks to the ultrahigh optical carrier frequency. In this work, we demonstrate Fourier synthesis arbitrary waveform generation(AWG) with integrated optical microresonator solitons. The RF temporal waveform is synthesized through line-by-line amplitude and phase shaping of an optical soliton microcomb, which is down-converted to the RF domain through dual-comb optical coherent sampling. A variety of RF waveforms with tunable repetition cycles are shown in our demonstration. | 12 | Molecular Cloning, Expression and Characterization of Peroxisome Proliferators-Activated Receptors Gamma in the Sea Urchin(Strongylocentrotus intermedius)显示文摘Peroxisome proliferators-activated receptor gamma(PPARγ) plays important regulatory roles in adipocyte differentiation. In this study, we cloned the full-length sequence of the PPARγ gene and analyzed its expression profile in different developmental stages and tissues of Strongylocentrotus intermedius. The full-length cDNA of PPARγ contains 2286 base pairs(bp) with a putative open reading frame of 1755 bp, and the gene encodes encoding a polypeptide of 584 amino acid residues. The predicted molecular mass of the protein is 67.27 kDa, and its theoretical isoelectric point(pI) is 10.07. The protein contains conserved motifs, including an RRM(RNA recognition motif) domain. PPARγ expression with the highest level was observed in the gonad, and the lowest was observed in the tube feet of S. intermedius. Time-course expression measurements at different developmental stages showed that the highest expression level of PPARγ is in the eggs and its weakest expression level is in the 32-cells stage. Knock-down of PPARγ by specific siRNA revealed that UCP2 expression is significantly decreased in the gonads and intestines 48 h post-transfection, indicating that the UCP2 is a downstream target gene of PPARγ.This finding suggests that PPARγ and UCP2 have positive regulatory effects in regulating adipocyte differentiation. Changes in fatty acid levels in the gonads before and after PPARγ interference were assessed, and decreased C18:2(trans, n-6) and C20:3(n-6) levels were observed 48 h after siRNA transfection. The results showed the function of PPARγ in fatty acid anabolism, The data are helpful to improve the current understanding of the fatty acid synthesis pathways and regulatory mechanisms in S. intermedius.
