| 1 | Betanodavirus: Mitochondrial disruption and necrotic cell death显示文摘Betanodaviruses cause viral nervous necrosis, an infectious neuropathological condition in fish that is characterized by necrosis of the central nervous system, including the brain and retina. This disease can cause mass mortality in larval and juvenile populations of several teleost species and is of global economic importance. The mechanism of brain and retina damage during betanodavirus infection is poorly understood. In this review, we will focus recent results that highlight betanodavirus infection-induced molecular death mechanisms in vitro. Betanodavirus can induce host cellular death and post-apoptotic necrosis in fish cells. Betanodavirus-induced necrotic cell death is also correlated with loss of mitochondrial membrane potential in fish cells, as this necrotic cell death is blocked by the mitochondrial membrane permeability transition pore inhibitor bongkrekic acid and the expression of the antiapoptotic Bcl-2 family member zf Bcl-x L. Moreover, this mitochondria-mediated necrotic cell death may require a caspase-independent pathway. A possible cellular death pathway involving mitochondrial function and the modulator zf Bcl-xs is discussed which may provide new insights into the necrotic pathogenesis of betanodavirus. | Jiann-Ruey Hong | 2013 | World Journal of Virology2013,2,1: | 2 |
| 2 | Anti-fungal properties of Puroindoline A against Aspergillus glaucus显示文摘Aspergillus glaucus can grow in low moisture grain,and is one of the main fungi responsible for agricultural product losses.Puroindoline A(PINA)is a tryptophan-rich alkaline adiponectin that can effectively inhibit numerous plant bacteria and fungi.However,the mechanism of PINA against A.glaucus remains unclear.Herein,we found that recombinant PINA(rPINA)could inhibit A.glaucus mycelia growth on salt Czapek dox agar(SCDA)medium and spore germination on Czapek dox(CD)medium.Scanning electron microscopy revealed that incomplete morphological characteristics of both A.glaucus spores and mycelia occurred following rPINA treatment.Laser scanning confocal microscopy(LSCM)showed that rPINA could enter the interior of spores.Flow cytometry and propidium iodide(PI)staining illustrated membranes of spores were severely damaged,especially after treatment with 0.9 mg/mL rPINA for 12 h,and spores with intact membranes were reduced by 30.7%.Additionally,rPINA reduced the mitochondrial membrane potential(MMP)by 5,5′,6,6′-tetrachlorr-1,1′,3,3′-tetraethyl-imidacarbocyanine iodide(JC-1)staining,and caused DNA damage by 4′,6-diamidino-2-phenylindole(DAPI)staining.These results indicated that rPINA may damage cell membranes and DNA structure and reduceMMP,thereby inhibiting the growth of A.glaucus.The antifungal mechanism has been demonstrated in this study,and results show that rPINA has application potential in preventing postharvest loss in the agricultural industry. | Wenjing Yuan Pingping Tian Ang Lyu Yangyong Lyu Wei Zhang Shan Wei Yuansen Hu | 2020 | Grain & Oil Science and Technology2020,3,1: | 0 |
| 3 | Relationship Between Membrane Damage Induced by Osmotic Stress and Lipid Peroxidation and Some Free Radicals显示文摘To treat leaf discs with solutions of various osmotic potentials of polyethylene glycols (MW 6000) by adopting the floating treatment could increase the membrane permeability, decrease the formation of malondialdehyde,and reduce the activity of peroxidase. Nevertheless, the activities of superoxide dismutase and catalase were not obviously altered. In verifying if the membrane damage was caused by lipid peroxidation initiated by active oxygen species, diethyldithiocarbamate was chosen as inhibitor superoxide dismutase,aminotriazole as catalase and mannitol as scavenger of hydroxyl free radicals.The results have shown that there is not any correlation between lipid peroxidation and membrane damage.Therefore,membrane damage caused by water stress is probably not due to free radical initiation. | 王邦锡 孙莉 黄久常 | 1993 | Science China Chemistry1993,36,1: | 0 |
